Previous studies of ag-related NH3 concentrations and/or emissions have used a variety of techniques, with very few studies offering adequate instrumental comparisons. In the fall of 2007, NH3 concentrations/emissions were monitored for a one week period at a waste treatment lagoon on a 6000-cow dairy in Idaho using five separate methodologies. Up to twenty-five Ogawa passive samplers were dispersed around the perimeter of the lagoon, with a concentrated bank of samplers arrayed along the predicted downwind side of the facility. A URG acid/base gas denuder system, assembles with three series denuders configured for NH3 collection, was collocated at a single …

Transcriptional, Epigenetic, And Signal Events In Antifolate Therapeutics, Alexandra Racanelli Jun 2009

Transcriptional, Epigenetic, And Signal Events In Antifolate Therapeutics, Alexandra Racanelli

Theses and Dissertations

A targeted approach to the development of antifolate therapies has been sought for many years. Central to the success of such development is an understanding of the molecular mechanisms dictating the sensitivity of cells to antifolates and the fundamental differences of these processes between normal and neoplastic phenotypes. This dissertation addressed transcriptional mechanisms and cell-signaling events responsible for the efficacy of antifolate therapies. Transcriptional processes and cell signaling pathways are often aberrant in neoplastic tissues, providing a potential point of distinction between a normal and neoplastic cellular state. Folylpolyglutamate synthetase (FPGS) catalyzes the formation of poly-γ-glutamate derivatives of folates and …

Stand Density In South Florida Tropical Forests: Implications For The Function And Management Of Everglades Tree Islands, M. Ross, P. L. Ruiz, J. P. Sah, L. Lopez, N. Colbert Jun 2009

Stand Density In South Florida Tropical Forests: Implications For The Function And Management Of Everglades Tree Islands, M. Ross, P. L. Ruiz, J. P. Sah, L. Lopez, N. Colbert

North American Forest Ecology Workshop

Within the continental US, the broadleaved forests of south Florida are exceptional in the abundance and diversity of tree species of tropical origin. Dry tropical forests are regionally most extensive in the upper Florida Keys, but are also represented on the mainland as fragments on limestone rocklands, and as “tree islands” embedded in the Everglades marsh. The exposed Everglades tree islands have a history of human use reaching back thousands of years, and are subject to frequent disturbance from tropical storms and hurricanes. They are sensitive to the hydrology of the surrounding marsh, which can lead to gradual changes in …

Aspen Ecology In The United States: Recent Advances And Future Needs, Paul C. Rogers Jun 2009

Aspen Ecology In The United States: Recent Advances And Future Needs, Paul C. Rogers

North American Forest Ecology Workshop

This presentation (along with a similar Canadian perspective) will offer a context for subsequent papers in the Advances in North American Aspen Ecology session. Collectively, we hope to outline major areas of research in North America, especially as they relate to perceived management needs. There is a long and rich history of ecological research in aspen systems of the United States. Probably the most striking long-term change, from a managerial perspective, has been a shift toward biological benefits of aspen as opposed to past avoidance, or even active denudation, of these systems in favor of more commercial species. This talk …

Intestinal Hco3- Secretion In Fish: A Widespread Mechanism With Newly Recognized Physiological Functions, Josi R. Taylor Jun 2009

Intestinal Hco3- Secretion In Fish: A Widespread Mechanism With Newly Recognized Physiological Functions, Josi R. Taylor

Open Access Dissertations

Intestinal HCO3- secretion and the excretion of resultant CaCO3 precipitates have become a recognized characteristic of seawater osmoregulation in teleosts; however, this is the first report of this osmoregulatory strategy outside of teleosts and also includes evidence for its use in green turtles, Chelonia mydas. Furthermore, the effects of feeding on intestinal HCO3- secretion were newly investigated in teleosts. Intestinal base secretion via apical Cl-/HCO3- exchange was found to increase following feeding, at a magnitude sufficient to offset the "alkaline tide" commonly associated with digestion. Intestinal HCO3- secretion in marine teleosts draws HCO3- from both endogenous (via hydration of intracellular …

Hexabromocyclododecane Decreases The Lytic Function And Atp Levels Of Human Natural Killer Cells, Natasha C. Hinkson, Margaret M. Whalen Jun 2009

Hexabromocyclododecane Decreases The Lytic Function And Atp Levels Of Human Natural Killer Cells, Natasha C. Hinkson, Margaret M. Whalen

Chemistry Faculty Research

This study investigates the effect of hexabromocyclododecane (HBCD) on the lytic function of human natural killer (NK) cells and on ATP levels in NK cells. NK cells are capable of lysing tumor cells, virally infected cells, and antibody-coated cells. HBCD is a brominated cyclic alkane used primarily as an additive flame retardant. If HBCD interferes with NK cell function, this could increase risk of tumor development and/or viral infection. NK cells were exposed to various concentrations of HBCD for 24 and 48 h and 6 days before determining lytic function and ATP levels. ATP levels and lytic function were also …

Findsite: A Combined Evolution/Structure-Based Approach To Protein Function Prediction, Jeffrey Skolnick, Michal Brylinski Jun 2009

Findsite: A Combined Evolution/Structure-Based Approach To Protein Function Prediction, Jeffrey Skolnick, Michal Brylinski

Faculty Publications

A key challenge of the post-genomic era is the identification of the function(s) of all the molecules in a given organism. Here, we review the status of sequence and structure-based approaches to protein function inference and ligand screening that can provide functional insights for a significant fraction of the ∼50% of ORFs of unassigned function in an average proteome. We then describe FINDSITE, a recently developed algorithm for ligand binding site prediction, ligand screening and molecular function prediction, which is based on binding site conservation across evolutionary distant proteins identified by threading. Importantly, FINDSITE gives comparable results when high-resolution experimental …

