The rising temperature of the world's oceans has become a major threat to coral reefs globally as the severity and frequency of mass coral bleaching and mortality events increase. In 2005, high ocean temperatures in the tropical Atlantic and Caribbean resulted in the most severe bleaching event ever recorded in the basin. Methodology/Principal Findings: Satellite-based tools provided warnings for coral reef managers and scientists, guiding both the iming and location of researchers' field observations as anomalously warm conditions developed and spread across the greater Caribbean region from June to October 2005. Tumor necrosis factor alpha (TNF-α) is a critical mediator of inflammation, and its production is tightly regulated, with control points operating at nearly every step of its biosynthesis. We sought to identify uncharacterized TNF-α 3' untranslated region (3'UTR)-interacting proteins utilizing a novel screen, termed the RNA capture assay. We identified CARHSP1, a cold-shock domain-containing protein. Knockdown of CARHSP1 inhibits TNF-α protein production in lipopolysaccharide (LPS)-stimulated cells and reduces the level of TNF-α mRNA in both resting and LPS-stimulated cells. mRNA stability assays demonstrate that CARHSP1 knockdown decreases TNF-α mRNA stability from a half-life (t(1/2)) of 49 min to a t(1/2) …

Protein phosphorylation is a post translational modification of proteins in which a serine, a threonine or a tyrosine residue is phosphorylated by an enzyme, kinase. Phosphorylation of proteins is a reversible and very important regulatory mechanism that occurs in both prokaryotes and eukaryotes. Phosphorylation turns many protein enzymes on and off, preventing or causing many diseases such as diabetes, cancer and rheumatoid arthritis. The phosphorylation on tyrosine residues of proteins is essential for transmission of signals for cell growth, proliferation and differentiation. Protein tyrosine phosphatases (PTPs) in concert with protein tyrosine kinases (PTKs) regulate many signal transduction pathways by controlling …

Reading Holiness: Agnes Grey, Ælfric, And The Augustinian Hermeneutic, Jessica Caroline Brown

Although Anne Brontë's first novel, Agnes Grey, presents itself as a didactic treatise, Brontë's work departs from many accepted Evangelical tropes in the portrayal of its moral protagonist. These departures create an exemplary figure whose flaws potentially subvert the novel's didactic purposes. The character of Agnes is not necessarily meant to be directly emulated, yet Brontë's governess is presented as a tool of moral instruction. The conflict between the novel's self-proclaimed didactic purpose and the form in which it presents that purpose raises a number of interpretive questions. I argue that many of these questions can be answered through …

Vertebrates display a wide variety of parental care behaviours, including the guarding of offspring pre and post nutritional independence as well as the direct provision of nutrients during the early development period. The Amazonian cichlid Symphysodon spp. (discus fish) is unusual among fish species, in that both parents provide offspring with mucus secretions to feed from after hatching. This extensive provision of care, which can last up to a month, imposes a physiological demand on both parents and gives rise to conflict between the parent and offspring. The transition from secondary education to post-secondary education is stressful for all students. The Individuals with Disabilities Education Improvement Act (IDEIA 2004) only mandates transition plans for students receiving special education services. Career assessments and transition services should encourage career exploration which requires students to have knowledge about oneself and the world of work. When career assessments provide students with this information, they may be more informed on what major to choose. Objectives: The objective of this study was to evaluate the influence of acute pulmonary tuberculosis and the effect of drug therapy on markers of oxidative stress (malondialdehyde [MDA] and total antioxidant status [TAS]), C-reactive protein (CRP) and iron body status indices. Methods:Forty patients with active pulmonary tuberculosis from the Advisory Clinic for Chest and Respiratory Diseases in Mosul City, Iraq,were included in this study, with fifty healthy age and sex matched subjects as controls. Assessment of serum concentrations of MDA, TAS, CRP, serum iron, total iron binding capacity, transferring saturation percent and ferritin were done for both patients and controls. …

Objectives: The Nutritional Advice Protocol, established by the Department of Health to combat diabetes, has been implemented in primary health care throughout Oman since 2003. This study aimed to assess the effectiveness of dietary and lifestyle advice and determine the perception and attitudes of Omani adults with type 2 diabetes to diabetes management. Methods: A cross-sectional epidemiological survey was conducted on 98 patients diagnosed with type 2 diabetes in Al-Buraimi Governorate, Oman. Infection with the hepatitis C virus (HCV) is a worldwide problem. Patients with chronic HCV infection who are non-responders to standard therapy represent a growing population within the HCV epidemic.Novel, more efficient and tolerable therapies are urgently needed. This review discusses the recent results showing that targeting miR-122, a micro-ribonucleic acid (MicroRNA) that enhances HCV replication, is a new anti-HCV therapy with a high barrier to resistance.

Given the serious social problems confronting Americans and others world-wide, the authors propose that Dewey's 1932 challenge to teachers is worthy of reconsideration by educators at all levels. In times similar to our own, Dewey challenged teachers to cultivate students' capacities to identify their happiness with what they can do to improve the conditions of others. At first glance, his challenge seems utopian. In order to justify their proposal for re-consideration, the authors explicate Dewey’s challenge by discussing two aspects of the thinking that lies behind it. Background: The subclass Enoplia (Phylum Nematoda) is purported to be the earliest branching clade amongst all nematode taxa, yet the deep phylogeny of this important lineage remains elusive. Free-living marine species within the order Enoplida play prominent roles in marine ecosystems, but previous Molecular phylogenies have provided only the briefest evolutionary insights; this study aimed to firmly resolve internal relationships within the hyper diverse but poorly understood Enoplida. In addition, we revisited the Molecular framework of the Nematoda using a rigorous phylogenetic approach in order to investigate patterns of early splits amongst the oldest lineages (Dorylaimia and Enoplia). In Vitro Gene Regulatory Networks Predict In Vivo Function Of Liver, Youping Deng, David R. Johnson, Junmei Ai, Edward J. Perkins, Sharon A. Meyer

Background: Evolution of toxicity testing is predicated upon using in vitro cell based systems to rapidly screen and predict how a chemical might cause toxicity to an organ in vivo. However, the degree to which we can extend in vitro results to in vivo activity and possible mechanisms of action remains to be fully addressed.

Results: Here we use the nitroaromatic 2,4,6-trinitrotoluene (TNT) as a model chemical to compare and determine how we might extrapolate from in vitro data to in vivo effects. We found 341 transcripts differentially expressed in common among in vitro and in vivo assays …
