An Impaired 20s Proteasome Contributes To The Accumulation Of Oxidized Proteins In Multiple Sclerosis And Its Animal Model, Jianzheng Zheng
An Impaired 20s Proteasome Contributes To The Accumulation Of Oxidized Proteins In Multiple Sclerosis And Its Animal Model, Jianzheng Zheng
Biomedical Sciences ETDs
Carbonylated (oxidized) proteins are known to accumulate in the brain of patients with multiple sclerosis (MS) and in the spinal cord of rats with acute experimental autoimmune encephalomyelitis (EAE). Yet, our knowledge regarding mechanism(s) underlying the build-up of protein carbonyls in these inflammatory demyelinating disorders is quite limited. The objectives of this dissertation were (1) to measure the changes in protein carbonylation during disease progression, and to identify the target cells and modified proteins in the cerebellum of EAE animals, prepared by active immunization of C57/BL6 mice with MOG35-55 peptide, (2) to determine if the accumulation of carbonylated proteins in …

Singular Value Decomposition Metrics Show Limitations Of Detector Design In Diffuse Fluorescence Tomography, Frederic Leblond, Kenneth M. Tichauer, Brian W. Pogue
Dartmouth Scholarship
The spatial resolution and recovered contrast of images reconstructed from diffuse fluorescence tomography data are limited by the high scattering properties of light propagation in biological tissue. As a result, the image reconstruction process can be exceedingly vulnerable to inaccurate prior knowledge of tissue optical properties and stochastic noise. In light of these limitations, the optimal source-detector geometry for a fluorescence tomography system is non-trivial, requiring analytical methods to guide design. Analysis of the singular value decomposition of the matrix to be inverted for image reconstruction is one potential approach, providing key quantitative metrics, such as singular image mode spatial …

The goal of this study was to test whether the breeding system and/or the degree of inbreeding of field colonies of the Formosan subterranean termite, Coptotermes formosanus, Shiraki (Isoptera: Rhinotermitidae) influences bacterial load on the cuticle of foraging workers. We enumerated bacterial load on the cuticle of groups of workers foraging in 20 inground monitoring stations surrounding the French Market in New Orleans, LA, and identified bacteria species using 16S rRNA gene sequencing. We used microsatellite genotyping to assign the 20 worker groups to seven simple family colonies (headed by a single pair of reproductives) and four extended family colonies …

Population Forecasting Model Of Nilaparvata Lugens And Sogatella Furcifera (Homoptera: Delphacidae) Based On Markov Chain Theory, Xiang Hui Yan, Huai Liu, Jin Jun Wang, Zhi Mo Zhao, Fangneng Huang, Deng Fa Cheng
Faculty Publications
Nilaparvata lugens (Stål) and Sogatella furcifera (Horváth) are the two most important long-distance migratory insect pests that cause great yield losses to rice in China. Accurate long-term population forecast is needed to implement effective management strategies for these two rice pests. In this paper, a transition probability matrix of 5-yr steps of Markov chain theory was constructed based on 31-yr light-trapping data of the two pests from 1977 to 2007 in Jiangkou County, Guizhou, China. The weight of each step for the transition probability matrix was calculated according to its prediction accuracy. Insect occurrence levels in the sixth year were …

Iota-Carrageenan Hydrolysis By Pseudoalteromonas Carrageenovora Ifo12985, Bernadette M. Henares, Erwin P. Enriquez, Fabian M. Dayrit, Nina Rosario L. Rojas
Chemistry Faculty Publications
We report iota-carrageenan hydrolysis by Pseudoalteromonas carrageenovora IFO 12985. Kappa-carrageenase and lambda-carrageenase were previously isolated from this organism, but iota-carrageenase activity had not been reported in the literature. P. carrageenovora was grown in iota-carrageenan-based liquid medium. Using the zone of depression assay, transfer of aliquots of the culture to solid medium with 2% iota- and kappa-carrageenan showed extensive hydrolysis of iota-carrageenan. Analysis of the hydrolysates by C-13 Nuclear Magnetic Resonance spectroscopy confirmed degradation of the iota-carrageenan. Hydrolytic activity of P. carrageenovora grown in iota-carrageenan was compared with that of the same organism grown in kappa-carrageenan. Cell-free supernatants from each yielded …

A confirmatory method has been developed to allow for the analysis of fourteen prohibited medicinal additives in pig and poultry compound feed. These compounds are prohibited for use as feed additives although some are still authorised for use in medicated feed. Feed samples are extracted by acetonitrile with addition of sodium sulphate. The extracts undergo a hexane wash to aid with sample purification. The extracts are then evaporated to dryness and reconstituted in initial mobile phase. The samples undergo an ultracentrifugation step prior to injection onto the LC-MS/MS system and are analysed in a run time of 26 minutes. The …

The Chestershaw: The Views And Ideas That Shaped The Friendly Rivalry Of G.K. Chesterton And George Bernard Shaw, Jonathan Vowell
Electronic Theses and Dissertations Archive
G.K. Chesterton and George Bernard Shaw both lived at the turn of the twentieth century, which was a time of incredible social and philosophic change and upheaval. In reaction to the social chaos and uncertainty, both men sought to change the world through their artistic and polemical works and thus were friends, yet both men wanted to change the world into different things for different reasons and thus were enemies. Their intellectual, friendly rivalry was a hallmark of early twentieth-century public discourse and English letters. Unfortunately, while their artistic contributions remain, most of their intellectual contributions have been either misunderstood …

