In poultry, sperm transferred by natural mating or AI into the distal end of the vagina immediately begin their ascent to the utero-vaginal junction (UVJ) at the anterior end of the vagina. However, due to an intense selection process in the vagina, less than 1% of the sperm transferred actually reach the UVJ. Those sperm that do reach the UVJ enter numerous tubular invaginations of the vagina’s surface epithelium located in the UVJ mucosa, collectively referred to as the sperm-storage tubules (SST). Sperm residing in the SST lumen are capable of surviving up to several weeks while retaining their fertilizing …

Transplantation of male germ line cells into sterilized recipients has been used in mammals for conventional breeding as well as for transgenesis. We have previously adapted this approach for the domestic chicken and we present now an improvement of the germ cell transplantation technique by using an enriched subpopulation of c-Kit-positive spermatogonia as donor cells. Dispersed c-Kit positive testicular cells from 16 to 17 week-old pubertal donors were transplanted by injection directly into the testes of recipient males sterilized by repeated gamma irradiation. We describe the repopulation of the recipient’s testes with c-Kit positive donor testicular cells, which resulted in …

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
Trophectoderm cell lines were established from 8-day in vitro-cultured embryos of cattle derived from fertilization (IVF), somatic cell nuclear transfer (NT), or parthenogenetic activation (P) of in vitro-matured oocytes and from five 8-day-old in vivo (V) embryos. The most abundant cellular proteins of 5 V-, 16 NT-, 12 P-, and 16 IVF-derived cell lines were compared by 2D-gel electrophoresis and mass spectrometry; that is, the unaltered thiourea/urea extract of each cell culture was analyzed. Common protein spots (n = 118) were examined, and 95% were identified with significant scores from protein and gene database searches. Of the proteins …

A Computer Model For Simulating Population Development Of The Indianmeal Moth (Lepidoptera: Pyralidae) In Stored Corn, James E. Throne, Richard T. Arbogast
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
The Indianmeal moth, Plodia interpunctella (Hu¨bner) (Lepidoptera: Pyralidae), is a common pest of stored corn, Zea mays L. We developed a computer model to simulate population development of the Indianmeal moth in stored corn by using previously published data describing immature developmental times and survivorship, and adult longevity and fecundity. The model accurately simulated population development of Indianmeal moths in corn stored during fall and into winter of three separate storage seasons in South Carolina. This is the period when the Indianmeal moth is a pest in stored corn in South Carolina. The model predicted that populations would increase after …
Blackboard Jungle: Delinquency, Desegregation, And The Cultural Politics Of Brown, Anders Walker
Blackboard Jungle: Delinquency, Desegregation, And The Cultural Politics Of Brown, Anders Walker
All Faculty Scholarship
In 1955, Metro-Goldwyn-Mayer released a controversial film about juvenile delinquency entitled Blackboard Jungle. Georgia Governor Ernest Vandiver subsequently used the film as a metaphor for what would happen to southern schools were Brown v. Board of Education enforced, marking the beginnings of a much larger campaign to re-articulate southern resistance to integration in popular terms. Taking the intersection between discourses of delinquency and desegregation at mid-century as a starting point, this article advances three claims. One, the NAACP’s reliance on sociological evidence in Brown was a strategic attempt to align black interests with concerns over child development popular at the …

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
Background: Two genotypically and microbiologically distinct strains of Mycobacterium avium subsp. paratuberculosis (MAP) exist - S and C MAP strains that primarily infect sheep and cattle, respectively. Concentration of iron in the cultivation medium has been suggested as one contributing factor for the observed microbiologic differences. We recently demonstrated that S strains have defective iron storage systems, leading us to propose that these strains might experience iron toxicity when excess iron is provided in the medium. To test this hypothesis, we carried out transcriptional and proteomic profiling of these MAP strains under iron-replete or -deplete conditions.
Results: We first complemented …

Association Analysis Of Prnp Gene Region With Chronic Wasting Disease In Rocky Mountain Elk, Stephen N. White, Terry R. Spraker, James O. Reynolds, Katherine I. O’Rourke
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
Background: Chronic wasting disease (CWD) is a transmissible spongiform encephalopathy (TSE) of cervids including white-tailed (Odocoileus virginianus) and mule deer (Odocoileus hemionus), Rocky Mountain elk (Cervus elaphus nelsoni), and moose (Alces alces). A leucine variant at position 132 (132L) in prion protein of Rocky Mountain elk confers a long incubation time with CWD, but not complete resistance. However, variants in regulatory regions outside the open reading frame of PRNP have been associated with varying degrees of susceptibility to prion disease in other species, and some variants have been observed in similar …

