Monitoring, Identification, And Intervention For Metabolic Disorders In Veterans With Psychotic Disorders, Michael Swtye Jul 2009

Monitoring, Identification, And Intervention For Metabolic Disorders In Veterans With Psychotic Disorders, Michael Swtye

Yale Medicine Thesis Digital Library

In light of growing evidence that certain antipsychotics may cause potentially life-threatening metabolic side-effects, the purpose of this study was to determine how regularly mental health clinicians (MHCs) currently monitor and manage metabolic abnormalities in overweight and obese patients with psychotic disorders. We hypothesized that MHCs monitor, identify and intervene for metabolic abnormalities in their patients at significantly lower rates than primary care physicians (PCCs), and that such rates may jeopardize patient health. We performed a one-year cross-sectional medical record review of primary care and mental health routine outpatient visit notes from the West Haven Campus of the Veteran Affairs …

The Wrong Wright Stuff : Mapping Human Error In Aviation, Stacey Hendrickson Jul 2009

The Wrong Wright Stuff : Mapping Human Error In Aviation, Stacey Hendrickson

Psychology ETDs

The Aviation Safety Reporting System (ASRS) was instituted to aid the Federal Aviation Administration in tracking trends in aviation incidents so that, ultimately, safety measures and training could be implemented to decrease the occurrence of accidents and incidents within the industry. The current system relies on hand coding of reports to recognize current trends and alert the proper parties. Although the filing party may enter some codified data describing the surrounding scenario (e.g., time of day, weather), there is no opportunity to specify a category if the problem is human error. Considering the prevalence of human error within these incidents …

Chimeras And Jagunços: Positivist Discourse In Euclides Da Cunha's Os Sertões, Justin Barber Jul 2009

Chimeras And Jagunços: Positivist Discourse In Euclides Da Cunha's Os Sertões, Justin Barber

History ETDs

The present study elucidates the discursive practice of positivism, as particularly manifested in late nineteenth-century Brazil. Founded in 1889, the Republic of the United States of Brazil was constructed as a positivist state by republican idealists and pragmatic oligarchs; the political embodiment of ordem e progresso (order and progress). It was a form of government meant to overcome Brazils imagined and concrete backwardness by strictly adhering to the Natural Laws identified by Auguste Comte, Charles Darwin, and, in particular, Herbert Spencer. Positivist progress was, however, contingent upon the elimination of the pathological, amorphous 'savage lower races. Non-white degeneration in Brazil, …

Theses and Dissertations

The Affinity 2.0 has been recently upgraded to the Affinity 2.5. Both instruments purport to measure sexual interest using viewing time as a form of measurement using non-pornographic images of people of varying ages and genders. The Affinity 2.5 increased the number of slides by 42.9%. This increase may have impacted the expected Chi square weights established for the Affinity 2.0 for non-pedophilic, exclusively heterosexual males. The purpose of this study was to create new expected Chi square weights for the Affinity 2.5 for non-pedophilic, exclusively heterosexual males. Additionally, this study re-examined the temporal stability of the Affinity 2.0 using …

Ccm2 Molecular Signaling Pathway, Arianne J. Boylan Jul 2009

Ccm2 Molecular Signaling Pathway, Arianne J. Boylan

Yale Medicine Thesis Digital Library

Cerebral cavernous malformations (CCM) are a central nervous system vascular malformation often responsible for hemorrhagic strokes. Molecular genetic studies have identified three genes (CCMl KRITI, CCM2 Malcavernin, and CCM3 PDCDIO) and four possible loci responsible for the pathogenesis of these lesions. CCMl functions through integrin signaling and regulation of RACl activity and may be involved in the MAPK and JNK signaling cascades. We hypothesized that CCM2 likely functioned through the same pathways and that CCM3 expression is regulated by these stress-induced signaling cascades. We showed that CCM2 likely functions through the MAPK pathway as the mouse homolog, osmosensing scaffold protein …

Palmitic Acid Increases Enac Expression And Contributes To Insulin Resistance In Renal Distal Tubule, Timmy Sullivan Jul 2009

Palmitic Acid Increases Enac Expression And Contributes To Insulin Resistance In Renal Distal Tubule, Timmy Sullivan