"The Young Blood Hungers": Mapping Young Black Manhood In Marita Bonner's Frye Street Fiction, Rachel Leigh Smith
Electronic Theses and Dissertations Archive
Marita O. Bonner, early twentieth century African American public intellectual and creative writer, wrote particularly about the experiences of blacks in Chicago. Though most Bonner scholarship focuses primarily on her working class female characters, this study provides close readings of the young male figures in the short stories, "One Boy's Story," "The Makin's," "The Whipping," "There Were Three," "Tin Can," and "Nothing New." I analyze how these texts confront notions of family, personal identity, and violence, and how Bonner configures young life as a volatile liminal space of human development. As seen in Bonner's short stories and in her essay …

This paper presents techniques for signal conditioning and feature estimation to be used for profiling sensors which perform broad scale classifications. Methods for improving existing feature estimation techniques for linear array profiling systems are provided. A 360 profiling system and the accompanying algorithms used for signal conditioning and feature estimation are presented. The algorithms are validated by demonstrating classification results for the broad scale human, animal, and vehicle three class problem for the linear array, and the human and animal two class problem for the 360 profiling sensor.
Modeling Light-Dark Cycle Memory In The Mammalian Scn, Benjamin L. Coffey
Modeling Light-Dark Cycle Memory In The Mammalian Scn, Benjamin L. Coffey
Electronic Theses and Dissertations Archive
The suprachiasmatic nuclei (SCN), the principal mammalian circadian oscillator, contain several thousand clock neurons that oscillate spontaneously with ~24-hour periods in its ventrolateral region. Without light, this network synchronizes through action potentials releasing VIP, compromising on a free-running period near 24 hours. We entrained Siberian hamsters to various light-dark cycles and tracked their activity into constant darkness showing that they retain memory for a particular light-dark cycle to which they were entrained before returning to their own free-running period. Using Leloup-Goldbeter mammalian clock neurons, we model the ventrolateral SCN network and show that light acting weakly upon a strongly rhythmic …
Stochastic Simulation Of Human Pulmonary Blood Flow Based On Anatomic And Elasticity Data, Jun Shi
Stochastic Simulation Of Human Pulmonary Blood Flow Based On Anatomic And Elasticity Data, Jun Shi
Electronic Theses and Dissertations Archive
In order to fully understand the function of the lungs, it is important to determine the transit time of the pulmonary circulation. However, a practical model to estimate the transit time is difficult to be found in literature, because of the high complexity of the whole pulmonary circulation system. This study developed a computing model of steady blood flow in human lungs based on detailed morphology and elasticity data by Huang et al (1996) [5], which incorporated a Diameter-Defined Strahler System to simulate a human lung branching system. A stochastic simulation approach was introduced. Considering Huang’s connectivity data between different …

Efficient Separation And Sensitive Detection Of Listeria Monocytogenes Using Magnetic Nanoparticles, Microfluidics And Interdigitated Microelectrode Based Impedance Immunosensor, Damira Ashimkhanovna Kanayeva
Graduate Theses and Dissertations
Listeria monocytogenes continues to be a major foodborne pathogen that causes food poisoning and sometimes death in immunosuppressed people and abortion in pregnant women. Nanoparticles have recently drawn attentions for use in immunomagnetic separation techniques due to their greater surface area/volume ratio and better stability against sedimentation in the absence of a magnetic field. Interdigitated microelectrodes and microfluidics make material transfer more efficient and biological/chemical interaction between the surface and solution phase much quicker. Magnetic nanoparticles (Fe3O4) with a 30 nm diameter were functionalized with rabbit anti-L. monocytogenes antibodies via biotin-streptavidin bonds and then amalgamated with target bacterial cells …

Applications Of Site-Specific Recombination Systems In Transgene Expression And Marker Gene Removal, Mehmet Aydin Akbudak
Applications Of Site-Specific Recombination Systems In Transgene Expression And Marker Gene Removal, Mehmet Aydin Akbudak
Graduate Theses and Dissertations
Site Specific Recombination systems, such as FLP-FRT and Cre-lox, have been successfully used for site-specific gene integration and marker-gene deletion in plant systems. They are very useful tools in the integration of single-copy full-length transgene cassettes into the genome because the transgene integration via conventional methods often generate multi-copy locus. Such complex locus containing direct and inverted repeats of full-length and truncated copies of the transgene cassette generate aberrant RNA resulting in gene silencing. Therefore, for stable gene expression, a single copy transgene locus is preferred. However, even single copy locus sometimes succumbs to gene silencing. Although the mechanism is …

Characterization Of Neuronal Groups Regulating Sexual And Agonistic Behavior In Male Chicken (Gallus Gallus), Jingjing Xie
Graduate Theses and Dissertations
The study aimed to understand the neuronal regulation of male sexual and agonistic behavior in broiler breeders. First, brain structures associated with sexual and agonistic behavior were identified by mapping Fos expression. The ventromedial subnucleus of medial portion of bed nucleus of the stria teriminalis (BSTM2) was specifically activated by male courtship behavior. The medial preoptic nucleus (POM) and lateral septum (SL) were associated with both sexual and agonistic behaviors. The bed nucleus of the pallial commissure (NCPa) and the paraventricular nucleus (PVN) were closely related to stress. Second, Fos-ir neurons were phenotyped by double labeling Fos with aromatase (ARO) …