T10b9 Monoclonal Antibody: A Short-Acting Nonstimulating Monoclonal Antibody That Spares Γδ T-Cells And Treats And Prevents Cellular Rejection, Thomas H. Waid, John S. Thompson, Maria Siemionow, Stephen A. Brown

Internal Medicine Faculty Publications

T10B9.1A-31/MEDI-500 is a nonmitogenic immunoglobulin M kappa murine monoclonal antibody (mAb) directed against the alpha-beta (αβ) heterodimer of the T-lymphocyte receptor complex. The hybridoma was first produced by fusing spleen cells from BALB/C mice immunized with human peripheral blood T-lymphocytes with SP2/O-Ag14 mutant myeloma cells. The mAb is produced and purified using multistep ion exchange and molecular sieve chromatography protocols. T10B9 has been used successfully to treat acute cellular rejection in renal transplantation and as an immunosuppression induction agent in heart and simultaneous kidney-pancreas transplantation. Because T10B9 is nonmitogenic and causes minimal cytokine release, both treatment of rejection and induction …

A Real-Time Prostate Cancer Detection Technique Using Needle Insertion Force And Patient-Specific Criteria During Percutaneous Intervention, Kaiguo Yan, T. Podder, L. Li, J. Joseph, D. R. Rubens, E. M. Messing, L. Liao, Y. Yu

Department of Radiation Oncology Faculty Papers

In this article, the authors present a novel real-time cancer detection technique by using needle insertion forces in conjunction with patient-specific criteria during percutaneous interventions. Needle insertion experiments and pathological analysis were performed for developing a computer-aided detection (CAD) model. Backward stepwise regression method was performed to identify the statistically significant patient-specific factors. A baseline force model was then developed using these significant factors. The threshold force model that estimated the lower bound of the cancerous tissue forces was formulated by adding an adjustable classifier to the baseline force model. Trade-off between sensitivity and specificity was obtained by varying the …

The Effects Of Spatial And Temporal Heterogeneity On The Population Dynamics Of Four Animal Species In A Danish Landscape, Richard M. Sibly, Jacob Nabe-Nielsen, Mads C. Forchhammer, Valery E. Forbes, Christopher J. Topping Jun 2009

Topping

Valery Forbes Publications

Background: Variation in carrying capacity and population return rates is generally ignored in traditional studies of population dynamics. Variation is hard to study in the field because of difficulties controlling the environment in order to obtain statistical replicates, and because of the scale and expense of experimenting on populations. There may also be ethical issues. To circumvent these problems we used detailed simulations of the simultaneous behaviours of interacting animals in an accurate facsimile of a real Danish landscape. The models incorporate as much as possible of the behaviour and ecology of skylarks Alauda arvensis, voles Microtus agrestis, …

Thyroid Hormone Regulation Of Cholesterol Metabolism, Lindsey R. Boone Jun 2009

Thyroid Hormone Regulation Of Cholesterol Metabolism, Lindsey R. Boone

USF Tampa Graduate Theses and Dissertations

In this study, we examined the effects of thyroid hormone on regulatory processes of cholesterol metabolism. Specifically, the pathways of cholesterol synthesis and cholesterol efflux were investigated. Hepatic HMG-CoA reductase (HMGR) is the rate-limiting enzyme in cholesterol synthesis. Hypothyroid rats exhibit decreased expression of this gene, which can be induced by subsequent treatment with thyroid hormone. The mechanism of this activation was previously unknown. Utilizing in vivo electroporation, we identified HMGR promoter elements necessary for the induction of HMGR by thyroid hormone. The -316/-321 element, the sterol response element, and nuclear factor-y site were all found to be necessary to …

Assessing The Impact Of Biodiversity On Tourism Flows: A Model For Tourist Behaviour And Its Policy Implications, Giulia Macagno, Maria Loureiro, Paulo A.L.D. Nunes, Richard Tol

Fondazione Eni Enrico Mattei Working Papers

This analysis provides an example of how biodiversity can be measured by means of different indicators, and how the latter can be used to assess the influence of the biodiversity profile of a region on the tourism flows towards it. Previous studies have considered environmental amenities as one of the determinants of tourism destination choice. The central hypothesis of this paper is that the destination’s biodiversity profile can be considered as a key component of environmental amenities. The main objective of this study is to propose a different perspective on this topic, considering the role of biodiversity on tourists’ choice …

Conservation Of The Glucan Phosphatase Laforin Is Linked To Rates Of Molecular Evolution And The Glucan Metabolism Of The Organism, Matthew S. Gentry, Rachel M. Pace Jun 2009

Conservation Of The Glucan Phosphatase Laforin Is Linked To Rates Of Molecular Evolution And The Glucan Metabolism Of The Organism, Matthew S. Gentry, Rachel M. Pace

Molecular and Cellular Biochemistry Faculty Publications

BACKGROUND: Lafora disease (LD) is a fatal autosomal recessive neurodegenerative disease. A hallmark of LD is cytoplasmic accumulation of insoluble glucans, called Lafora bodies (LBs). Mutations in the gene encoding the phosphatase laforin account for approximately 50% of LD cases, and this gene is conserved in all vertebrates. We recently demonstrated that laforin is the founding member of a unique class of phosphatases that dephosphorylate glucans.

RESULTS: Herein, we identify laforin orthologs in a protist and two invertebrate genomes, and report that laforin is absent in the vast majority of protozoan genomes and it is lacking in all other invertebrate …
