Electronic Theses and Dissertations
The dissertation will give an introduction, background and our research progress in the areas of antifolates. The design and synthesis of both classical and nonclassical bicyclic furo[2,3-d]pyrimidines, pyrrolo[2,3-d]pyrimidines and tricyclic pyrimido[4,5-b]indoles as potential antifolates have been described. As a part of this study, a total of one hundred and fifty-three new compounds have been synthesized and characterized. Of these, sixty-seven final compounds were submitted for biological evaluation.

Background: MicroRNAs (miRs) are non-coding RNA molecules involved in post-transcriptional regulation, with diverse functions in tissue development, differentiation, cell proliferation and apoptosis. miRs may be less prone to degradation during formalin fixation, facilitating miR expression studies in formalin-fixed paraffin-embedded (FFPE) tissue.
Results: Our study demonstrates that the TaqMan Human MicroRNA Array v1.0 (Early Access) platform is suitable for miR expression analysis in FFPE tissue with a high reproducibility (correlation coefficients of 0.95 between duplicates, p < 0.00001) and outlines the optimal performance conditions of this platform using clinical FFPE samples. We also outline a method of data analysis looking at differences in miR abundance between FFPE and fresh-frozen samples. By dividing the profiled miR into abundance strata of high (Ct35), we show that reproducibility between technical replicates, equivalent dilutions, and FFPE vs. frozen samples is best in the high abundance stratum. We also demonstrate that the miR expression profiles of FFPE samples are comparable to those of fresh-frozen samples, with a correlation of up to 0.87 (p < 0.001), when examining all miRs, regardless of RNA extraction method used. Examining correlation coefficients between FFPE and fresh-frozen samples in terms of miR abundance reveals correlation coefficients of up to 0.32 (low abundance), 0.70 (medium abundance) and up to 0.97 (high abundance).
Conclusion: Our study thus demonstrates the utility, reproducibility, and optimization steps needed in miR expression studies using FFPE samples on a high-throughput quantitative PCR-based miR …

Background: The family plays a vital role in shaping adolescent behaviours. The present study investigated the associations between family structure and substance use among Hong Kong Chinese adolescents.
Methods: A total of 32,961 Form 1 to 5 (grade 7-12 in the US) Hong Kong students participated in the Youth Smoking Survey in 2003-4. An anonymous questionnaire was used to obtain information about family structure, daily duration of parent-child conversation, smoking, alcohol drinking and drug use. Logistic regression was used to calculate the adjusted odds ratios (OR) for each substance use by family structure.

This paper will be an analysis of the psychological disorder of major depression. First, the classification of this disorder using the DSM-IV-TR will be reviewed. The areas of etiology, differential diagnosis, and treatment will then be considered. Following, a theory-specific approach to the disease including theoretical framework, diagnosis and treatment, and outcomes of using this approach, will be examined. Finally a personal reflection on the subject of major depression, and lessons learned from this project, will be discussed.

Thousands of children throughout the United States are currently awaiting placement with adoptive families. The literature indicates that gay- and lesbian-headed households can well meet the needs of these children. Research suggests that sexual prejudice, religious fundamentalism and attitudes about gay and lesbian adoption may influence practice decisions regarding placement. This dissertation study examined the influences of religious fundamentalism, sexual prejudice, contact with sexually diverse individuals, and attitudes towards gay men and lesbians as adoptive parents on intent to place children in need with gay and lesbian couples. In this study, the cell lines HeLa, 3T3-Swiss albino, CHO-Kl, and ARPE-19 were cultured and co-incubated with variable concentrations of S-phosphorylated thiols and Nacetylcysteine. Using MTT assays, the cultures were analyzed to determine the viability and proliferation of the cultured cell lines. Additionally, the dose measurement of the ¹³⁷Cs Gamma Source used in this experiment was calculated in order to prepare for future studies. Based on the data from this experiment, further testing can be conducted to determine the effect that the cell lines co-incubated with s-phosporylated thiols has compared to the control when exposed to ultraviolet and gamma radiation. …

Margonis (1986) criticizes Heidegger’s philosophy and those who would attempt to adopt his views for the purpose of thinking education because of the "abstract nature of his discussions," which suggest "proposals regarding our political, economic and educational lives from the place of metaphysical argumentation" (p. 125). To the contrary, Dwyer, et al (1988) claim the Heidegger’s philosophy, "clearly suggests an educational theory" (p. 100). This, is perhaps an overly optimistic claim, for it glosses over the difficulty associated with plumbing the depths of Heidegger’s vast corpus in order to speculate on the legitimate potential his philosophy has for contemporary educational …

