Purification Process Development Of Protein Subunit Based Vaccine Candidates Produced Using Recombinant E. Coli Expression System (Part - I), Davinder Chawla, Yan-Ping Yang

Vaccine Technology III

Proteins are key components of prophylactic vaccines against infectious diseases. Protein subunit based vaccine is an attractive alternative to the traditional detoxified bacterial or inactivated viral vaccine approach due to its highly purified and well characterized product nature. Purification of protein antigens to achieve consistent product purity and quality is an integral part of the protein subunit vaccine product development process. Expression levels of the recombinant proteins in bacterial expression system may be extremely high following rapid technology advancement. The challenges and approaches used to develop purification processes for novel protein vaccine candidates expressed at g/L level will be discussed.

Development Of Recombinant Protein Based Chemical Conjugate Malaria Vaccines Targeting The Pre-Erythrocytic Stage, Transmission Blocking, Or Both, David L. Narum, Nicholas Macdonald, David Jones, Ruth Ellis, Yimin Wu, Patrick E. Duffy

Vaccine Technology III

The development of a Plasmodium falciparum malaria vaccine is critical for future control and elimination programs. Recombinant protein based chemical conjugate vaccines, covering different parasite stages, are being developed due to complexity of the parasite and sub-optimal immunogenicity of recombinant malaria proteins in humans, respectively. Chemical conjugation of recombinant malaria proteins to carrier proteins improves their immunogenicity in animal studies. A transmission blocking vaccine comprised of the ookinete protein Pfs25 chemically conjugated to Pseudomonas aeruginosa ExoProtein A (EPA) is currently being developed for pilot scale cGMP production. Bulk lots of Pfs25 and EPA have already been produced and released following …

A Functional Calcium-Transporting Atpase Encoded By Chlorella Viruses, Maria Cristina Bonza, Holger Martin, Ming Kang, Gentry L. Lewis, Timo Greiner, Sonia Giacometti, James L. Van Etten, Maria Ida De Michelis, Gerhard Thiel, Anna Moroni

James Van Etten Publications

Calcium-transporting ATPases (Ca2+ pumps) are major players in maintaining calcium homeostasis in the cell and have been detected in all cellular organisms. Here, we report the identification of two putative Ca2+ pumps, M535L and C785L, encoded by chlorella viruses MT325 and AR158, respectively, and the functional characterization of M535L. Phylogenetic and sequence analyses place the viral proteins in group IIB of P-type ATPases even though they lack a typical feature of this class, a calmodulin-binding domain. A Ca2+ pump gene is present in 45 of 47 viruses tested and is transcribed during virus infection. Complementation analysis of …

Mitochondria-Centric Activation Induced Cell Death Of Cytolytic T Lymphocytes And Its Implications For Cancer Immunotherapy, Arvind Chhabra Jun 2010

Mitochondria-Centric Activation Induced Cell Death Of Cytolytic T Lymphocytes And Its Implications For Cancer Immunotherapy, Arvind Chhabra

UCHC Articles - Research

Premature death of the adoptively transferred cytolytic T lymphocytes (CTL) by means of activation induced cell death (AICD) represents one of the major constraints in devising an effective anti-cancer immune intervention strategy. Understanding the mechanism of AICD is, therefore, critical for developing methods to interfere with this death process. Although the existing paradigm on AICD centers around the initiation of the cascade of events originating from the engagement of death receptors leading to the activation of effector caspases and eventually resulting in cell death, recent findings have questioned the universal role of caspases as the cell death executioners. We here …

Recognizing And Managing Common Crop Disease In South Dakota, Martin A. Draper, Darrel Deneke Jun 2010

Recognizing And Managing Common Crop Disease In South Dakota, Martin A. Draper, Darrel Deneke

