Excision Dynamics Of Vibrio Pathogenicity Island-2 From Vibrio Cholerae: Role Of A Recombination Directionality Factor Vefa, Salvador Almagro-Moreno, Michael G. Napolitano, E. Fidelma Boyd

Vibrio Pathogenicity Island-2 (VPI-2) is a 57 kb region present in choleragenic V. cholerae isolates that is required for growth on sialic acid as a sole carbon source. V. cholerae non-O1/O139 pathogenic strains also contain VPI-2, which in addition to sialic acid catabolism genes also encodes a type 3 secretion system in these strains. VPI-2 integrates into chromosome 1 at a tRNA-serine site and encodes an integrase intV2 (VC1758) that belongs to the tyrosine recombinase family. ntV2 is required for VPI-2 excision from chromosome 1, which occurs at very low levels, and formation of a non-replicative circular intermediate.

In this report, we analyze trends in interleukin-6 (IL-6) production in people living with human immunodeficiency virus (PLWH), as a function of smoking behaviors and race, using a clinic-based case-control design. Peripheral blood mononuclear cells (PBMCs) were collected from a multiethnic cohort of 120 PLWH to measure IL-6 in culture supernatant. The data indicated that stimulated PBMC produced significantly higher levels of IL-6 in smokers than nonsmokers. However, as expected, this relationship was substantially modified by race. The distinctive production of IL-6 across different racial groups highlights the need for additional studies and suggests that African American smokers have enhanced …

Undisturbed archeological deposits at Fort Charlotte—a component of Grand Portage National Monument, Minnesota—reflect the daily activities and social dynamics of the Canadian fur trade. These remains are threatened by both natural and human factors, and the park has sought methods to monitor the site, protect its archeological resources from destruction, and maintain the potential for significant research into all aspects of the fur trade. This thesis explores the potential of Fort Charlotte as a significant archeological site, discusses trends and current attitudes toward historic preservation, and offers recommendations for the preservation of 21CK7. While much is known about the relationship between stress and the steroid hormone, corticosterone (CORT) in adult animals, less is known about the dynamics of CORT secretion in developing animals. My research examined factors affecting levels of baseline and stress-induced CORT in a free-living, non-migratory passerine, the Florida scrub-jay (Aphelocoma coerulescens). Nuclear Magnetic Resonance Affinity Screening Methods For Functional Annotation Of Proteins And Drug Discovery, Matthew D. Shortridge

With nearly 1,350 complete genome sequences available our understanding of biology at the molecular level has never been more complete. A consequence of these sequencing projects was the discovery of large functionally unannotated segments of each genome. The genes (and proteins they encode) found in these unannotated regions are considered “hypothetical proteins”. Current estimates suggest between 12%-50% of the known gene sequences are functionally unannotated. Incomplete functional annotation of the various genomes significantly limits our understanding of biology. Pragmatically, identifying the functions of these proteins could lead to new therapeutics; making functional annotation of paramount importance.

In the mid to late 1700s, men of letters became more and more interested in the natural world. From studies in astronomy to biology, chemistry, and medicine, these "philosophers" pioneered what would become our current scientific categories. While the significance of their contributions to these fields has been widely appreciated historically, the interconnection between these men and their literary counterparts has not. A study of the "Romantic man of science" reveals how much that figure has in common with the traditional "Romantic" literary figure embodied by poets like William Wordsworth and Samuel Taylor Coleridge. Frequency Dependence In The Initiation Of Ultrafast Laser-Induced Damage, Jeremy R. Gulley

Numerous studies have investigated the role of photoionization in ultrafast laser-induced damage of bulk dielectrics. This study examines the role of spectral width and instantaneous laser frequency in laser-induced damage using a frequency dependent multiphoton ionization model and numerical simulation of an 800 nm laser pulse propagating through fused silica. When the individual photon wavelengths are greater than 827 nm, an additional photon is required for photoionization, reducing the probability of the event by many orders of magnitude. Simulation results suggest that this frequency dependence may significantly affect the processes of laser-induced damage and filamentation.

BACKGROUND: Thiamine (vitamin B1) deficiency (TD) causes mild impairment of oxidative metabolism and region-selective neuronal loss in the central nervous system (CNS). TD in animals has been used to model aging-associated neurodegeneration in the brain. The mechanisms of TD-induced neuron death are complex, and it is likely multiple mechanisms interplay and contribute to the action of TD. In this study, we demonstrated that TD significantly increased intracellular calcium concentrations [Ca2+]i in cultured cortical neurons.