Background: Strong patterns of habitat association are frequent among tropical forest trees and contribute to the maintenance of biodiversity. The relation of edaphic differentiation to tradeoffs among leaf functional traits is less clear, but may provide insights into mechanisms of habitat partitioning in these species rich assemblages.
Methodology/Principal Findings: We quantify the leaf economics spectrum (LES) for 16 tree species within a Bornean forest characterized by highly pronounced habitat specialization. Our findings suggest that the primary axis of trait variation in light-limited, lowland tropical forests was identical to the LES and corresponds with the shade tolerance continuum. There was no …

Extracellular RNA is becoming increasingly recognized as a signaling molecule. Virally derived double stranded (ds)RNA released into the extracellular space during virus induced cell lysis acts as a powerful inducer of classical type I interferon (IFN) responses; however, the receptor that mediates this response has not been identified. Class A scavenger receptors (SRAs) are likely candidates due to their cell surface expression and ability to bind nucleic acids. In this study, we investigated a possible role for SR-As in mediating type I IFN responses induced by extracellular dsRNA in fibroblasts, a predominant producer of IFNb. The present study examined the effect of dissolved organic carbon (DOC), both added and inherent, on Cu toxicity in glochidia, the larvae of freshwater mussels. Using incremental additions of natural DOC concentrate and reconstituted water, a series of acute copper toxicity tests were conducted. An increase in DOC from 0.7 to 4.4 mg C/L resulted in a fourfold increase (36–150 μg Cu/L) in the 24-h median effective concentration (EC50) and a significant linear relationship (r2=0.98, p=0.0008) between the DOC concentration and the Cu EC50 of Lampsilis siliquoidea glochidia. When Counting Sheep Becomes Counting Worries: Insomnia, Rumination, And Depression, Huiting Lui, Partrick Rich, Emily Maurer, Shivani Santoki
Modern Psychological Studies
Previous studies have examined how both external and internal factors are related to sleep disorders. Internal factors such as depression have been linked to insomnia and other sleep disorders. The current study examines the relationship between rumination, insomnia, and depression. The study also examines how sleep rumination, the act of persistently worrying about how much sleep one is getting, might affect the relationship between insomnia and depression. In the results, depression was strongly correlated with rumination (r = .635, p < .001). Though there was no correlation between sleep rumination and depression, there was a significant correlation between depression and sleep discrepancy, the difference between the amount of sleep and the amount of sleep desired (r = .51, p < .001). This finding suggests that more research should be conducted on sleep discrepancy to see how it may relate to other disorders and psychological concepts.

The presidency of Jose Luis Zapatero Rodriguez has seen significant legisla tion in Spain that includes the legalization of same-sex marriage in 2005 and La ley de la memoria historica in 2007.1 Upon consideration of the changes in Spain during the first decade of the twenty-first century, we as critics must pause to consider how literature and film respond to the topics of homosexuality and history. La enfermedad respiratoria es la segunda causa de muerte (diarrea es la primera) en las terneras lecheras no destetadas. Los problemas respiratorios han aumentado en un 34 por ciento en los últimos 20 años, causando cerca de 21 por ciento de la mortandad de los terneros (NAHMS 2007). Las terneras que sobreviven continúan con un desempeño pobre al convertirse en vacas adultas. Para prevenir este problema, es importante tener en cuenta tanto causas predisponentes como determinantes.

In this article, the author examines conflicts over whether to maintain a brain dead pregnant woman on life-sustaining treatment. The author cautions that on the rare occasions when courts are confronted with such a conflict, they should employ a consistent methodology for resolution of the conflict and attempt to honor the wishes of the post-mortem mother and her family. The author draws on relevant areas of law to demonstrate the existence of a legal fiction that protects the interests of post-mortem pregnant women in refusing medical treatment. This article discusses how pharmaceutical innovation achieves remarkable improvements in human health but a significant portion of the U.S. population cannot afford prescription drugs. The author examines ways that patent protection, generics, supply chain complexity, and the cost of innovation and promotion affect access and affordability. The author then looks at the influences of marketing strategies and industry trends such as the patent cliff and pipeline for new drugs, innovations in biotechnology and genomics, comparative effectiveness analysis, and payor and employer strategies on drug prices. Rationing Health Care: It's A Matter Of The Health Care System's Structure, David Orentlicher

The article examines two primary policy proposals for how the U.S. should allocate its limited health care dollars: a centralized model in which a commission establishes rationing guidelines, and a decentralized model in which rationing decisions are made by health care providers on a case by case basis. The author finds significant advantages with each position, leading the author to assert that a combination of each is key to an effective rationing policy: a centralized control of structure coupled with decentralized physician-level decision making. While mindful that formal rationing guidelines alone are unfeasible to effectuate cost-effective care, the author introduces …
