Caged Morpholino Oligonucleotide For Control Of Gene Expression In Zebrafish, Chad Michael Jarreau

To control gene expression in vivo with spatial and temporal precision remains a significant hurdle in laboratory studies of development as well as clinical genetic therapies. Here we demonstrate such control over gene expression by use of photochemistry to reversibly inactivate the hybridization of a nucleic acid analog used for specific protein knockdown. A morpholino oligonucleotide, commonly used for knockdown of protein expression in developmental studies, was “caged” using carbodiimide conjugate chemistry which yielded photocleavable adducts that can be removed with light exposure. fac-[Re^I(CO)₃L]ⁿ complexes serve as models for short-lived fac-[^99mTc^I(CO)₃L] imaging tracers. Dangling groups on L, needed to achieve desirable biodistribution, complicate the NMR spectra, which are not readily understood. In fac-[Re^I(CO)₃L]⁺ with less complicated L, NH groups (exo-NH) projecting toward the L face sometimes showed an upfield shift attributable to steric shielding of the exo-NH group from the solvent by the chelate rings. World competition has encouraged United States rice farmers and rice mills to be efficient in farming and production practices. Efforts to augment economic competitiveness include development of new varieties, improvements in milling practices, and identification of uses for rice products and by-products. The research detailed in this dissertation adds to the body of knowledge in milling practices and identification of uses for rice bran. To improve the prediction of milled rice quality at industrial scale, correlations for milling quality among laboratory, pilot, and industrial scale mills were identified for Clearfield 161. This study evaluated the acute time course of the ergogenic effect of concurrent activation potentiation (CAP). Forty-two men and women, including CAP non-responders and responders, performed a 5 second isometric knee extension on a dynamometer with the use of remote voluntary contractions (RVC). Mean torque was assessed in seven 500 millisecond (ms) time periods. A two-way repeated measures ANOVA revealed significant main effects for time period (p ≤ 0.001), but no significant interaction between time period and CAP non-responders and responders (p > 0.05). The ergogenic effects of CAP are accrued during the first 1000ms. Concurrent activation potentiation responders produce greater …

The reexpression of the fetal γ-globin gene in adult erythrocytes is of therapeutic interest due to its ameliorating effects in β-hemoglobinopathies. We recently showed that Methyl CpG Binding Domain Protein2 (MBD2) contributes to the silencing of the chicken embryonic ρ-globin and human fetal γ-globin genes. We further biochemically characterized an erythroid MeCP1 complex that is recruited by MBD2 to mediate the silencing of these genes. These observations suggest that the disruption of the MeCP1 complex could augment the expression of the fetal/embryonic globin genes. The happiness revolution is coming to legal scholarship. Based on empirical data about the how and why of positive emotions, legal scholars are beginning to suggest reforms to legal institutions. In this article we aim to redirect and slow down this revolution.

One of their first targets of these legal hedonists is the jury system for tort damages. In several recent articles, scholars have concluded that early findings about hedonic adaptation and affective forecasting undermine tort awards for pain and suffering, mental anguish, loss of enjoyment of life, and other non-economic damages. Restrictive eating attitudes and behaviors have been hypothesized to be related to processes of intrasexual competition. According to this perspective, within-sex competition for status serves the adaptive purpose of attracting mates. As such, status competition salience may lead to concerns of mating desirability. For heterosexual women and gay men, such concerns revolve around appearing youthful and, thus, thinner. Following this logic, we examined how exposure to high-status and competitive (but not thin or highly attractive) same-sex individuals would influence body image and eating attitudes in heterosexual and in gay/lesbian individuals. This paper seeks to explain and describe the xenophobia and xenophobic attacks of Zimbabwean refugees and migrants in South Africa. The political and social instability in Zimbabwe has led to mass exodus of Zimbabweans in search of stability and a means to survive. Many of these Zimbabweans have sought opportunity and refuge in South Africa only to be met with hurdles that metastasized into xenophobia. Research was done on the target population of Zimbabweans in Central Methodist Church in Johannesburg, along with interviews conducted through Medicines Sans Frontiers, and active players in the civil society in Cape Town.

