Children's skeletons mature at different rates, and they can be affected by a variety of factors including disease, hormone imbalance or genetics. The assessment of skeletal maturity is a frequently performed procedure that allows the detection of hormonal, growth or genetic disorders. Several methods have been developed to estimate skeletal maturity. Most methods evaluate hand/wrist radiographs using indicators such as the ratios of various bone widths, the onset of the ossification of epiphysis and epiphyseal-diaphyseal fusion. This study determined the tissue-specific effect of adipose stem cells (ASC) within a melanoma environment for development of a cell-based melanoma therapy. Analysis included a subcutaneous B16-F1 melanoma model using thirty-one C57BL/6 wild-type mice. Melanoma xenografts were treated with cell-based therapies of CFDA-SE-labeled human fibroblasts HF20x (control), non-differentiated ndASC or hematopoietic-differentiated HdASC. No tumor regression was observed in presence of cell-based therapies, thus, the HdASC group demonstrated an increase in tumor growth accompanied with an up-regulated macrophage response, and increased angiogenesis. In addition, this group demonstrated a decrease in Melan-A tumor marker and interferon-γ expression suggesting that ASC-supported tumor angiogenesis …

Chlorinated aliphatic hydrocarbons (CAHs) are often found as groundwater contaminants because of past industrial activities and disposal practices. CAHs pose a threat to human health and thus, create a need to find both natural and engineered processes that can remove these chlorinated compounds from the environment. Natural attenuation by oxidative biodegradation is especially important because it can allow for mineralization to carbon dioxide, a nontoxic end-product. Charaterization Of Chlorpyrifos Toxicity On The Pancreatic Beta Cell Line Rinm5f, Zhongyu Yan

Chlorpyrifos (CPF) is a well-known organophosphate (OP) insecticide that inhibits acetylcholinesterase (AChE). Organophosphates have been shown to induce hyperglycemia in both humans and animals that cannot be explained by AChE inhibition alone. Pancreatic β-cells are responsible for secreting insulin and playing a fundamental role in glucose homeostasis. The aim of this research was to investigate CPF toxicity on the insulin secreting β-cell line RINm5f (RIN). In the first part of this dissertation, we determined that cytotoxic concentrations of CPF between 50 µM and 200 µM induced a dose-dependent increase in RIN cell apoptotic and necrotic death (p<0.05). Lower doses, i.e., 5-25 µM did not induce apoptosis or necrosis. In further mechanistic studies, inhibitors of c-Jun N-terminal kinase (JNK) and P38 pathways, i.e., SP600125 and SB202190, successfully attenuated CPF-induced apoptosis (p<0.05), suggesting cell death is mediated by JNK and/or p38. In addition, N-Acetyl-L-cysteine (NAC), an antioxidant, inhibited apoptosis suggesting that oxidative stress is also involved in RIN cell death. In the second part of this dissertation, low dose CPF's effects on insulin synthesis and secretion was studied. Our results showed that RIN cells treated with CPF for 4 hr exhibited a dose-dependent decrease in insulin secretion in response to stimulation by 50 mM potassium (p<0.05). Insulin secretion stimulated by 25 mM glucose also showed significant decreases upon incubation with various doses of CPF (p<0.05). Insulin synthesis involves proteolytic cleavage of proinsulin at two specific sites. The enzymes responsible for promoting proinsulin cleavage, i.e, prohormone convertase (PC) 1/3 and 2 (PC2), contain a serine hydrolase catalytic triad, His-Asp-Ser, similar to AChE. CPF modification of PC enzymes similar to AChE, could potentially interrupt the conversion of proinsulin to insulin and disrupt insulin secretion. PC2 enzyme activity was significantly inhibited by CPF at concentrations as low as 10 µM (p<0.01). RIN cells, for the first time, were found to secrete proinsulin was through a constitutive instead of a regulated pathway. In summary, our results show that CPF not only exerts a direct cytotoxic effect at concentrations above 50 µM, but also significantly impairs insulin secretion at lower doses. Low dose CPF could potentially affect endocrine function in vivo by inhibiting the proinsulin processing enzyme PC2.

A shortage of housing affordable to low- and moderate-income households in job-rich suburbs is one of the biggest obstacles to upward mobility for those households, yet our understanding of the factors influencing the distribution of affordable housing development is weak. Focusing on suburbs in Hennepin County, Minnesota, I use qualitative case studies of eight cities and a statistical analysis of 39 to demonstrate that the geography of economic incentives to develop affordable units has the strongest influence. Contrary to common beliefs, opposition of middle-class suburbanites to affordable housing is a weak factor. Public policy should therefore focus on altering economic …

Platinum group metals (PGMs) have been shown to accumulate in various tissues of organisms but their toxicity is not well-known. Raman spectroscopy is a powerful analytical technique that enables direct and non-destructive chemical characterization of bone tissues. PGMs were injected into chick embryos in the following concentrations: 0.1, 1.0, 5.0, or 10.0 ppm solutions of Pt(IV), Rh(III), Pd(II) or PGMs mixtures (0.1 ppm or 1.0 ppm of each metal). Nuclear magnetic resonance (NMR) based metabolomics is a developing research field with broad applicability, including the identification of biomarkers associated with pathophysiologic changes, sample classification based on the mechanism of toxicity, and clinical diagnosis. Intrinsic to these applications is the need for statistical and computational techniques to facilitate the associated data analysis. Further, a typical 1H NMR spectrum of pure proteins, biofluids, or tissue may contain thousands of resonances (i.e., peaks), thus, a pure visual inspection is insufficient to fully utilize the spectral information.