SDSU Extension Extra Archives

Diseases of plants may be expressed differently under varying environmental conditions or in various host species. Disease is the outward expression of an infection, yet in some cases there may be no outward evidence of disease. Outward expression may be some deviation from normal growth, change in appearance, or alteration of the quality or value of the crop. Plant diseases may be caused by biotic agents, infectious microorganisms, or abiotic causes such as chemical deposition, nutrient imbalances, or environmental factors. This publication concentrates on biotic or infectious diseases and provides information on common abiotic problems. Infectious plant diseases may be …

Spirituality As A Life Line: Women Living With Hiv/Aids And The Role Of Spirituality In Their Support System, Jennifer L. Peterson, Malynnda A. Johnson, Kelly E. Tenzek Jun 2010

Spirituality As A Life Line: Women Living With Hiv/Aids And The Role Of Spirituality In Their Support System, Jennifer L. Peterson, Malynnda A. Johnson, Kelly E. Tenzek

Journal of Interdisciplinary Feminist Thought

For many women living with HIV/AIDS, incorporating spirituality into their lives helps them organize their experience, empower self-reconstruction, and manage stigma. (Stanley, 1999). Because of the potential relationship of spirituality and social support, the specific aim of this study is to examine the role of spirituality in the support experiences of women living with HIV or AIDS. In this case, spirituality or a connection to God, offered the women an opportunity to develop meaning and perspective taking, to have a source of support, to provide control through a more powerful being, and to offer a path to community.

Sizing Up Pharmacotherapy For Obesity., Michael A. Valentino, Andre Terzic, Scott A. Waldman

Department of Pharmacology and Experimental Therapeutics Faculty Papers

Obesity has increased over the last 20 years, from a condition affecting only a small portion of populations in developed countries, into a global pandemic. The impact of obesity can be appreciated in the context of the populations at risk, and it is estimated that >1 billion adults worldwide are overweight (BMI >25 kg/m2), 300 million of whom are clinically obese (BMI >30 kg/m2). In the United States, 65% of adults are overweight, and 32.2% of them are obese, a prevalence that has doubled over 20 years. In industrialized countries, obesity rates have tripled, coinciding with adoption of a Western …

Two-Sided Exact Tests And Matching Confidence Intervals For Discrete Data, Michael P. Fay Jun 2010

Two-Sided Exact Tests And Matching Confidence Intervals For Discrete Data, Michael P. Fay

The R Journal

There is an inherent relationship between two-sided hypothesis tests and confidence intervals. A series of two-sided hypothesis tests may be inverted to obtain the matching 100(1-)% confidence interval defined as the smallest interval that contains all point null parameter values that would not be rejected at the α level. Unfortunately, for discrete data there are several different ways of defining two-sided exact tests and the most commonly used two sided exact tests are defined one way, while the most commonly used exact confidence intervals are inversions of tests defined another way. This can lead to inconsistencies where the exact test …

What Leona Helmsley Can Teach Us About The Charitable Deduction, Ray D. Madoff Jun 2010

What Leona Helmsley Can Teach Us About The Charitable Deduction, Ray D. Madoff

Chicago-Kent Law Review

Leona Helmsley named a number of beneficiaries under her will (both human and canine), but among the unnamed beneficiaries are scholars interested in studying the role of philanthropy in the United States. By directing that an estimated $8 billion be used for the benefit of dogs, Mrs. Helmsley brought in to high relief policy issues regarding the appropriateness of the unlimited charitable deduction. I argue that these concerns are equally applicable, albeit less obvious, when it comes to more traditional charitable bequests. In this paper I will discuss the appropriateness of the unlimited estate tax deduction (particularly in light of …

Ethnobotanical Leaflets

Nine plant species belonging to the Euphorbiaceae family and used in traditional medicine in Akwa Ibom State of Nigeria were evaluated for in vitro antimicrobial activity using agar diffusion method. The stem bark of Maesobotrya dusenii gave the most significant effect followed by its root bark. The inhibitory effect of M. dusenii stem bark extract (37 mm) on Pseudomonas aeruginosa was higher than that of Chloramphenicol (35 mm). However, Alchornea laxiflora leaf extract showed the weakest activity. The minimum inhibitory concentration of the extracts ranged between 12.5 and 250 μg/mL. The results of the antimicrobial effects validated the use of …

