During the course of this research a novel method which couples the molecular recognition-triggered photoamplification chain in diaryl ketone adducts of dithiane with a "turn-off" or "turn-on" fluorescence-based assay for the detection of biological targets and ligands, regardless of their nature, through a molecular recognition event has been developed. This research has included several key steps, the most significant being: (1) the design of fluorophore adducts or dyads which recover fluorescence upon photocleavage for a "turn-on" assay and the identification of fluorophores which are quenched upon the photochemical release of a quencher for a "turn off" assay; (2) Optimization of …

The conservation of prairie dogs is highly contested due to the embedded view that they are pests. This research addressed the ecological and social viability of prairie dog colonies in Denver, Colorado. Remote sensing analysis was applied to identify potentially viable areas for urban prairie dog colonies. In order to assess the social viability of urban colonies, knowledge and attitudinal surveys were distributed to residents near existing colonies and residents near potential colonies. Statistical analysis of responses provided insight into relationships between proximity to colonies, ecological knowledge, attitudes towards prairie dogs, demographics, and the presence of educational literature. Perishable artifacts, such as basketry, cordage, and sandals are rare cultural materials due to the environments in which they are preserved and their inherent non-durability. Where recovered, researchers have used them to study expressed identity and trace population movements over time and space. On this premise, previously un-described sandal assemblages from Trinchera Cave, Colorado and the Kenton Caves, Oklahoma/New Mexico were age dated, analyzed, and compared to other known sandal collections throughout North America, including Franktown Cave, Colorado. While depression is considered the most common mental illness regardless of age, gender, ethnicity, and socioeconomic status, compared to research on the general population, depression among psychologists has received little attention. While an abundance of literature on stereotypes exists with regards to the workplace, less research exists that assesses how stereotypes could affect an employee and motivate him or her to leave the organization. The present study addressed the relationship among job identification, gender identification, stereotype threat perception at work, anxiety, work specific efficacy, job satisfaction, and turnover intent. A hypothesized model that connected the possible relations among stereotype threat, gender identification, job identification, job anxiety, work specific self-efficacy, job satisfaction, and turnover intent was tested using data from 267 working women.

In the biomedical field the need for intra organ pressure measurement can only be facilitated by adapting existing pressure sensing technology to the specific tissues under test. The customization of these sensors has only driven up cost and the need to explore new technologies has become increasingly more important. For this dissertation, we explore the use of a new technology, particularly electrochemical pressure sensing to provide a low-cost pressure sensor to fill this need. In 1999, after several exploratory meetings, the college administration established an ad-hoc interdisciplinary assessment committee to begin a conversation about what students were taught and how faculty knew what was learned and what was not. At the first meeting of this committee, composed of representatives of the college’s seventeen teaching departments, the library, student affairs, institutional planning, research, and assessment, and academic affairs, several impediments to a formalized college-wide assessment initiative immediately became apparent. While a culture of informal assessment already existed as instructors daily grappled with effectively teaching their students, the notion of a widespread institutionalized plan was alien. …

From 2007-2009, we studied the current (2006) and historic (1892, mid-1950s) extent, current age distribution, and plant species composition of plains cottonwood (Populus deltoides) and non-cottonwood riparian stands along eight study segments of the Missouri River between Fort Benton, Montana and Kansas City Missouri, covering 930 river miles (1500 km) or over 1/3 of the river's length. These segments included all of the unchannelized and unimpounded segments below Fort Benton, as well as portions of two impounded and one channelized segment. The purpose of this study was to examine a physician's recommendation for ColoRectal Cancer screenings among Caucasians and minorities (African American, Asian American, and Latino). Past research on CRC has suggested that a history of CRC in a first degree relative elevates a person's lifetime risk. Furthermore, without a physician recommendation for early screening, the patient's chances of early detection are substantially lowered. This is why it is imperative to examine the complex factors that contribute to minorities who have lower CRC screening rates compared to Caucasians.

This thesis defines the Emergent Church movement and discusses how it is situated among American Evangelical Christendom (A.E.C.). Furthermore, this thesis explores how Emergent Church texts use the rhetoric of conversation to question established biblical foundations. Through conversations the Emergent Church uses discourse that differs from traditional A.E. Christian language. Within these differences postmodern threads emerge. The theoretical framework for this study is Jean-Francois Lyotard's "The Postmodern Condition: A Report on Knowledge". Two key authors considered are Rob Bell and Brian McLaren.

This thesis contends that the notion of an uncanny valley can be extended to explain the lack of lesbian presence in the traditional dramatic canon. Because mainstream theatre audiences are unfamiliar with lesbian dramatic representation, lesbian theatre often provokes an uncanny valley response in general audiences and this negative response leads to little commercial success for lesbian plays. California recently declared a state of emergency over drought followiing three years of below average rain and snowfall. There is a consensus among many involved in water supply in California for finding new sources of water, conservation, and increasing recharge to replenish the groundwater with surface water that would have been lost to run off. This study investigates the amount of stormwater that could potentially be reinvested as groundwater recharge by simply transforming selected impermeable parking lots of the commercial, industrial, and institutional sectors into pervious pavement that allow water to infiltrate rather than be lost as urban runoff.