Endocrine disruption is a complex phenomenon in the sense that endocrine disrupting chemicals (EDCs) are known to act via multiple modes of action, but the mechanisms of actions are poorly understood. In crustaceans, calcium (Ca2+) apart from its role as a second messenger, is also a major constituent of the calcified exoskeleton which undergoes a periodic mineralization/demineralization process known as the molting cycle. This special feature examines synergies and trade-offs among these objectives. A prototype multi-stage cyclone system consisting of an impaction inlet and five axial flow cyclone stages has been developed to classify simulants of Lunar and Martian dusts for various research and development needs of NASA’s space exploration missions. Individual axial flow cyclone stages can be either independently operated with an inline connection to other particle devices or cascaded together for particle separation and collection. The Nebraska Sand Hills are a distinctive eco-region in the semi-arid Great Plains of the western United States. The water table underlying the Sand Hills is part of the High Plains/ Ogallala aquifer, an important water resource for the central Great Plains. Lake levels are affected directly by fluctuations in the water table, which is recharged primarily by local precipitation and responds quickly to climatically induced changes in regional water balance. Instrumental records are available for only 50–100 years, and paleolimnological data provide important insights into the extremes and variability in moisture balance over longer time scales. Recent decisions--all relying on a stylized example first provided by the Ortho court--hold that a multi-product seller that uses a bundled discount in a way that excludes an equally or more efficient competitor engages in predatory bundling. According to these decisions, a bundle can be considered “predatory” even when the price of the bundle exceeds its cost. This Article shows that the Ortho court's stylized example and its monopoly leveraging theory are erroneous. The sale and distribution of turtles with a carapace smaller than four inches in diameter was banned by federal law in 1975 on the grounds that such animals were frequently associated with human Salmonella infections. However, the popularity of these small turtles as pets has resurfaced, and in many places in Michigan they are being sold illegally. Type I cells are one of two main cell types located within the carotid body. These cells respond to hypoxia, hypercapnia, and acidosis by releasing excitatory and inhibitory neurotransmitters. This causes increased firing of the carotid sinus nerve and restores blood gas levels to their physiological values. While previous studies have shown whether individual neurotransmitters are excitatory or inhibitory, this work demonstrates how the interplay between two neurotransmitters may potentially shape the output of the carotid body. In this research, a state-of-health (SOH) monitoring method is developed for automotive EPGS system, focusing on alternator related faults. Parity relations, characterizing the correlation among EPGS system signals under normal operating conditions, are generated using principal/minor component analysis techniques. The Importance Of Perceived Similarity Within Faculty-Faculty Mentoring Dyads, Emily N. Polander

This study investigated the effects of gender similarity, perceived similarity, and relationship type (formal vs. informal) within faculty-faculty mentoring dyads on various mentoring outcomes from the protégé's perspective. Perceived similarity was expected to be a stronger predictor of relationship satisfaction, affective commitment, job satisfaction, and turnover intent than gender similarity. Perceived similarity was also examined as a potential mediator of relationship type and relationship satisfaction. Tenure-track faculty who reported having mentors (N = 45) answered questions regarding their primary career mentor and other workplace attitudes. Results indicated that perceived similarity had a positive, greater effect than gender similarity on relationship …

The current study examines the use of a frame-of-reference (FOR) in the evaluation of core self-evaluations, stressors and strains. The study investigates the prediction that using a FOR that matches predictors and outcomes will result in a stronger relationship between the constructs being measured, specifically CSE, stressors, and strains. In addition, the study examines the extent to which following this procedure will add incremental validity after non-contextualized predictors are controlled. This work investigates two questions regarding DNA replication. The first aim examines the interactions of DUE-B with replication proteins and the second explores the cellular effects of replicating a polypurine polypyrimidine sequence in human cells.

DUE-B siRNA decreases the chromatin binding of essential replication proteins Cdc45, PCNA and RPA. DUE-B also co-immunoprecipitates with Cdc45 and TopBP1. In vitro kinase assays suggest that the checkpoint protein ATR may phosphorylate DUE-B. These experiments lend further evidence that DUE-B plays an important role in the initiation of eukaryotic DNA replication.

In this dissertation, I analyze the 20th century text, A Room of One's Own, by Virginia Woolf (2005), and I engage with Woolf's concept of a woman's need for a room of her own in which she can be free to think for herself, study, write, or pursue other interests away from the oppression of patriarchal societal expectations and demands. Through library-based research, I identify four screens in Woolf's work through which she viewed and critiqued culture, and I use these screens to reconceptualize "a room of one's own" in 21st Century terms. The Rhetoric of the Regional Image: Interpreting the Visual Products of Regional Planning investigates the manner in which visual conventions and visual contexts of regional visioning scenarios affect their interpretation by urban and regional planners, who use visual communication to meet the technical and rhetorical demands of their professional practice. The research assesses Central Florida‘s ―How Shall We Grow?‖ regional land use scenario using focus groups and interviews with planning professionals, a corresponding survey of community values, and rhetorical analysis to explore the ―How Shall We Grow?‖ scenario as persuasive communication.
