Background Dysregulation of the insulin-like growth factor (IGF) system, a common consequence of adiposity-induced insulin resistance, may be a key underlying mechanism linking excess body weight with colon cancer. Evidence has been derived from studies of cancer and polyps. Supporting data about aberrant crypt foci (ACF), putative pre-polyp changes, have been generated only from animal studies to date. Methods We randomly selected 26 patients with sex-specific elevated waist-hip-ratio (WHR) and 26 with normal values from a series of 150 patients seeking routine colonoscopy at the University of Connecticut Health Center. Cross-sectional analyses were performed of ACF number (<5, ≥5) in relation to total IGF1, IGF-binding protein-3 (IGFBP3), insulin, body mass index (BMI), WHR and waist circumference (WC). Visualized ACF in the 20 cm of the distal colon were counted using advanced endoscopic imaging. Results Patients …

Statistical approaches to evaluate higher order SNP-SNP and SNP-environment interactions are critical in genetic association studies, as susceptibility to complex disease is likely to be related to the interaction of multiple SNPs and environmental factors. Logic regression (Kooperberg et al., 2001; Ruczinski et al., 2003) is one such approach, where interactions between SNPs and environmental variables are assessed in a regression framework, and interactions become part of the model search space. In this manuscript we extend the logic regression methodology, originally developed for cohort and case-control studies, for studies of trios with affected probands. Introduction: Multiple myeloma (MM), a malignancy of plasma cells, accounts for 10% of all haematological malignancies and is currently incurable. Although it can be treated, the disease tends to relapse after several years and becomes increasingly resistant to conventional therapy. Investigations into using humoral therapy for MM are now underway with a view that novel therapeutic agents may provide a more targeted therapy for MM.

Materials and Methods: Here, phage display, a faster and more efficient method compared to classical hybridoma fusion technology, was used as a proof-of-concept to isolate several single-chain Fragment variables (scFv) against Ku86.

Background: Aged skeletal muscle is characterized by an increased incidence of metabolic and functional disorders, which if allowed to proceed unchecked can lead to increased morbidity and mortality. The mechanism(s) underlying the development of these disorders in aging skeletal muscle are not well understood. Protein kinase B (Akt/PKB) is an important regulator of cellular metabolism and survival, but it is unclear if aged muscle exhibits alterations in Akt function. Here we report a novel dysfunction of Akt in aging muscle, which may relate to S-nitrosylation and can be prevented by acetaminophen intervention.

In response to water quality concerns in the Jordan Lake Reservoir and state and federal mandates, several cities in North Carolina are being required for the first time to reduce nutrient loads in stormwater from previously developed lands; that is, install retrofits. It is anticipated that similar requirements will become necessary for other urban areas as North Carolina. The goal of this study is to evaluate the feasibility of alternative approaches to stormwater management for existing developments within North Carolina cities. Race Disparities In Birth Outcomes In The U.S. South And The Rest Of The Nation, Lenna Nepomnyaschy

University of Kentucky Center for Poverty Research Discussion Paper Series

There are well-documented and as yet unexplained disparities in birth outcomes by race in the United States, even after controlling for socioeconomic status. This paper examines the sources of disparities in low birth weight between blacks and whites in the U.S., by focusing on differences in disparities between two very distinct geographic areas, the Deep South and the rest of the country. Two findings from prior research drive the analyses: First, health overall is worse in the Deep South states; Second, race disparities are smaller in the Deep South than in the rest of the nation. Community Wildfire Protection Plans: Reducing Wildfire Hazards In The Wildland Urban Interface, Gail Wells

Joint Fire Science Program Digests

The 2003 Healthy Forests Restoration Act calls for local communities in the wildland-urban interface to collaborate on developing Community Wildfire Protection Plans (CWPPs) to reduce their wildfire hazard. To craft a successful CWPP, a community must collaborate effectively. A Joint Fire Science Program-sponsored research team studied 15 communities as they developed CWPPs. They found that social networks, learning communities, and community capacity were key indicators of success, and that working together on a CWPP can enhance a community’s capacity to collaborate, helping it address future challenges more skillfully.

The American Association of School Librarians’ Standards for the 21st Century Learner make clear that information skills alone are not sufficient for student success; students must also value those skills, use them in a productive and responsible manner, and have the motivational “dispositions in action” to support successful research and independent lifelong learning. Self-determination theory highlights perceived competence and autonomy as two basic psychological needs that support intrinsically-motivated behavior. Acquisition and compilation of water quality data for a ten year time period (1996 – 2006) from 589 stream and river stations was conducted to support nutrient criteria development for the multi–state Red River Basin shared by Arkansas, Louisiana, New Mexico, Oklahoma and Texas, USA. Twenty–three water quality parameters were collected from five data sources (USGS, ADEQ, LDEQ, OCC, OWRB, and TCEQ) and an additional 13 parameters were acquired from at least one source. Data for the primary biological parameter of interest, chlorophyll a, was sparse and available from only two sources. Following compilation of data, medians were calculated for …

Heterosternuta sulphuria is an endemic aquatic species of concern in Arkansas, with a priority score of 80 out of 100 and a conservation rank of S1and G1. A need of the Arkansas Wildlife Action Plan (AWAP) was to obtain baseline information on distribution and population status of H. sulphuria. Here, we report new H. sulphuria records for 39 sites across 10 counties in the Ozark Highlands and Boston Mountain ecoregions and a determined habitat type of shallow margins and small bedrock pools of perennial streams and spring seeps. The cultural and political implications of landscape change and urban growth in the western U.S. are well-documented. However, comparatively little scholarship has examined the effects of urbanization on sense of place in the southern U.S. We contribute to the literature on competing place meanings with a case study from the rural “Sewee to Santee” region of northern Charleston County, SC. Our research highlights conflicting cultural, environmental, and racial politics and their roles in struggles over place meanings. Structural Evidence For Consecutive Hel308-Like Modules In The Spliceosomal Atpase Brr2, L. Zhang, T. Xu, Corina Maeder, L.-O. Bud, J. Shanks, J. Nix, C. Guthrie, J. A. Pleiss, R. Zhao

Brr2 is a DExD/H-box helicase responsible for U4/U6 unwinding during spliceosomal activation. Brr2 contains two helicase-like domains, each of which is followed by a Sec63 domain with unknown function. We determined the crystal structure of the second Sec63 domain, which unexpectedly resembles domains 4 and 5 of DNA helicase Hel308. This, together with sequence similarities between Brr2's helicase-like domains and domains 1-3 of Hel308, led us to hypothesize that Brr2 contains two consecutive Hel308-like modules (Hel308-I and Hel308-II). Our structural model and mutagenesis data suggest that Brr2 shares a similar helicase mechanism with Hel308. We demonstrate that Hel308-II interacts with …

Cockburn Sound is the largest of the very few protected marine embayments along the lower west coast of Western Australia. It has been recognised as playing an integral role in the life history strategies of many marine species, including the highly valued snapper Pagrus auratus and blue swimmer crab Portunus pelagicus. Currently, the adult stocks of snapper in the West Coast Bioregion and blue swimmer crabs in Cockburn Sound are at depleted levels, most likely a result of high fishing pressure and below average recruitment in recent years for both species.
