Physiology And Behavior Of The Jumbo Squid Dosidicus Gigas, Lloyd A. Trueblood Jan 2010

Physiology And Behavior Of The Jumbo Squid Dosidicus Gigas, Lloyd A. Trueblood

Open Access Dissertations

Dosidicus gigas is a large, epipelagic squid known to vertically migrate, spending night on the surface, and descending during the day into the oxygen minimum layer (OML). This dissertation seeks to examine physiological and behavioral adaptations of D. gigas to the environment in which it dwells. Life in the open ocean is unique in that there is little physical refuge. In order to capture prey in this environment, many pelagic fishes engage at high relative swimming speeds attempting to strike before being detected. This requires high metabolic rates and increasing anaerobic potential with increasing body size to overcome drag. I …

Roles Of Src Family Kinase Signaling During Fertilization And The First Cell Cycle In The Marine Protostome Worm Cerebratulus, Stephen A. Stricker, David J. Carroll, Wai L. Tsui Jan 2010

Roles Of Src Family Kinase Signaling During Fertilization And The First Cell Cycle In The Marine Protostome Worm Cerebratulus, Stephen A. Stricker, David J. Carroll, Wai L. Tsui

Ocean Engineering and Marine Sciences Faculty Publications

For eggs to generate a calcium response during fertilization, the sperm of many deuterostome animals must first activate a group of egg kinases, called Src family kinases (SFKs). However, whether SFK activation is also required for fertilization-induced calcium signals in eggs of protostomes remains unknown. Thus, in this study, unfertilized oocytes of the marine protostome worm Cerebratulus were treated with either PP2 to inhibit SFKs or with U73122 to block phospholipase C activity downstream of SFK. Compared with control fertilizations, the inhibitors significantly reduced post-insemination levels of polar body formation and cleavage, but apparently did so via different mechanisms, based …

Early Development Of Graphical Literacy Through Knowledge Building, Jianwei Zhang, Yongcheng Gan, Marlene Scardamalia, Huang-Yao Hong

Educational Theory and Practice Faculty Scholarship

This study examined growth in graphical literacy for students contributing to an online, multimedia, communal environment as they advanced their understanding of biology, history and optics. Their science and history studies started early in Grade 3 and continued to the end of Grade 4; students did not receive instruction in graphics production, nor were they required to produce graphics. Results show that students spontaneously produced graphics that advanced along seven dimensions, including effective representation of complex ideas, use of source information and captions, and aesthetic quality. On average, the scores for the seven dimensions were higher for Grade 4 students …

Characterization And Optimization Of Microelectrode Arrays For Glutamate Measurements In The Rat Hippocampus, Pooja Mahendra Talauliker Jan 2010

Characterization And Optimization Of Microelectrode Arrays For Glutamate Measurements In The Rat Hippocampus, Pooja Mahendra Talauliker

University of Kentucky Doctoral Dissertations

An overarching goal of the Gerhardt laboratory is the development of an implantable neural device that allows for long-term glutamate recordings in the hippocampus. Proper L-glutamate regulation is essential for hippocampal function, while glutamate dysregulation is implicated in many neurodegenerative diseases. Direct evidence for subregional glutamate regulation is lacking in previous in vivo studies because of limitations in the spatio-temporal resolution of conventional experimental techniques. We used novel enzyme-coated microelectrode arrays (MEAs) for rapid measurements (2Hz) of extracellular glutamate in urethane-anesthetized rats. Potassium-evoked glutamate release was highest in the cornu ammonis 1 (CA1) subregion and lowest in the cornu ammonis …

Assessing Impact Of Affect Recognition On Therapeutic Relationship, Julianne V. Sutter Jan 2010

Assessing Impact Of Affect Recognition On Therapeutic Relationship, Julianne V. Sutter

University of Kentucky Master's Theses

Therapeutic alliance and its relationship to client nonverbal behavior, specifically facial expressions, were examined. Therapist interpretation of the client nonverbal behavior, or affect, influences the therapeutic alliance and process. Based on a sample of clients from a graduate school therapy training facility, results suggest therapist training in facial expressions, and how they relate to client emotion, improve the therapeutic alliance between therapist and client. After a micro-expression training for therapists, clients reported higher life functioning on the Outcome Rating Scale (ORS) and an improved therapeutic alliance on the Session Rating Scale (SRS). Overall, these findings support the benefit of incorporating …

The Characterization And Calculation Of Diet Quality For A Low-Income Population In Quito, Ecuador, Rachel Ann Beyatte Jan 2010

The Characterization And Calculation Of Diet Quality For A Low-Income Population In Quito, Ecuador, Rachel Ann Beyatte