Annual spring-seeded forage crops use less water than cereal grains, including durum (Triticum turgidum L. var. durum), and may be suitable to replace summer fallow. We conducted an experiment from 2002 through 2006 comparing yield, quality, and water and N use of durum and three annual forages, barley (Hordeum vulgare L.), barley interseeded with pea [Pisum sativum L. ssp. arvense (L.) Poir.], and foxtail millet [Setaria italica (L.) Beauv.] in 2-yr rotations. Durum in rotation with summer fallow and alfalfa (Medicago sativa L.) were included. Averaged over 5 yr, alfalfa had higher forage yield …

The past four decades have been witness to the realization of ideas which, when first contemplated, seemed to resemble the prose of science fiction writers. Some seemingly far-fetched notions, such as robot pets, anti-sleeping pills, and ugly unisex jumpsuits, are now readily available. However, there are currently no colonies on the moon, and we are all still waiting to order our personal jetpacks from Amazon.com. Some are likely to remain in the realm of science fiction and will only be realized with the help of movie and television special effects artists. Yet through these entertaining glimpses into a potential future, …

Cytocompatible Cross-Linking Of Electrospun Zein Fibers For The Development Of Water-Stable Tissue Engineering Scaffolds, Quiran Jiang, Narendra Reddy, Yiqi Yang
Department of Agricultural and Biological Systems Engineering: Faculty Publications
This paper reports a new method of cross-linking electrospun zein fibers using citric acid as a non-toxic cross-linker to enhance the water stability and cytocompatibility of zein fibers for tissue engineering and other medical applications. The electrospun structure has many advantages over other types of structures and protein-based biomaterials possess unique properties preferred for tissue engineering and other medical applications. However, ultrafine fiber matrices developed from proteins have poor mechanical properties and morphological stability in the aqueous environments required for medical applications. Efforts have been made to improve the water stability of electrospun protein scaffolds using cross-linking and other approaches, …

Microwave-Based Alkali Pretreatment Of Switchgrass And Coastal Bermudagrass For Bioethanol Production, Deepak R. Keshwani, Jay J. Cheng
Department of Agricultural and Biological Systems Engineering: Faculty Publications
Switchgrass and coastal bermudagrass are promising lignocellulosic feedstocks for bioethanol production. However, pretreatment of lignocelluloses is required to improve production of fermentable sugars from enzymatic hydrolysis. Microwave-based alkali pretreatment of switchgrass and coastal bermudagrass was investigated in this study. Pretreatments were carried out by immersing the biomass in dilute alkali reagents and exposing the slurry to microwave radiation at 250 W for residence times ranging from 5 to 20 minutes. Simons’ stain method was used to quantify changes in biomass porosity as a result of the pretreatment. Pretreatments were evaluated based on yields of total reducing sugars, glucose, and xylose. …
Interleukin 6 And 8 Gene Expression Responses To Resistance Exercise And The Correlation To Muscle Mass, Vivien Massie
Interleukin 6 And 8 Gene Expression Responses To Resistance Exercise And The Correlation To Muscle Mass, Vivien Massie
Theses : Honours
The post exercise inflammatory response is a key signalling mechanism regulating muscle protein synthesis. The purpose of this research was firstly to determine whether muscle mass in non-strength trained individuals was associated with the inflammatory muscle gene response after a single bout of eccentric muscle loading. Secondly, to determine whether changes in muscle cross-sectional area after a chronic increase in muscle loading (resistance training) is related to the inflammatory gene response to a single bout of muscle loading. Eleven male participants (21.6 ± 4.1 years) volunteered for this study. Each participant completed a preliminary testing session that consisted of two …
Do Different Levels Of Fishing Pressure, Through Spatial Management, Influence Communities And Ecological Interactions In Seagrass Meadows In South Western Australia?, Karina Inostroza
Theses : Honours
Marine Protected Areas (MPAs) are internationally recognised as a significant spatial management and cost-effective strategy to restore and conserve the marine ecosystems structure and function from human impact. MPAs have been declared with the aim to address a single or a set of management objectives in response to anthropogenic threats, such as fishing, which can have repercussions on biodiversity through indirect interactions or top-down control within an ecosystem or across ecosystems where consumers migrate to forage. However, this indirect impact of fishing remains poorly understood, and as a consequence, the effectiveness of MP As in conserving those ecological processes and …
Where New Meanings Spring: The Relationship Between Indigenous Cultural Meanings For Freshwater Springs And Management Practices: Analysis Of Stories From Kalbarri, Western Australia, Tamara Lee Murdock
Theses : Honours
While Indigenous peoples' practices have been acknowledged to change and evolve, whether Indigenous cultural meanings invested in a specific place also change and/or evolve over time, and the affect these changes may have on land and water practices has generally been ignored. This study explores the relationship between Indigenous cultural meanings and land and water stewardship practices, and whether these change over time. A qualitative research design was employed in this study to emphasise the complex and dynamic nature of language and the relationship between people, culture and nature. This study utilised interviews collected from traditional Indigenous people concerning stories …
The Seventeenth-Century Battaglie For Lute In Italy, Aiden Deasy
The Seventeenth-Century Battaglie For Lute In Italy, Aiden Deasy
Theses : Honours
Italy had been the theatre of war for centuries. Political fragmentation ignited dynastic rivalries, carving out powerful autonomous city-states whose leaders amassed within their courts artists, musicians and writers in order to chronicle their military, religious and political vision. The musical depictions of particular military exploits, or battaglia, became popular springboards for colourful, programmatic compositions. These works were often performed in public in order to garner support for campaigns. Originally a vocal genre whose distant relative can be traced back to the caccia, or hunting song, the battaglie were often transcribed for instruments, in particular the lute, thus …