Peer Reviewed Articles

The reduction of infant mortality (IM) continues to be a primary goal throughout much of the developing world, and a suite of methods have been proposed with which to achieve it. However, most attempts at IM reduction have taken place within the context of an almost bewilderingly complex regimen of simultaneous changes occurring in the physical environment, sociopolitical structure, and health care. Which of these various elements is truly causal in reducing IM and which may be merely a correlated but nonprimary agent? During the past two decades Mexico has constituted an extraordinary field laboratory with which to decipher the …

“‘Not In Canada’: The Non-Ebola Panic And Media Misrepresentation Of The Black Community”, Charles T. Adeyanju, Temitope Oriola Jun 2010

“‘Not In Canada’: The Non-Ebola Panic And Media Misrepresentation Of The Black Community”, Charles T. Adeyanju, Temitope Oriola

African Journal of Criminology and Justice Studies

This paper investigates the case of a non-Ebola panic in Hamilton, Ontario in 2001. Using in-depth interviews of journalists, hospital staff, members of the Congolese-Black community and content analysis of coverage by four Canadian newspapers, this study shows that the media problematized the case by cross-articulating a health scare of Ebola with immigration, crime and “race”. Utilizing extant “late modernity” literature and its attendant anxiety over risks, the study shows how a “scapegoat” was created, feasted upon by the media and aroused the collective conscience of mainstream Canadian society. Consequently, the voice of the community was misrepresented and underrepresented in …

Therapeutic Modalities And Educational Approaches In Clergy Stress Reduction, Shaun Tress Campbell Jun 2010

Therapeutic Modalities And Educational Approaches In Clergy Stress Reduction, Shaun Tress Campbell

Loma Linda University Electronic Theses, Dissertations & Projects

Clergy are listed among administrators, social workers, healthcare providers, fire fighters, and police officers as professionals with high levels of occupational stress (Hill et al., 2003; Greenberg, 1990). Clergy are valuable resources to the American culture in that they perform an array of services to comfort and assist others. From natural catastrophes to personal misfortune, pastors are often expected to be present to offer consolation or to extend ecclesiastical services. Researchers have found that pastors are the primary mental health counselors for tens of millions of Americans (Weaver et al., 2002). Darling et al. (2004) report, "Clergy in U.S. culture …

Global Warming: Myths & Realities, Dennis Jaynes

Social Sciences

The topic of Global Warming has been surrounded with controversy since it was first proposed. From evidence that has come from years of studies, a majority of the world’s governments and scientists are in agreement that the Earth’s global temperatures are warming. However, there are others who are skeptical of the science; they spread their message by trying to discredit the scientists or have even gone as far as to cover it up. The intent of this paper is to examine the myths and compare them side by side with the latest scientific data to determine what is real and …

Influence Of Type 2 Bovine Viral Diarrhea Virus NPro On Enhancement Of Bovine Respiratory Syncytial Virus Replication Mediated By Antagonism Of Host Cell Interferon Type I Responses, Abdulrahman Abdulaziz A. Alkheraif Jun 2010

Alkheraif

School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research

Bovine viral diarrhea virus (BVDV) is a member of the genus Pestivirus, Family Flaviviridae. The virus can infect many species of animals of the order Artiodactyla. The BVDV genome encodes an auto protease, Npro, that degrades interferon regulatory factor-3 (IRF-3) reducing type I interferon (IFN-I) production from host cells. Bovine respiratory syncytial virus (BRSV) is a member of the genus Pneumovirus, Family Paramyxoviridae. Concurrent infection with BVDV and BRSV causes more severe respiratory and enteric disease than infection with either virus alone. Our hypothesis was that Npro modulates the innate immune responses to BVDV infection …
