Pharmaceutical Sciences Faculty Publications

Background: The use of neo-adjuvant chemotherapy in treating osteosarcoma has improved patients’ average 5 year survival rate from 20% to 70% in the past 30 years. However, for patients who progress after chemotherapy, its effectiveness diminishes due to the emergence of multi-drug resistance (MDR) after prolonged therapy.

Methodology/Principal Findings: In order to overcome both the dose-limiting side effects of conventional chemotherapeutic agents and the therapeutic failure resulting from MDR, we designed and evaluated a novel drug delivery system for MDR1 siRNA delivery. Novel biocompatible, lipid-modified dextran-based polymeric nanoparticles were used as the platform for MDR1 siRNA delivery; and the efficacy …

Background: Birth weight is negatively associated with cardiovascular diseases and diabetes, but the associations are less well-established in developing populations where birth weight is often unavailable. We studied the association of birth weight and cardiovascular risk, using birth rank as an instrumental variable, in Southern China.

Methods: We used published data on birth weight by birth rank from an appropriate population and baseline data from the Guangzhou Biobank Cohort Study phases 2 & 3 (2005-8) to examine the adjusted associations, using instrumental variable analysis, of birth weight with clinically measured cardiovascular risk factors and the metabolic syndrome in older …

Mechanism Of Yeast Prion Portein Aggregation And Strain Formation, Tejas Baba Kalastavadi May 2010

Mechanism Of Yeast Prion Portein Aggregation And Strain Formation, Tejas Baba Kalastavadi

All Theses and Dissertations (ETDs)

Misfolding and aggregation of the prion protein: PrP) causes fatal neurodegenerative diseases in many mammalian species, including humans. Mutations in the gene encoding PrP are associated with ~15% of the incidences, while, the vast majority of the cases are sporadic. Interestingly, prion diseases also display pathological variation, suggesting that there are multiple different strains. To elucidate the mechanism of prion protein aggregation and strain formation, I have taken advantage of the yeast prions [PSI+] and [RNQ+] and their protein determinants Sup35p and Rnq1p, respectively. Using a Sup35-PrP chimera, I have investigated the effect of the disease associated oligopeptide repeat domain: …

Explanatory Models In Behavioral Endocrinology, Sylvia Rolloff May 2010

Explanatory Models In Behavioral Endocrinology, Sylvia Rolloff

All Theses and Dissertations (ETDs)

While the progress of science has been described by some as proceeding by a series of "crucial experiments," I claim that an important factor in the development of many scientific fields is that of what I call "crucial resolutions" of outstanding anomalies. In contrast to crucial experiments, crucial resolutions are proposed to solve persistent anomalies or conflicting theoretical positions - often in advance of experimental confirmation. I illustrate this point using specific examples from the history of endocrinology. I claim this general structure of explanatory models can serve as a template for philosophical and historical investigations in other scientific fields.

The Role Of Actin Depolymerizing Factor In Regulating Actin Dynamics In Toxoplasma Gondii, Simren Mehta May 2010

The Role Of Actin Depolymerizing Factor In Regulating Actin Dynamics In Toxoplasma Gondii, Simren Mehta

All Theses and Dissertations (ETDs)

Apicomplexan parasites utilize a unique process of rapid motility termed gliding, which is coupled to their invasion of host cells. Gliding and invasion are dependent on parasite actin filaments, yet parasite actin is mostly non-filamentous. Filaments have been detected only transiently during gliding, suggesting that parasite actin filaments are rapidly assembled and disassembled during gliding motility. Little is known about what regulates the turnover of parasite actin filaments. In higher eukaryotes the Actin Depolymerizing Factor: ADF)/Cofilin proteins are essential regulators of actin filament turnover. ADF is one of the few actin binding proteins conserved in apicomplexan parasites. To investigate the …

Writing American Soldiers: Nineteenth-Century Varieties Of Military Experience, Benjamin Cooper May 2010

Writing American Soldiers: Nineteenth-Century Varieties Of Military Experience, Benjamin Cooper

All Theses and Dissertations (ETDs)

In the late 1980s when Paul Fussell was defending the use of the atomic bomb in World War II, he did so on the grounds that he was a soldier. He knew better because his experiences in the war told him the bomb was necessary. Reactions to Fussell were quick to reject his sense of a personal knowledge of war, and such repudiations were nothing new. When Daniel Aaron claimed the Civil War was left unwritten, he had to overlook scores of novels, short stories, and personal essays written by soldiers who were deliberative artists in their own right, men …

Development And Study Of Inhibitors Of Heat Shock Protein 70 Induction, Rongsheng Wang May 2010

Development And Study Of Inhibitors Of Heat Shock Protein 70 Induction, Rongsheng Wang

