Self-Assembly And Disassembly Of The Snare Complex: Examined Using Circular Dichroism And Atomic Force Microscopy, Jeremy D. Cook, Won Jin Cho, Timothy L. Stemmler, Bhanu P. Jena

In this study, we report for the first time that both t-SNAREs and v-SNARE and their complexes in buffered suspension, exhibit defined peaks at CD signals of 208 and 222 nm wavelengths, consistent with a higher degree of helical secondary structure. Surprisingly, when incorporated in lipid membrane, both SNAREs and their complexes exhibit reduced folding. In presence of NSF-ATP, the SNARE complex disassembles, as reflected from the CD signals demonstrating elimination of α-helices within the structure.

Two novel sulfate-reducing bacteria, strains CY1 and CY2, were isolated from heavy-metal-contaminated sediments of Lake Coeur d'Alene, Idaho, USA. Strains CY1 and CY2 were found to contain c-type cytochromes and to reduce sulfate, sulfite, thiosulfate, elemental sulfur, DMSO, anthraquinone disulfonate and fumarate using lactate as an electron donor. In a comparison of 16S rRNA gene sequences, CY1 and CY2 were found to be 100% identical, but only 97 and 92.4% similar, respectively, to the type strains of Desulfovibrio mexicanus and Desulfovibrio aminophilus. Adenylyl cyclases (ACs) catalyze the conversion of ATP into the second messenger cAMP and play a key role in signal transduction. In a recent study (Mol Pharmacol 70: 878-886, 2006), we reported that 2',3'-O-(2,4,6-trinitrophenyl)-substituted nucleoside 5'-triphosphates (TNP-NTPs) are potent inhibitors (K(i) values in the 10 nM range) of the purified catalytic subunits VC1 and IIC2 of membranous AC (mAC). The crystal structure of VC1: IIC2 in complex with TNP-ATP revealed that the nucleotide binds to the catalytic site with the TNP-group projecting into a hydrophobic pocket. Phylogenies derived from gene order data may prove crucial in answering some fundamental open questions in biomolecular evolution. Yet very few techniques are available for such phylogenetic reconstructions. One method is breakpoint analysis, developed by Blanchette and Sankoff 2 for solving the breakpoint phylogeny.' Our earlier studies 5;6 confirmed the usefulness of this approach, but also found that BPAnalysis, the implementation developed by Sankoff and Blanchette, was too slow to use on all but very small datasets. Prediction Of Antigenic Epitopes On Protein Surfaces By Consensus Scoring, Shide Liang, Dandan Zheng, Chi Zhang, Martin Zacharias

Background: Prediction of antigenic epitopes on protein surfaces is important for vaccine design. Most existing epitope prediction methods focus on protein sequences to predict continuous epitopes linear in sequence. Only a few structure-based epitope prediction algorithms are available and they have not yet shown satisfying performance.
Results: We present a new antigen Epitope Prediction method, which uses ConsEnsus Scoring (EPCES) from six different scoring functions - residue epitope propensity, conservation score, sidechain energy score, contact number, surface planarity score, and secondary structure composition. Applied to unbounded antigen structures from an independent test set, EPCES was able to predict antigenic eptitopes …

Great northern common bean (Phaseolus vulgaris L.) ‘Coyne’ (Reg. No. CV-287, PI 655574) was developed by the dry bean breeding program at the University of Nebraska Agricultural Research Division and released in 2008. It was bred specifically for adaptation to Nebraska growing conditions and for enhanced resistance to common bacterial blight (CBB), a major disease of common bean caused by the seed-borne bacterium Xanthomonas campestris pv. phaseoli (Smith) Dye, and bean common rust Uromyces appendiculatus (Pers.:Pers) Unger. Coyne is a great northern F7:8 line derived from a three-way cross (G95023/Weihing//BelMiNeb-RMR-11). The first cross was made in winter 2003. …