The Critical Aesthetics Of Disorder: A Porteña Crisis Of Size, Melissa Maldonado-Salcedo
Publications and Research
In Argentina, contemporary notions of progress originate within the whitening projects of the nineteenth century. In this work I propose that the imprints of these Colonial modernization projects are still visible on the porteña body. For this reason I examine the Porteño fashion industry alongside the pervasive Barbie culture. I believe that Porteñas embody the national crisis of identity. I seek to pose the possibility — instead of arguing the fact — that the fashion industry's power over Porteñas (women who live in Buenos Aires Capital) toughened post the economic crisis of 2001. The reason for this is rooted in …

Geography
Understanding the relationships between climate and carbon exchange by terrestrial ecosystems is critical to predict future levels of atmospheric carbon dioxide because of the potential accelerating effects of positive climate-carbon cycle feedbacks. However, directly observed relationships between climate and terrestrial CO2exchange with the atmosphere across biomes and continents are lacking. Here we present data describing the relationships between net ecosystem exchange of carbon (NEE) and climate factors as measured using the eddy covariance method at 125 unique sites in various ecosystems over six continents with a total of 559 site-years. We find that NEE observed at eddy covariance sites is …

Creating Units For Second Grade Learners Of English As A Foreign Language, Laura Elizabeth Messerly
Creating Units For Second Grade Learners Of English As A Foreign Language, Laura Elizabeth Messerly
Honors Program Theses
Teaching English as a Foreign Language is becoming more popular as English is in high demand in countries throughout the world because of globalization (Niklov, 2006). The chief difference between English as a Foreign Language (EFL) and English as a Second Language (ESL) is that the language is being learned in a non-English speaking community (.EFL) versus an English speaking community (ESL). These non-English speaking communities are being affected by multinational corporations. As businesses grow and globalize, flourishing across continents, factories and branches are sprouting up in places all over the world. Technology, too, has contributed to the spread of …

N-P Co-Limitation Of Primary Production And Response Of Arthropods To N And P In Early Primary Succession On Mount St. Helens Volcano, John G. Bishop, Niamh B. O'Hara, Jonathan H. Titus, Jennifer L. Apple, Richard A. Gill, Louise Wynn
Biology
Background: The effect of low nutrient availability on plant-consumer interactions during early succession is poorly understood. The low productivity and complexity of primary successional communities are expected to limit diversity and abundance of arthropods, but few studies have examined arthropod responses to enhanced nutrient supply in this context. We investigated the effects of nitrogen (N) and phosphorus (P) addition on plant productivity and arthropod abundance on 24-yr-old soils at Mount St. Helens volcano. Methodology/Principal Findings:We measured the relative abundance of eight arthropod orders and five families in plots that received N, P, or no nutrients for 3-5 years. We also …

Comparative In Vivo Study Of Gp96 Adjuvanticity In The Frog Xenopus Laevis, Hristina Nedelkovska, Tanya Cruz-Luna, Pamela Mcpherson, Jacques Robert
Biology
We have developed in the amphibian Xenopus laevis a unique non-mammalian model to study the ability of certain heat shock proteins (hsps) such as gp96 to facilitate cross-presentation of chaperoned antigens and elicit innate and adaptive T cell responses. Xenopus skin graft rejection provides an excellent platform to study the ability of gp96 to elicit classical MHC class Ia (class Ia) restricted T cell responses. Additionally, the Xenopus model system also provides an attractive alternative to mice for exploring the ability of gp96 to generate responses against tumors that have down-regulated their class Ia molecules thereby escaping immune surveillance. Recently, …