RESULTS: TD drastically potentiated AMPA-triggered calcium influx and inhibited pre-mRNA editing of GluR2, a Ca2+-permeable subtype of AMPA receptors. The Ca2+ permeability of GluR2 is regulated …

Background: More than 450 newborns die every hour worldwide, before they reach the age of four weeks (neonatal period) and over 500,000 women die from complications related to childbirth. The major direct causes of neonatal death are infections (36%), Prematurity (28%) and Asphyxia (23%). Pakistan has one of the highest perinatal and neonatal mortality rates in the region and contributes significantly to global neonatal mortality. The high mortality rates are partially attributable to scarcity of trained skilled birth attendants and paucity of resources. The role of protein isoaspartyl methyltransferase (PIMT) in repairing a wide assortment of damaged proteins in a host of organisms has been inferred from the affinity of the enzyme for isoaspartyl residues in a plethora of amino acid contexts. The identification of PIMT target proteins in plant seeds, where the enzyme is highly active and proteome long-lived, has been hindered by large amounts of isoaspartate-containing storage proteins. Mature seed phage display libraries circumvented this problem. In Qβ RNA, sequestering the coat gene ribosome binding site in a putatively strong hairpin stem structure eliminated synthesis of coat protein and activated protein synthesis from the much weaker maturation gene initiation site, located 1300 nucleotides upstream. As the stability of a hairpin stem comprising the coat gene Shine–Dalgarno site was incrementally increased, there was a corresponding increase in translation of maturation protein. The effect of the downstream coat gene ribosome binding sequence on maturation gene expression appeared to have occurred only in cis and did not require an AUG start codon or initiation of coat protein synthesis. M. malabathricum grows well in acidic soil with high Al solubility, thereby it can be used as a model plant for tolerance to aluminum and acid stresses. The goal of determining which of hundreds of thousands of SNPs are associated with disease poses one of the most challenging multiple testing problems. Using the empirical Bayes approach, the local false discovery rate (LFDR) estimated using popular semiparametric models has enjoyed success in simultaneous inference. However, the estimated LFDR can be biased because the semiparametric approach tends to overestimate the proportion of the non-associated single nucleotide polymorphisms (SNPs). One of the negative consequences is that, like conventional p-values, such LFDR estimates cannot quantify the amount of information in the data that favors the null hypothesis of no disease-association.

Epigenetic modifications play a key role in the patho-physiology of many tumors and the current use of agents targeting epigenetic changes has become a topic of intense interest in cancer research. DNA methyltransferase (DNMT) inhibitors represent a promising class of epigenetic modulators. Research performed yielded promising anti-tumorigenic activity for these agents in vitro and in vivo against a variety of hematologic and solid tumors. These epigenetic modulators cause cell cycle and growth arrest, differentiation and apoptosis. Purification and Characterization of Fibrinolytic Proteases from Mushroom Volvariela volvaceae Extract. Edible straw mushroom (V. volvaceae) has been known used for improvement of blood circulation due to its fibrinolytic content. The objective of the study is to purify and characterize fibrinolytic protease from straw mushroom extract. Purification were performed through several steps, i.e. precipitation using ammonium sulphate 75%, dialyzed membran (cut-off 10 kDa), and ion-exchange chromatography using DEAE Sepharose. Genomic Integrity Detection of In Vitro Irradiated Banana Using Microsatellite Marker. The research aims to detect genomic integrity of in vitro irradiated banana using microsatellite marker. These studies were done on banana cv. Pisang Mas irradiated by 15 Gy of gamma ray. The DNA was isolated from each accesion following Dixie. Amplification of DNA products were done by Perkin Elmer Gene Amp PCR 2400 using ten primers, and then electroforesis in agarose 1%. Genetic Diversity of Pandanus from West Java based on ISSR Markers. The genetic diversity of 10 species of Pandanus from West Java was examined using inter simple sequence repeat (ISSR) technique. Samples were collected from different localities in West Java. Two primers (ISSR2 and ISSR7) were selected as reliable amplifying ISSR markers. The two primers amplified 19 band position, with amplification size ranged from 200 to 1500 bp. Genetic distance for samples ranged from 0.267 to 0.957.
