Benzodiazepines (BDZs) enhance GABA(A) receptor inhibition by direct actions on central BDZ receptors (CBRs). Although some BDZs also bind mitochondrial receptors [translocator protein (18 kDa) (TSPO)] and promote the synthesis of GABA-enhancing neurosteroids, the role of neurosteroids in the clinical effects of BDZs is unknown. In rat hippocampal slices, we compared midazolam, an anesthetic BDZ, with clonazepam, an anticonvulsant/anxiolytic BDZ that activates CBRs selectively. Midazolam, but not clonazepam, increased neurosteroid levels in CA1 pyramidal neurons without changing TSPO immunostaining. Midazolam, but not clonazepam, also augmented a form of spike inhibition after stimulation adjacent to the pyramidal cell layer and inhibited …

A robust and efficient algorithm is proposed to quasiunsteadily predict attitudes of floating bodies with arbitrary configurations. Numerical techniques for accuracy and computing time saving are addressed. A floating cylinder was modeled as a benchmark, by theoretical and experimental approaches, to validate the developed program. Attitudes of two kinds of floating bodies, which are a body composed of multiple objects and a submarine-like submersible, were simulated to show the feasibility of the present algorithm. Background Intravenous injection of mice with attenuated Plasmodium berghei sporozoites induces sterile immunity to challenge with viable sporozoites. Non-intravenous routes have been reported to yield poor immunity. Because intravenous immunization has been considered to be unacceptable for large scale vaccination of humans, assessment was made of the results of intradermal immunization of mice with Plasmodium yoelii, a rodent malaria parasite whose infectivity resembles that of human malaria. Methods Mice were immunized with two injections of isolated, radiation-attenuated P. yoelii sporozoites, either by intravenous (IV) or intradermal (ID) inoculation. High fat feeding increases hepatic fat accumulation and is associated with hepatic insulin resistance. AMP Activated Protein Kinase (AMPK) is thought to inhibit lipid synthesis by the acute inhibition of glycerol-3-phosphate acyltransferase (GPAT) activity and transcriptional regulation via SREBP-1c. The purpose of this study was to determine if chronic activation of AMPK prevented an increase in GPAT1 activity in rats fed a high fat diet. Rats were fed a control (C), or a high fat (HF) diet (60% fat) for 6 weeks and injected with saline or a daily AICAR dose of 0.5 mg/g body weight. Chronic AMPK activation by …

A new technique, called nanoinjection, is being developed to insert foreign DNA into a living cell. Such DNA transfection is commonly used to create transgenic organisms vital to the study of genetics, immunology, and many other biological sciences. In nanoinjection, DNA, which has a net negative charge, is electrically attracted to a micromachined lance. The lance then pierces the cell membranes, and the voltage on the lance is reversed, repelling the DNA into the cell. It is shown that DNA motion is strongly correlated to ion transport through a process called electrophoresis. Gel electrophoresis is used to move DNA using …

Natural exposure to hypersaline environments occurs in many fish species (e.g. in tidal pools, lakes, estuaries, or due to migration). Fish in environments that are more concentrated than their extracellular fluids experience continual diffusive water loss which they combat by drinking. Intestinal Na+ and Cl- absorption drives water uptake and concentrates impermeable MgSO4 in the lumen. Epithelial Cl- absorption occurs in part by apical Cl-/HCO3- exchange, with HCO3- provided by transepithelial transport and/or by carbonic anhydrase-mediated hydration of endogenous CO2. Hydration of CO2 also liberates H+, which is excreted across either the basolateral or apical membranes. Transport of water, ions, …

A method for quantifying a neurodegenerative disorder in a patient that includes obtaining a fluid sample from the subject; measuring a protein biomarker complex in said fluid sample and correlating the measurement with mild cognitive impairment or Alzheimer's disease status. The biomarkers include those that comprise at least one of a transthyretin protein and/or a prostaglandin-H2 D-isomerase protein, and at least one second, different protein selected from a transthyretin, prostaglandin-H2 D-isomerase, beta-2-microglobulin, cystatin C, superoxide dismutase [Cu—Zn], plasma retinol-binding protein, phosphatidylethanolamine-binding protein, carbonic anhydrase 2, prostaglandin-H2 D-isomerase, and/or serotransferrin protein.