University of Kentucky Master's Theses

Diet is an important factor in disease prevention and health promotion. According to Global Health, in 2006 four of the top five leading causes of death in Ecuador were heart disease, cerebrovascular diseases, diabetes mellitus and hypertensive diseases which are all associated with dietary intake. Knowledge of eating patterns enables the formulation of public policies directed toward health promotion and disease prevention. Studies using indices that measure dietary quality and summarize the main characteristics of healthful eating habits enables the evaluation of possible associations between health determinants and health indicators. Most studies using score-based approaches have generally shown more varied …

Multiwall Carbon Nanotubes Alter The Thermal Profile And Antibiotic Elution Of Orthopaedic Bone Cement, Alison Carroll Tickle Jan 2010

Multiwall Carbon Nanotubes Alter The Thermal Profile And Antibiotic Elution Of Orthopaedic Bone Cement, Alison Carroll Tickle

University of Kentucky Master's Theses

Multiwall carbon nanotubes (MWNTs) have extraordinary mechanical and thermal transport properties. They significantly improve the static and dynamic mechanical properties of acrylic orthopaedic bone cement when added to the dry cement polymer powder. Understanding the role MWNTs play on bone cement polymerization temperatures will lead to improved mechanical integrity of the cement-bone interface in joint arthroplasties. It was determined through thermal testing that MWNTs increased the polymerization time of the methylmethacrylate by 45-460% and decreased the peak exothermic temperature of bone cement with and without antibiotics. The flow of heat produced during polymerizing cement was reduced 25-85% with the addition …

Resistant Starches Types 2 And 4 Have Differential Effects On The Composition Of The Fecal Microbiota In Human Subjects, Ines Martinez, Jaehyoung Kim, Patrick R. Duffy, Vicki L. Schlegel, Jens Walter

Department of Food Science and Technology: Faculty Publications

Background: To systematically develop dietary strategies based on resistant starch (RS) that modulate the human gut microbiome, detailed in vivo studies that evaluate the effects of different forms of RS on the community structure and population dynamics of the gut microbiota are necessary. The aim of the present study was to gain a community wide perspective of the effects of RS types 2 (RS2) and 4 (RS4) on the fecal microbiota in human individuals.

Methods and Findings: Ten human subjects consumed crackers for three weeks each containing either RS2, RS4, or native starch in a double-blind, crossover design. Multiplex sequencing …

Cultivating Farmers' Rights: Reconciling Food Security, Indigenous Agriculture, And Trips, Lauren Winter Jan 2010

Cultivating Farmers' Rights: Reconciling Food Security, Indigenous Agriculture, And Trips, Lauren Winter

Vanderbilt Journal of Transnational Law

This Note discusses strategies for cultivating Farmers' Rights internationally. The rise of international treaties awarding intellectual property rights in plant genetic resources to plant breeders brought with it an erosion of agricultural biodiversity as well indigenous farmer lifestyles. Farmers' Rights emerged in recognition of the role of traditional farmers play in conserving, creating, and promoting genetic diversity in the food supply and of the importance of maintaining traditional agriculture practices. This Note argues that Farmers' Rights can be realized internationally through concerted effort. The Note proposes that Farmers' Rights could be realized if national governments create laws and infrastructure that …

Legal Techniques For Dealing With Scientific Uncertainty In Environmental Law, Jorge E. Vinuales Jan 2010

Legal Techniques For Dealing With Scientific Uncertainty In Environmental Law, Jorge E. Vinuales

Vanderbilt Journal of Transnational Law

This Article analyzes how scientific uncertainty is handled in international environmental law. It identifies ten legal techniques used for this purpose (i.e., precautionary reasoning; framework-protocol approach; advisory scientific bodies; law-making by treaty bodies; managerial approaches to compliance; prior informed consent; environmental impact assessment and monitoring; provisional measures; evidence; and facilitated liability) and links them to four different stages of development of environmental regimes (i.e., advocacy, design, implementation, and reparation). These techniques are illustrated by reference to some fifteen environmental treaties and other instruments as well as through a detailed case study focusing on the climate change regime.

Nineteenth-Century Principles For Twenty-First-Century Pleading, Adam Mcdonell Moline Jan 2010

Nineteenth-Century Principles For Twenty-First-Century Pleading, Adam Mcdonell Moline

Emory Law Journal

Two recent Supreme Court decisions, Bell Atlantic Corp. v. Twombly and Ashcroft v. Iqbal, upended the standards of pleading under the Federal Rules. In both cases, the plaintiffs had filed complaints so unsubstantiated that the Court concluded they could have no other purpose than to abuse the discovery process against the defendants. Rather than subject the defendants to these unfair burdens, the Court struck language from a fifty-year-old precedent, Conley v. Gibson, that would have allowed the suits to proceed. The Court¿s solution created three new problems. Lower court judges found little guidance in its new, amorphous ¿plausibility¿ standard. Critics …