Methylphenidate is the preferred treatment for attention-deficit hyperactivity disorder in preschool-aged children, even though few human studies have assessed the long-term consequences of early methyphenidate use. In rats, exposure to methylphenidate during a developmental period that approximates early childhood increases morphine-induced antinociception in adulthood. However, because morphine activates three distinct opioid receptors it is unknown which receptor is resonsible for this methylphenidate effect. The purpose of the present study was to assess the long-term effects of early methylphenidate exposure on DAMGO- and U50, 488-induced antinociception, a mu-opioid and kappa-opioid selective agonist.

This study investigated whether or not having a permanent connection was beneficial to young adults after they left foster care. It showed that even with the presence of an identified mentor to guide youth post-emancipation, these youth were still facing serious difficulties.

According to the Center for Disease Control (2005), African American women account for 64% of the nearly 127,000 women living with HIV/AIDS in the United States. The author conducted qualitative interviews with 30 adult Black males with a history of practicing Down Low (DL) sexual behavior-- engaging in homoerotic activities with their female partners. The purpose of this study is to provide insight into the DL subculture and to obtain useful information that is crucial to understanding how the DL male's personal struggles with sexuality and masculinity contribute to the disporportionate amount of Black women contracting HIV/AIDS.

In the eastern United States, two invasive specialist insects share a native host plant, Eastern hemlock, Tsuga canadensis. In recent years, much research has focused on the impacts of the hemlock woolly adelgid (Adelges tsugae) because of the detrimental effects it has on hemlock growth and survival. In contrast, the invasive elongate hemlock scale (Fiorinia externa) is thought to have only minor impacts on hemlock. The bacterium Borrelia burgdorferi, the causative agent of Lyme disease, is a limited-genome organism that must obtain many of its biochemical building blocks, including N-acetylglucosamine (GlcNAc), from its tick or vertebrate host. GlcNAc can be imported into the cell as a monomer or dimer (chitobiose), and the annotation for several B. burgdorferi genes suggests that this organism may be able to degrade and utilize chitin, a polymer of GlcNAc. We investigated the ability of B. burgdorferi to utilize chitin in the absence of free GlcNAc, and we attempted to identify genes involved in the process. We also examined …

Molecular-assisted alpha taxonomy has recently become an effective practice in reassessing biodiversity and floristics for a variety of different organisms. This paper presents a series of examples that have been drawn from biodiversity work being carried out on the marine red algae of Bermuda. Molecular sequencing of DNA from Bermuda samples has already begun to greatly alter the makeup of the flora as it was known just decades ago, and will help set a new database for future comparison as climate change affects species composition in the islands.

Chromatin remodeling is an essential part of transcription initiation. We show that at heat shock gene promoters functional interactions between individual ATP-dependent chromatin remodeling complexes play critical role in both nucleosome displacement and Pol II recruitment. Using HSP12, HSP82 and SSA4 gene promoters as reporters, we demonstrated that while inactivation of SNF2, a critical ATPase of the SWI/SNF complex, primarily affects the HSP12 promoter, depletion of STH1- a SNF2 homolog from the RSC complex reduces histone displacement and abolishes the Pol II recruitment at all three promoters. The present experiment examined the influence of excitotoxic lesions of the basolateral amygdala (BLA) on morphine-induced saccharin avoidance. Neurologically intact subjects rapidly learned to avoid drinking the taste conditioned stimulus (CS), an effect that was sustained throughout the experiment. Although the BLA-lesioned (BLAX) rats showed CS avoidance over the first few trials, the effect was not sustained. That is, by the end of the experiment, the BLAX rats were drinking the same amount of saccharin after seven saccharin-morphine trials as they did on the first trial (i.e., prior to the morphine injections). Potential interpretations of the results are discussed including …

OBJECTIVE: To review evidence for the use of intrathecal rituximab and trastuzumab in the management of leptomeningeal carcinomatosis.
DATA SOURCES: A search of MEDLINE (1966-July 2010) and International Pharmaceutical Abstracts (1970-July 2010) was performed using search terms intrathecal, trastuzumab, rituximab, and monoclonal antibody. Additionally, American Society of Clinical Oncology, San Antonio Breast Conference, American Association for Cancer Research, and American Society of Hematology meeting abstracts were searched.
STUDY SELECTION AND DATA EXTRACTION: Publications were reviewed for inclusion. Those reporting use of rituximab and trastuzumab intrathecally are reviewed and include 1 Phase 1 trial, 2 small prospective studies, 1 case series, …
