Semiparametric Two-Part Models With Proportionality Constraints: Analysis Of The Multi-Ethnic Study Of Atherosclerosis (Mesa), Anna Liu, Richard Kronmal, Xiao-Hua Zhou, Shuangge Ma

SUMMARY. In this article, we analyze the coronary artery calcium (CAC) score in the Multi-Ethnic Study of Atherosclerosis (MESA), where about half of the CAC scores are zero and the rest are continuously distributed. When the observed data has a mixture distribution, two-part models can be the natural choice. With a two-part model, there are two covariate effects, with one in each part of the model. Determination of whether the two covariate effects are proportional can provide more insights into the process underlying development and progression of CAC. Pooled Nucleic Acid Testing To Identify Antiretroviral Treatment Failure During Hiv Infection, Susanne May, Anthony Gamst, Richard Haubrich, Constance Benson, Davey Smith

Abstract Background: Pooling strategies have been used to reduce the costs of polymerase chain reaction based screening for acute HIV infection in populations where the prevalence of acute infection is low (<1%). Only limited research has been done for conditions where the prevalence of screening positivity is higher (>1%). Methods and Results: We present data on a variety of pooling strategies that incorporate the use of PCR-based quantitative measures to monitor for virologic failure among HIV-infected patients receiving antiretroviral therapy. For a prevalence of virologic failure between 1% and 25%, we demonstrate relative efficiency and accuracy of various strategies. CONTEXT: The established relationship between lymph node metastasis and prognosis in colorectal cancer suggests that recurrence in 25% of patients with lymph nodes free of tumor cells by histopathology (pN0) reflects the presence of occult metastases. Guanylyl cyclase 2C (GUCY2C) is a marker expressed by colorectal tumors that could reveal occult metastases in lymph nodes and better estimate recurrence risk.

OBJECTIVE: To examine the association of occult lymph node metastases detected by quantifying GUCY2C messenger RNA, using the reverse transcriptase-polymerase chain reaction, with recurrence and survival in patients with colorectal cancer.

Developing nontraditional approaches to the synthesis and characterization of multivalent compounds is critical to our efforts to study and interface with biological systems and to build new noncovalent materials. This paper demonstrates a biomimetic approach to the construction of discrete, modular, multivalent receptors via molecular self-assembly in aqueous solution. Scaffolds presenting 1−3 viologen groups recruit a respective 1−3 copies of the synthetic host, cucurbit[8]uril, in a noncooperative manner and with a consistent equilibrium association constant (Ka) value of 2 × 106 M−1 per binding site. Adiposity, Stress, And Stigmatization: A Biopsychosocial Approach To Cardiovascular Disease Prediction, Kristi E. White

The purpose of the present study was to use a biopsychosocial approach to investigate the role of body image concerns and appearance-related evaluation stress in the relationship between adiposity and cardiovascular reactivity and recovery. Participants included 106 Caucasian female undergraduates at the University of South Florida. The laboratory procedure consisted of resting baseline, speech preparation and delivery, and recovery phases. Participants also completed a variety of body image questionnaires. To manipulate appearance-related evaluation stress, participants were randomly assigned to present their speech to a video camera or an audio recorder. Regulation Of Distinct Septin Rings In A Single Cell By Elm1p And Gin4p Kinases, Bradley S. Demay, Rebecca A. Meseroll, Patricia Occhipinti, Amy S. Gladfelter

Septins are conserved, GTP-binding proteins that assemble into higher order structures, including filaments and rings with varied cellular functions. Using four-dimensional quantitative fluorescence microscopy of Ashbya gossypii fungal cells, we show that septins can assemble into morphologically distinct classes of rings that vary in dimensions, intensities, and positions within a single cell. Notably, these different classes coexist and persist for extended times, similar in appearance and behavior to septins in mammalian neurons and cultured cells. We demonstrate that new septin proteins can add through time to assembled rings, indicating that septins may continue to polymerize during ring maturation. Different classes …

The Mrna Nuclear Export Machinery Is Targeted By Influenza Virus And Antivirals, Neal G. Satterly

Proper mRNA nuclear export is essential for harmonious growth and maintenance of a cell. An effective weapon influenza virus employs to hijack a host cell is its ability to inhibit such export. Exactly how influenza virus achieves this inhibition is not fully known. Here, we demonstrate that upon infection, influenza virus degrades two nucleopore proteins (Nup98 and Nup96), which play a key role in mRNA nuclear export. Also, a main virulence factor of influenza virus (non-structural protein 1, NS1) binds directly to NXF1 and E1B-AP5, two key constituents of the mRNA export pathway (NXF1/NXT pathway) responsible for exporting bulk (~70%) …

The African Plio-Pleistocene hominins known as australopiths evolved a distinctive craniofacial morphology that traditionally has been viewed as a dietary adaptation for feeding on either small, hard objects or on large volumes of food. A historically influential interpretation of this morphology hypothesizes that loads applied to the premolars during feeding had a profound influence on the evolution of australopith craniofacial form. Here, we test this hypothesis using finite element analysis in conjunction with comparative, imaging, and experimental methods.