Although a species’ locomotor capacity is suggestive of its ability to escape global climate change, such a suggestion is not necessarily straightforward. Species vary substantially in locomotor capacity, both ontogenetically and within/among populations, and much of this variation has a genetic basis. Accordingly, locomotor capacity can and does evolve rapidly, as selection experiments demonstrate. Importantly, even though this evolution of locomotor capacity may be rapid enough to escape changing climate, genetic correlations among traits (often due to pleiotropy) are such that successful or rapid dispersers are often limited in colonization or reproductive ability, which may be viewed as a trade-off. …

Kim’s essay “Anger, Cognition, Ideology: What Crash Can Show Us About Emotion” argues for the relevance and importance of cognitive studies to ethnic and postcolonial literary studies, and vice versa. After surveying recent developments in the field of cognitive studies, the essay combines cultural and cognitive approaches in order to examine anger in and around the 2005 Paul Haggis film Crash.

The Effects Of Flower Color Transitions On Diversification Rates In Morning Glories (Ipomoea Subg. Quamoclit, Convolvulaceae), Stacey Dewitt Smith, Richard E. Miller, Sarah P. Otto, Richard G. Fitzjohn, Mark D. Rausher

As in many clades of flowering plants, the Quamoclit clade of morning glories (Ipomoea subgenus Quamoclit) exhibits unequal proportions of different flower colors, with pigmented species outnumbering unpigmented species by nearly a factor of 7. We examined three possible macroevolutionary explanations for this pattern: (1) asymmetric transition rates between pigmented and unpigmented flowers; (2) low transition rates preventing the flower colors from reaching equilibrium; and (3) differential diversification. In order to discriminate among these explanations, we employ the newly-developed Binary-State Speciation and Extinction (BiSSE) model, which jointly estimates transition rates among states and the speciation and extinction rates …

Objective: The minor T-allele of rs780094 in the glucokinase regulator gene (GCKR) associates with a number of metabolic traits including higher triglyceride levels and improved glycemic regulation in study populations of mostly European ancestry. Using data from the Atherosclerosis Risk in Communities (ARIC) Study, we sought to replicate these findings, examine them in a large population-based sample of African American study participants, and to investigate independent associations with other metabolic traits in order to determine if variation in GKCR contributes to their observed clustering. The effect of the plant-derived nonpsychotropic cannabinoid, cannabidiol (CBD), on the function of hydroxytryptamine (5-HT)3A receptors expressed in Xenopus laevis oocytes was investigated using two-electrode voltage-clamp techniques. CBD reversibly inhibited 5-HT (1 μM)-evoked currents in a concentration-dependent manner (IC50 = 0.6 μM). CBD (1 μM) did not alter specific binding of the 5-HT3A antagonist [3H]3-(5-methyl-1H-imidazol-4-yl)-1-(1-methylindol-3-yl)propan-1-one (GR65630), in oocytes expressing 5-HT3A receptors. In the presence of 1 μM CBD, the maximal 5-HT-induced currents were also inhibited. The EC50 values were 1.2 and 1.4 μM, in the absence and presence of CBD, indicating that CBD acts as a noncompetitive antagonist of 5-HT3 …

Cell-cell adhesion mediated by desmosomes is crucial for maintaining proper epidermal structure and function, as evidenced by several severe and potentially fatal skin disorders involving impairment of desmosomal proteins. Pemphigus foliaceus (PF) and staphylococcal scalded skin syndrome (SSSS) are subcorneal blistering diseases resulting from loss of function of the desmosomal cadherin, desmoglein 1 (Dsg1). To further study the pathomechanism of these diseases and to assess the adhesive properties of Dsg2, we employed a recently established transgenic (Tg) mouse model expressing Dsg2 in the superficial epidermis. The confluent telangiectasia on her cheeks gave a permanent appearance of well-applied rouge. Law confirms but seldom determines the course of a society. Values and beliefs, instead, are the true polestars, incrementally implemented by the laws, customs, and policies. The Chiricahua Apache, a tribal society of hunters, gatherers, and raiders in the mountains and deserts of the Southwest, were squeezed between the growing populations and economies of the United States and Mexico. Raiding brought response, reprisal, and ultimately confinement at the loathsome San Carlos Reservation.