Ethical research provides great benefit to the public, but ethical research is not guaranteed. Research can go terribly wrong when research subjects are not protected. Egos of scientists and others in power can cause disastrous results, and that is what happened at the Dachau concentration camp in Germany. Because of incidents such as this, medical research at universities is now reviewed by Institutional Review Boards to protect subjects. Purpose: Latino adolescent girls are less physically active than the general adolescent population, which puts them at increased risk for obesity and chronic disease. The Physical Activity Partnership for Girls (PG) uses a community-based participatory research approach to assess community needs and priorities related to physical activity (PA) behaviors of middle school girls living in low-income, urban sectors of San Antonio, Texas. Increased dietary palatability may lead to excess food consumption which in turn causes an excess of caloric intake and weight gain. Although most obesity studies in rodents utilize a high-fat diet, food variation, a combination of different diets for example, may actually increase body weight and fat mass beyond that of solely high-fat feeding. Purpose: To examine the changes in weight gain and body composition in mice that consume either a high-fat diet or a combination of high-fat and stock diet. Purpose: Progenitor cells, produced in the bone marrow, migrate to the thymus where the thymic microenvironment plays a critical role in their differentiation and maturation into T cells. Fat accumulates in the thymus beginning at 1 year of age, causing the thymus to gradually involute as age progresses, thus resulting in a decline in the production of mature T-cells. Obesity augments thymic fat accumulation and hastens the decline in thymic output. Resveratrol, a phytoalexin produced in plants, has also been shown to regulate fat metabolism, in an attempt to reduce hepatic fat accumulation, via 5' adenosine monophosphate-activated protein kinase (AMPK). …

As modern fishery assessments change in an effort to be more accurate and encompass the range of potential ecosystem interactions, critical information on the ecology of species including life history, intra and inter-specific competitive interactions and habitat requirements must be added to the standard fishery-dependent and independent data sets. One species whose movements and habitat associations greatly affects exploitation patterns is lingcod, Ophiodon elongatus, which support an economically important fishery along the coastal waters of the Pacific Coast of North America. BACKGROUND: Age-related neurodegenerative diseases share a number of important pathological features, such as accumulation of misfolded proteins as amyloid oligomers and fibrils. Recent evidence suggests that soluble amyloid oligomers and not the insoluble amyloid fibrils may represent the primary pathological species of protein aggregates.
RESULTS: We have produced several monoclonal antibodies that specifically recognize prefibrillar oligomers and do not recognize amyloid fibrils, monomer or natively folded proteins. Like the polyclonal antisera, the individual monoclonals recognize generic epitopes that do not depend on a specific linear amino acid sequence, but they display distinct preferences for different subsets of prefibrillar oligomers. Increasing concern over the depleting fossil fuels and rising costs of chemical fertilizers have stimulated the need to find alternative crop biomass production through regionally available fertilizer sources. A field experiment was conducted to determine high biomass sorghum [(Sorghum bicolor (Moench) L.) dry matter yield, and N uptake due to application of urea, pelleted poultry litter, and class A municipal biosolids in two cropping systems of biomass sorghum and nitrogen use efficiencies of these three nutrient sources in northeast Arkansas in 2009. Ovarian cancer (OC) is the second most common gynecologic cancer; however it is responsible for the most gynecologic cancer-related deaths. Apoptosis evasion is an important mechanism in OC tumorigenesis, and the prototypic anti-apoptotic protein, B-cell lymphoma 2 (Bcl-2), is often overexpressed in OC tumors. Gaining a better understanding of the mechanism(s) behind Bcl-2 overexpression and potential extra-anti-apoptotic functions of Bcl-2 could elucidate the importance of elevated Bcl-2 in OC. In the current study, I show through immunohistochemical analysis of normal, benign, and OC tissue sections, that both epithelial and stromal Bcl-2 expression decreases with OC progression. Harvesting feedstock for biofuel production must not degrade soil, water, or air resources. Our objective is to provide an overview of field research being conducted to quantify effects of harvesting corn (Zea mays L.) stover as a bioenergy feedstock. Coordinated fi eld studies are being conducted near Ames, IA; St. Paul and Morris, MN; Mead, NE; University Park, PA; Florence, SC; and Brookings, SD., as part of the USDA-ARS Renewable Energy Assessment Project (REAP).
