Molecular Characterization Of Rhizoctonia Solani Ag4 Using Pcr-Rflp Of The Rdna-Its Region, Meli̇ke Çebi̇ Kiliçoğlu, İbrahi̇m Özkoç

Rhizoctonia solani isolates obtained from common beans (Phaseolus vulgaris L.) were included in an AG4 anastomosis group in accordance with hyphal anastomosis. In the subgrouping of AG4 isolates, PCR-RFLP patterns in the rDNA-ITS were used. After obtaining the genomic DNA belonging to R. solani AG4, an approximately 700 bp amplification product of the ITS1-5.8S-ITS2 region was obtained with PCR, using ITS1 and ITS4 universal primers. The PCR products were digested with MseI, HincII, AvaII, and MfeI restriction endonucleases, and different PCR-RFLP patterns were obtained for the AG4 HGI and AG4 HGII subgroups. Barley Germplasms Developed For Scald Disease Resistance Exhibited A High Level Of Genetic Diversity Based On Srap Markers, Ayten Di̇zkirici, Zeki̇ Kaya, H. Eli̇f Güren, Hi̇kmet Budak

The objective of this study was to assess the genetic diversity and genetic relationships among 59 Turkish barley (Hordeum vulgare L.) germplasm lines (maintained for scald disease resistance breeding) using sequence-related amplified polymorphism (SRAP) markers. Seventeen SRAP primer combinations produced 83 polymorphic markers, with a mean polymorphism of 73.5%. The dendrogram created based on Nei's unweighted-pair group method using arithmetic average (UPGMA) indicated that there were 4 main clusters, which was supported by principle component analysis (PCA). Indole-3-Acetic Acid And Gibberellic Acid Production In Aspergillus Niger, Işil Seyi̇s Bi̇lkay, Şafak Karakoç, Ni̇lüfer Aksöz

The effects of incubation time, temperature, pH, and agitation on indole-3- acetic acid and gibberellic acid production in Aspergillus niger were studied. For indole-3-acetic acid production, 6 days of incubation at 25 °C and pH 6.0 was found to be optimum. Optimum conditions for gibberellic acid production were 12 days of incubation at 30 °C and pH 5.0. Agitation increased both indole-3-acetic acid and gibberellic acid production.

Temporal Variations Of Surface Phytoplankton, Nutrients And Chlorophyll A In The Dardanelles (Turkish Straits System): A Coastal Station Sample In Weekly Time Intervals, Muhammet Türkoğlu

In this study, weekly distributions of inorganic nutrients and phytoplankton cell volumes were investigated in relation to the hydrology of the Dardanelles. The data were collected between March 2001 and March 2002. NO^{-}_{2}+NO^{-}_{3}, PO_{4}^{-3}, and SiO_4 concentrations ranged between 0.050-6.887 µM, 0.051-1.152 µM, and 0.64-10.74 µM, respectively. During the study, the highest nutrient values were measured between late fall and mid-winter. Inorganic N:P and Si:P ratios in the surface water were lower due to high PO_{4}^{-3} concentrations. The chlorophyll a concentrations ranged from 0.03 to 8.67 µg L^{-1}. Phytoplankton cell density and cell volume ranged from 1.54E + 05 to …

Genetic Diversity In Doubled Haploid Wheat According To The Acid-Page Method, Morphological Traits, And Baking Quality, Javid Ojaghi, Ellada Akhundova

This study aimed to assess the genetic diversity of 102 lines of doubled haploid wheat (obtained from CIMMYT) using the acid-PAGE method, morphological traits, and baking quality. Cluster analysis of the morphological traits separated all the genotypes into 4 groups, such that the genotypes with high yield were placed in 1 group. However, based on cluster analysis of their qualitative traits, the genotypes were classified into 3 main groups, while the genotypes with high protein content were placed in a distinct group. Molecular Fingerprinting Of Some Clinically Significant Nocardia And Related Strains By Restriction Polymorphism Ribosomal Rna Analyses, Kami̇l Işik, Michael Goodfellow