All Theses and Dissertations (ETDs)

Tumor and cancer cells that over express heat shock protein 70: HSP70) are found to be multidrug resistant and thermo tolerant, creating a hurdle to existing therapy. Although HSP70 is recognized as an increasingly important drug target, the protein structure of this highly conserved chaperone remains challenging for direct targeting. An alternative strategy is to inhibit the transcription of HSP70. Among known small molecule inhibitors of HSP70 induction, quercetin has very low toxicity and has the advantage of being easily modified for structure-activity studies. One part of the dissertation focuses on the identification of quercetin derivatives with improved specificity and …

Cd4 T-Cell Suppression By Cells From Toxoplasma Gondii-Infected Retinas Is Mediated By Surface Protein Pd-L1, Elizabeth Charles, Sunil Joshi, John D. Ash, Barbara A. Fox May 2010

Cd4 T-Cell Suppression By Cells From Toxoplasma Gondii-Infected Retinas Is Mediated By Surface Protein Pd-L1, Elizabeth Charles, Sunil Joshi, John D. Ash, Barbara A. Fox

Dartmouth Scholarship

In the inflamed retina, CD4(+) T cells can cause retinal damage when they are not properly regulated. Since tissue expression of major histocompatibility complex (MHC) class II and costimulatory molecules is a key mechanism for regulating effector T cells, we tested the hypothesis that upregulation of these proteins in the retina contributes to the regulation of CD4 T cells. Here we report that in retinas infected with the protozoan parasite Toxoplasma gondii, MHC class II is upregulated on infiltrating leukocytes as well as on resident retinal cells, including photoreceptors. Flow cytometric analysis indicated that B7 costimulatory family members (CD80, CD86, …

Landslide Hazard Mapping And Assessment In Himalayas, A. K. Pachauri

International Conferences on Recent Advances in Geotechnical Earthquake Engineering and Soil Dynamics

Himalayas offer a good opportunity to study landslides in hard terrains. The region is a well established seismic region of Asia where earthquakes are common occurrence. The on going plate motion and its collision with Asian plate is the main triggering factor for landslides, but there are geological factors that make the slopes susceptible for landslides to be triggered during rains. The Yatra routes of Char Dham the four pilgrimage centers where millions of Hindus go each year for salvation and heaven abode have to face road blocks each rainy season. The border road organization the pioneer organization looking after …

Comparison Of Erosional Features By Tsunami And Wind Waves, B. K. Pal, M. K. Panda May 2010

Comparison Of Erosional Features By Tsunami And Wind Waves, B. K. Pal, M. K. Panda

International Conferences on Recent Advances in Geotechnical Earthquake Engineering and Soil Dynamics

The erosion features from tsunami wave and wind wave are different according to the characteristics of the two kinds of waves. The tsunami wave is a shallow water wave, even in Deep Ocean, with very long wavelength and relatively high especially near shore. It does not break when attacking the shore. It composed of run-up and run-down. The waves which can scour the offshore sea bottom and deposit the sediment mostly sand on the coast called storm or tsunami over washes. The erosion features from storm wave are caused by the breaking waves and wind-driven currents. However, the erosion features …

Development Of Seismic Safety During Pre- And Co-Seismic Periods, Arun Bapat May 2010

Development Of Seismic Safety During Pre- And Co-Seismic Periods, Arun Bapat

International Conferences on Recent Advances in Geotechnical Earthquake Engineering and Soil Dynamics

Since the year 2000 a large number of earthquakes have taken place. But despite advances in disaster management and seismology, till now it has not been possible to save a single life during earthquake. Issue of forewarning based on reliable seismic precursors has not been considered. Large magnitude earthquakes in India, Indonesia, Pakistan, Iran, China, Italy, and Peru etc were unexpected jolts to the disaster managers. Practically each earthquake has taken heavy death toll in India, Pakistan, Iran, and China etc. If some precursory warning were issued, it would help in saving at least some lives. The issue of earthquake …

Ultrasonic Characterization Of Human Coronary Arteries And Atherosclerotic Plaques, Joseph Hoffman May 2010

Ultrasonic Characterization Of Human Coronary Arteries And Atherosclerotic Plaques, Joseph Hoffman

All Theses and Dissertations (ETDs)

The development, execution, and interpretation of studies investigating the physics of the interaction of ultrasound with normal and pathological coronary artery tissues are described in this dissertation. Ultrasound is a modality capable of visualizing and characterizing the lesions that define atherosclerosis. A better understanding of the physics underlying the mechanisms by which ultrasound interacts with arterial tissue and plaques may provide benefit to patients with coronary artery disease. The two-fold goal of the studies presented in this thesis was to better understand the fundamental physics of the relationship between ultrasound and coronary artery tissue and to use this knowledge to …