Yale Medicine Thesis Digital Library

Insulin resistance and obesity are two conditions associated with hypertension. Although the causes of hypertension are likely to be multifactorial, dysregulation of renal distal tubule sodium reabsorption may play a role. It has been shown that insulin increases sodium reabsorption in the distal tubule through the increased expression of epithelial sodium channels (ENaC); and obesity has been shown to cause insulin resistance in various tissues. Thiazolidinediones (TZDs) are insulin-sensitizing drugs whose use can be limited by a side effect of fluid-retention leading to edema and congestive heart failure. We set out to determine if the expression of ENaC in distal …

"Sparks From Law's Anvil": The Evolution Controversies In New Mexico, 1925-2008, Peter Incaudo

History ETDs

After attaining statehood in 1912, New Mexico witnessed an influx of American people, culture and controversies. This thesis tracks the development of controversies over the theory of evolution in the public schools of the state of New Mexico during the twentieth century and into the twenty-first century. Beginning with the states reaction to the 1925 Scopes trial, I discuss the reasons that the evolution controversy remained largely muted in 1920s New Mexico. Next, I examine the brief, but unique, 1927 evolution controversy sparked by a rumor of legislation to mandate the teaching of evolution. From there, I describe New Mexico's …

The 'Uncanny' And The Android, Noah Cooperstein Jul 2009

The 'Uncanny' And The Android, Noah Cooperstein

Foreign Languages & Literatures ETDs

The character of the android is found widely in film and literature. While she appears across the entire spectrum of genres, she most often makes her appearance in the uncanny text. This appearance is nearly always accompanied by some variation of the vision motif. Despite wide spread interest in both the Uncanny' and the android, to date, there is not a theory which accounts for the uncanny nature of the android and the prevalence of the vision motif in the android text. This paper will attempt to develop just such a theory. Any paper that addresses the 'Uncanny' must begin …

Salmonella Enterica Serovar Typhimurium Adhesion And Cytotoxicity During Epithelial Cell Stress Is Reduced By Lactobacillus Rhamnosus Gg, Kristen Burkholder, Arun K. Bhunia

Department of Food Science Faculty Publications

Background

Physiological stressors may alter susceptibility of the host intestinal epithelium to infection by enteric pathogens. In the current study, cytotoxic effect, adhesion and invasion of Salmonella enterica serovar Typhimurium (S. Typhimurium) to Caco-2 cells exposed to thermal stress (41°C, 1 h) was investigated. Probiotic bacteria have been shown to reduce interaction of pathogens with the epithelium under non-stress conditions and may have a significant effect on epithelial viability during infection; however, probiotic effect on pathogen interaction with epithelial cells under physiological stress is not known. Therefore, we investigated the influence of Lactobacillus rhamnosus GG and Lactobacillus gasseri …

The Effect Of Aging On The Innate Immune Response Of Vascular Smooth Muscle, Christopher Winterbottom Jul 2009

The Effect Of Aging On The Innate Immune Response Of Vascular Smooth Muscle, Christopher Winterbottom

Yale Medicine Thesis Digital Library

As our understanding of atherosclerosis has become more sophisticated, a picture of the disease has emerged which emphasizes the role of inflammation in the pathogenesis of the disease, including signaling cascades mediated by toll-like receptors (TLRs). Data has also emerged suggesting that the process of aging plays an important role in atherogenesis, through pathways at least partially mediated by changes in the function of vascular smooth muscle cells. The purpose of this study was to elucidate the role that aging might play in the TLR-dependent innate immune response of vascular smooth muscle. Ex vivo cell and tissue culture models were …

Ion Transport Behaviors Upstream And Downstream From The Sampling Cone Of An Inductively Coupled Plasma Mass Spectrometer, Haibin Ma Jul 2009

Ion Transport Behaviors Upstream And Downstream From The Sampling Cone Of An Inductively Coupled Plasma Mass Spectrometer, Haibin Ma

Theses and Dissertations

Inductively coupled plasma - mass spectrometry (ICP-MS) is the technique of choice worldwide for trace elemental determinations because of its excellent ionization ability, low detection limits and fast analysis speeds. However, the ICP-MS still suffers from some disadvantages, such as spectral overlap and severe matrix effects. Matrix effects or interferences, partly arise from changes in the analyte transmission through the interfacial region between the ICP and mass spectrometer with changes in sample matrix. Better understanding of the transmission behaviors of analyte through the sampling and skimmer cones will provide the insights needed to alleviate matrix interferences and to improve the …