Fifty-three Nocardia strains were the subject of a restriction polymorphic ribosomal RNA analysis (ribotyping) designed to distinguish between representatives of clinically significant species and related strains. The organisms were assigned to 19 groups using a combination of EcoRV gene restriction endonuclease patterns and a digoxigenin-labelled Streptomyces violaceoruber TK21 rDNA probe. Each ribotype group contained 4 to 13 restriction fragments that ranged in size from 20.7 to 0.9 kb. The N. brasiliensis, N. crassostreae, N. farcinica, N. otitidiscaviarum, and N. seriolea strains showed distinct ribotype patterns. Biological Activities Of Cu(Ii) And Hg(Ii) Complexes Of A Heptadentate Schiff Base Ligand, Emel Ergene, Hülya Si̇vas, Kadri̇ye Benkli̇

Two metal complexes were synthesized as [Hg(L)](ClO_4)_2 and [Cu(L)](ClO_4)2 by the template reaction between 2,6-bis(2 aminothiophenoxymethyl)pyridine and 2,2'-bipyridine-6,6'-dicarboxaldehyde in the presence of Hg(II) and Cu(II) perchlorate salts. The structures of the compounds were elucidated by IR, 1H-NMR, MASS, and Elemental Analyses values. Cytotoxicity of the compounds were investigated by MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay in normal and cancerous rat fibroblasts. Both compounds showed cytotoxicity on 2 cell lines as a dose-dependent manner. Compound [Hg(L)](ClO_4)_2 was more cytotoxic than [Cu(L)](ClO_4)2. Microbial biofilm and corrosion in cooling systems are the most common problems that damage expensive equipment, cause loss of production, and increase maintenance costs. Sulfate reducing bacteria were considered the major bacterial group involved in microbiologically influenced corrosion (MIC). The in vitro effect of certain fungicides, insecticides, and biopesticides on mycelial growth of Trichoderma harzianum was evaluated. Seven systemic fungicides, 2 contact fungicides, and 4 biopesticides were tested. A progressive increase in percent inhibition of radial growth in the fungus was observed as the concentrations of the fungicides increased. Among the systemic fungicides, hexaconazole was the most toxic, followed by propiconazole and triflumizole. Toxicity of the contact fungicides was lower than that of the systemic fungicides, among which copper oxychloride and copper hydroxide were highly compatible; no inhibition was observed at lower concentrations. To determine the potential biochemical effects of heavy metal pollution on aquatic organisms, pleopodal eggs, and stage-1 juveniles of the freshwater crayfish, Astacus leptodactylus (Eschscholtz, 1823), was exposed to a non-essential (Cd) and an essential (Cu) metal, either singly or in combination. Soil samples were collected from an industrial area in Kazanlı, Mersin, Turkey. Six heavy metals, Cr, Cu, Ni, Co, Cd, and Zn, were tested. ICP-AES analyses were carried out to detect the heavy metal levels of the soil samples. The analysis of test samples revealed high levels of Ni and Cr. A total of 272 Pseudomonas spp. and 161 Bacillus spp. strains were isolated from soils from 4 different sampling locations. The maximum tolerable metal concentrations (MTCs) of Cr, Cu, Ni, Co, Cd, and Zn for each isolate were determined. Resistance to Cr was found in 73.9% of the bacterial …

Fatty Acid Composition Of Total Lipids In Muscle Tissues Of Nine Freshwater Fish From The River Tigris (Turkey), Eli̇f İpek Cengi̇z, Erhan Ünlü, Mehmet Başhan