Presentation Of Citrullinated Peptides Derived From Unmodified Protein By Antigen Presenting Cells., Jamie Rimer

All Theses and Dissertations (ETDs)

ABSTRACT OF THE DISSERTATION Presentation of citrullinated peptides derived from unmodified protein by antigen presenting cells. By Jamie Michelle Rimer Doctor of Philosophy in Biology and Biomedical Sciences: Immunology) Washington University in St. Louis 2010 Professor Emil R. Unanue, Chair Autoimmune responses to citrullinated proteins have been associated with rheumatoid arthritis; however, little is known of the mechanism by which tolerance to citrullinated proteins is established or the biology of citrullination by cells of the immune system. We find that immunization of B10.BR mice with HEL in CFA gives rise to T cells that specifically recognize citrullinated epitopes. We generated …

Genetic And Phenotypic Differentiation Between Winemaking And Wild Strains Of Saccharomyces Cerevisiae, Katie Hyma May 2010

Genetic And Phenotypic Differentiation Between Winemaking And Wild Strains Of Saccharomyces Cerevisiae, Katie Hyma

All Theses and Dissertations (ETDs)

Traditionally, Saccharomyces cerevisiae has been associated with wine, beer and bread production, yet wild strains have also been isolated from natural habitats. While all strains of S. cerevisiae as well as other Saccharomyces species are capable of wine fermentation, a genetically distinct group of S. cerevisiae strains is primarily used to produce wine. These strains exhibit an apparent genetic bottleneck, which led to the hypothesis that wine stains have been domesticated from `wild' natural strains. However, it is unknown whether the genetic bottleneck was accompanied by selection for phenotypic differences. In this study we tested for phenotypes correlated with the …

Distinct Functions For Type I And Type Ii Interferon In Cancer Immunoediting, Mark Diamond May 2010

Distinct Functions For Type I And Type Ii Interferon In Cancer Immunoediting, Mark Diamond

All Theses and Dissertations (ETDs)

The idea that a functionally intact immune system can protect against cancer development forms the basis of the long-postulated and once controversial concept of cancer immunosurveillance. A substantial body of evidence, however, now exists defining a role for immunity in extrinsic tumor suppression - work that broadened our understanding of the tumor-immune interface and led to the cancer immunoediting hypothesis. The interferons, both type I: IFNα/β) and type II IFN: IFNγ), are critical mediators of cancer immunoediting, yet their respective roles in promoting anti-tumor immune responses remain unclear. Herein, we have examined the actions of IFNα/β and IFNγ during tumor …

Photoacoustic Reporter Gene Imaging And Optical Coherence Computed Tomography, Li Li May 2010

Photoacoustic Reporter Gene Imaging And Optical Coherence Computed Tomography, Li Li

All Theses and Dissertations (ETDs)

Advances in imaging technologies have always been the major driving forces for the evolution of biomedical research. Compared with other modalities, optical imaging possesses several prominent merits. Because light interacts with tissue at the microscopic level through many distinct physical mechanisms, optical methods allow sensitive exploration of various aspects of the life down to the single-molecule level. From the technical perspective, optical systems utilize safe non-ionizing radiation, could be implemented at relatively low cost, also have the potential to be miniaturized for portable or endoscopic applications. As a result, optical imaging tools are playing an increasingly important role in both …

Synthetic Corpora: A Synergy Of Linguistics And Computer Animation, Jerry C. Schnepp, Rosalee Wolfe, John Mcdonald May 2010

Synthetic Corpora: A Synergy Of Linguistics And Computer Animation, Jerry C. Schnepp, Rosalee Wolfe, John Mcdonald

Visual Communications and Technology Education Faculty Publications

Synthetic corpora enable the creation of computer-generated animations depicting sign language and are the complement of corpora containing videotaped exemplars. Any design for a synthetic corpus needs to accommodate linguistic processes as well as support the generation of believable, acceptable synthesized utterances. This paper explores one possibility for representing linguistic and extralinguistic processes that involve the face and reports on the outcomes of a user test evaluating the clarity of utterances synthesized by this approach.

Physics

The SPS at high intensities exhibits transverse singlebunch instabilities with signatures consistent with an Ecloud driven instability.[1] We present recent MD data from the SPS, details of the instrument technique and spectral analysis methods which help reveal complex vertical motion that develops within a subset of the injected bunch trains. The beam motion is detected via wideband exponential taper striplines and delta-sigma hybrids. The raw sum and difference data is sampled at 50 GHz with 1.8 GHz bandwidth. Sliding window FFT techniques and RMS motion techniques show the development of large vertical tune shifts on portions of the bunch of …