Relaxation Of Vibrationally Excited Trifluorobenzene And Tetrafluorobenzene By Collisions With Carbon Dioxide, Alan M. Johnson Jul 2009

Relaxation Of Vibrationally Excited Trifluorobenzene And Tetrafluorobenzene By Collisions With Carbon Dioxide, Alan M. Johnson

Theses and Dissertations

An investigation into the relaxation of highly vibrationally excited trifluorobenzene and tetrafluorobenzene following collisions with carbon dioxide was performed using diode laser transient absorption spectroscopy. A 248 nm excimer laser prepared the vibrationally hot (E'~41,000 cm-1) fluorobenzene molecules. Large amounts of translational and rotational energy are transferred through collisions between the hot donor molecule and CO2. Rate constants and collisional probabilities were calculated by probing the high J states (J=58~80) of CO2 in the vibrational ground state, 0000, with measurements taken 1 µsec, ¼ the mean gas collision time, following each excimer laser pulse. The energy transfer probability distribution function, …

Theses and Dissertations

This dissertation synthesizes and analyzes an emblematic sample of three prevalent psychological approaches to organizational change and learning, giving particular attention to the conception of cognition and emotion. It also explores some of the philosophical and psychological assumptions undergirding these approaches. A web model depicting various epistemological influences is offered as a tool for exploring influences on the psychological research within and beyond this study. A second conceptual model is also offered as a tool for considering the hierarchical treatment and preferential placement of cognition over emotion theory and practice. The project draws on general philosophy, psychology, evolutionary theory, and …

Sphingosine Kinase 1 Inhibitor, A Novel Inducer Of Autophagy, Daniel Meza Jul 2009

Sphingosine Kinase 1 Inhibitor, A Novel Inducer Of Autophagy, Daniel Meza

Theses and Dissertations

Autophagy is the process of “cell self-eating” which has been implicated both in cell survival and cell death. Sphingosine kinase 1 (SphK1) regulates the intracellular balance between ceramide and sphingosine, bioactive lipids associated with cell death, and sphingosine-1-phosphate (S1P), whose actions are associated with survival and proliferation. Previous studies have implicated upregulation of SphK1 in the induction of autophagy. In this study, SK1-I, a SphK1 specific competitive inhibitor, induced autophagy in a concentration and time dependent manner in HCT116 colorectal carcinoma cells. This autophagic response was observed to be more intense in wild type p53 expressing HCT116 cells than in …

Nanofiber Scaffolds With Gradations In Mineral Content For Mimicking The Tendon-To-Bone Insertion Site, Xiaoran Li, Jingwei Xie, Justin Lipner, Xiaoyan Yuan, Stavros Thomopoulos, Younan Xia

MIIR Faculty Research

We have demonstrated a simple and versatile method for generating a continuously graded, bonelike calcium phosphate coating on a nonwoven mat of electrospun nanofibers. A linear gradient in calcium phosphate content could be achieved across the surface of the nanofiber mat. The gradient had functional consequences with regard to stiffness and biological activity. Specifically, the gradient in mineral content resulted in a gradient in the stiffness of the scaffold and further influenced the activity of mouse preosteoblast MC3T3 cells. This new class of nanofiberbased scaffolds can potentially be employed for repairing the tendon-to-bone insertion site via a tissue engineering approach.

Single-Molecule Protein Unfolding In Solid State Nanopores, David Talaga, Jiali Li Jul 2009

Single-Molecule Protein Unfolding In Solid State Nanopores, David Talaga, Jiali Li

Department of Chemistry and Biochemistry Faculty Scholarship and Creative Works

We use single silicon nitride nanopores to study folded, partially folded, and unfolded single proteins by measuring their excluded volumes. The DNA-calibrated translocation signals of β-lactoglobulin and histidine-containing phosphocarrier protein match quantitatively with that predicted by a simple sum of the partial volumes of the amino acids in the polypeptide segment inside the pore when translocation stalls due to the primary charge sequence. Our analysis suggests that the majority of the protein molecules were linear or looped during translocation and that the electrical forces present under physiologically relevant potentials can unfold proteins. Our results show that the nanopore translocation signals …