Fatty acid compositions of total lipids in the muscle of 9 freshwater fish species were examined via gas chromatography. Palmitic acid and oleic acid were the predominant saturated (SFA) and monounsaturated fatty acids (MUFA) in all the species analyzed. The highest value of total SFA was detected in Liza abu (48.94%). Alburnus mossulensis had the greatest percentage of total MUFA (55.56%). Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) were the most abundant PUFA in all the species analyzed. Seventy soil samples were collected from 25 different plant varieties grown in fields in Aydın province. Arbuscular mycorrhizal fungi (AMF) were found in 59 soil samples; 58 of these were identified as Glomus sp. and 1 as Gigaspora sp. Due to its adaptation characteristics, growth rates, and colonization properties, Glomus sp. was selected for further study. The effects of Glomus sp. and BioOrganics (Glomus aggregatum, G. clarum, G. deserticola, G. intraradices, G. monosporus, G. mosseae, Gigaspora margarita, and Paraglomus brasilianum) on tomato, pepper, and cucumber plant development were evaluated. The antioxidant activity of the methanolic extracts of 4 plants from genus Helichrysum Mill. (Asteraceae) [H. arenarium (L.) Moench subsp. erzincanicum Davis & Kupicha, H. arenarium (L.) Moench subsp. rubicundum (C.Koch.) Davis & Kupicha, H. armenium DC. subsp. araxinum (Kirp.) Takht., and H. plicatum DC. subsp. pseudoplicatum (Nab.) Davis & Kupicha] was investigated with 2 complementary test systems, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging and phosphomolybdenum assay. Genetic variations are the raw materials of plant breeding and have been increasingly narrowed due to various reasons. Landraces are good sources of genetic variation, but need to be characterized first. This study was conducted to determine the levels of genetic variation in 3 barley materials originated in Turkey, all known as Tokak. Megagametophyte tissue in seeds from a Pinus bruia Ten. seed orchard was analyzed for 9 enzyme systems (aconitase, alcohol dehydrogenase, glutamate dehydrogenase, glutamate oxaloacetate-transaminase, malate dehydrogenase, menadione reductase, 6-phosphogluconate dehydrogenase, phosphoglucose isomerase, and shikimate dehydrogenase) encoded by 14 loci using starch gel electrophoresis. Twenty-eight clones and 3 ramets per clone were sampled from the orchard for analysis. Isozyme patterns of the clones clearly indicate that each clone had a different genotype when overall loci were taken into consideration. Sixty Bacillus spp. isolates were obtained from different agricultural fields in Kayseri province using selective media. Total DNA was isolated and analyzed by PCR using cry1 general primers. Of the 60 tested isolates, 17 carried the cry1 gene. B. thuringiensis subsp. kurstaki and all other isolates exhibited approximately 19-kb plasmid bands. In addition to the 270-bp band, the standard strain and isolate I47 yielded 180-bp PCR products. The isolates and standard strain were toxic to Ephestia kuehniella Zeller and Plodia interpunctella Hubner larvae. Propolis has a strong antimicrobial effect and limits the growth of microorganisms. This study was carried out to determine the effect of propolis on the storage life of Star Ruby grapefruit. Fruits were obtained from Mustafa Kemal University, Faculty of Agriculture, Dörtyol Experimental Research Station, Dörtyol, Hatay, Turkey. Soil compaction caused by heavy wheeling reduces water infiltration, root development, and yield, and increases bulk density and soil strength. The objective of this study was to determine changes in spatial variability patterns of soil penetration resistance before and after using the silage machine in a corn-growing field. Soil penetration resistance was measured in a 100 × 400 m field with 10 m intervals just before and after harvesting. Copper remains the primary biocide component used today to protect wood. Increased interest in the use of non-arsenic copper-based wood preservatives has also led to increased studies on copper-tolerant decay fungi. Oxalic acid production by brown-rot fungi is proposed as one mechanism of copper tolerance. This study evaluated oxalic acid production and copper losses from ACQ- and CCA-treated Pinus sylvestris L. and Populus × euramericana I 214 wood by brown-rot fungi: Postia placenta, Gloeophyllum trabeum, Tyromyces palustris, and 2 strains of Serpula lacrymans. Homopteran insects contain bacteria in their cells and tissues known as "secondary symbionts," which under special environmental circumstances act against their host insects. In this study, both molecular- and culture-based methods were used to characterize the bacteria associated with the whitefly in Jordan. We isolated, cultured, and identified 11 species of bacteria from nymphs (6 species), adults (8 species), and parasitized pupae (2 species) of the whitefly Bemesia tabaci collected from different vegetable crops planted in different localities in Jordan. The aim of this study was to determine the effectiveness of fennel (Foeniculum vulgare Mill.) leaves as food on development, survival, and reproduction in Podisus nigrispinus (Dallas). Newly hatched P. nigrispinus nymphs were used in this study. The prey consisted of 3rd-instar Alabama argillacea (Huebner) larvae. Fennel and cotton leaves were not sufficient to enable the full development of the predator to its adult stage. In drained Histosols used as pastures or arable fields the muck-forming process transforms organic soil formations into muck. The rush vegetation (Caricetum acutiformis and Glycerietum maximae) covering Histosols in the organic matter accumulation stage is replaced by meadow vegetation (Molinietalia order). During sustained drainage mucks are transformed into muck-like formations that contain less than 12% organic carbon. The aim of this study was to report the changes in organic matter initiated by the muck-forming process. Soil organic matter was fractionated into humic-acid carbon, fulvic-acid carbon, and humins.
