Ethylene Dichloride Disruption Of Fertility In Male Mice, Harold J. Daigle, Derek N. Cole, J. Andrew Carlson, William R. Lee, Vincent L. Wilson
Faculty Publications
Ethylene dichloride (EDC) is a high use compound in chemical industry today. Although a potent alkylating agent and carcinogen, EDC has not been associated previously with adverse fertility consequences. Intraperitoneal 5 to 40 mg/kg once a day for five days in C57BL/6 mice rendered males infertile for 6 months and longer. Two of three mice treated with EDC at 5 mg/kg recovered to fertility after a 3 and 5-week sterile period, respectively. Significant testicular pathology was evident within 8 days post treatment with EDC, which progressed to resemble Sertoli Cell-Only syndrome. Although the observed effects on Leydig cell populations were …

Pore Size Characterization Of Monolithic Capillary Columns Using Capillary Flow Porometry, Yan Fang
Pore Size Characterization Of Monolithic Capillary Columns Using Capillary Flow Porometry, Yan Fang
Theses and Dissertations
A simple capillary flow porometer (CFP) was assembled for pore structure characterization of monolithic capillary liquid chromatography columns based on ASTM standard F316-86. Determination of differential pressures and flow rates through dry and wet samples provided the necessary information to determine the through-pore throat diameter, bubble point pore diameter, mean flow pore diameter, and pore distribution. Unlike measurements in bulk using traditional techniques to provide indirect information about the pore properties of monolithic columns, monoliths can be characterized in their original chromatographic forms with this system. The performance of the new CFP was first evaluated by characterizing the pore size …

Inflammasome-Dependent Caspase-1 Activation In Cervical Epithelial Cells Stimulates Growth Of The Intracellular Pathogen Chlamydia Trachomatis, Ali A. Abdul-Sater, Evonne Koo, Georg Hacker, David M. Ojcius
All Dugoni School of Dentistry Faculty Articles
Inflammasomes have been extensively characterized in monocytes and macrophages, but not in epithelial cells, which are the preferred host cells for many pathogens. Here we show that cervical epithelial cells express a functional inflammasome. Infection of the cells by Chlamydia trachomatis leads to activation of caspase-1, through a process requiring the NOD-like receptor family member NLRP3 and the inflammasome adaptor protein ASC. Secretion of newly synthesized virulence proteins from the chlamydial vacuole through a type III secretion apparatus results in efflux of K+ through glibenclamide-sensitive K+ channels, which in turn stimulates production of reactive oxygen species. Elevated levels of reactive …

David Berkowitz Publications
This overview focuses on the (α,α-difluoromethylene)phosphonate mimic of phosphoserine (pCF2Ser) and its application to the study of kinase-mediated signal transduction – pathways of great interest to drug development. The most versatile modes of access to these chemical biological tools are discussed, organized by method of PCF2-C bond. The pCF2-Ser mimic may be site-specifically incorporated into peptides (SPPS) and proteins (expressed protein ligation). This isopolar, dianionic pSer mimic results in a “constitutive phosphorylation” phenotype, and is seen to support native protein-protein interactions that depend upon serine phosphorylation. Signal transduction pathways studied with this chemical biological …
The Regulation Of Alternative Splicing By Oncogenic Signaling Pathways., Jacqueline Shultz
The Regulation Of Alternative Splicing By Oncogenic Signaling Pathways., Jacqueline Shultz
Theses and Dissertations
In the presented study, we demonstrate that the alternative splicing of caspase 9 was dysregulated in a large percentage of NSCLC tumors and cell lines. These findings led to the hypothesis that survival pathways activated by oncogenic mutation regulated this mechanism. Indeed, the oncogenic PI3-Kinase/Akt pathway was demonstrated to regulate the alternative splicing of caspase 9. Further mechanistic studies demonstrate that multiple Akt isoforms can regulate the alternative splicing of caspase 9 in NSCLC. Akt was additionally shown to mediate the exclusion of the exon 3,4,5,6 cassette of caspase 9 via the phospho-state of the RNA trans-factor, SRp30a. Mutagenesis studies …
Literary Bodies: The Novel As Experience, Britt Dienes
Literary Bodies: The Novel As Experience, Britt Dienes
USF Tampa Graduate Theses and Dissertations
For my MA thesis I propose to examine a series of novels that combine motifs of the body with structural and linguistic experimentation that parallels the state of the bodies within the text. Using Tsitsi Dangarembga's 1988 "bodybildungsroman" Nervous Conditions, Sherley Anne Williams' 1986 neo -slave narrative Dessa Rose, Samuel Beckett's 1938 existential novel Murphy, Vikram Seth's 1986 poetic novel Golden Gate, and Vladimir Nabokov's 1962 poetic novel Pale Fire, I will argue that these texts portray the body as a readable space of culture, a legible site of conflict or creation.
I contend that these novels …

Journal Articles: Biochemistry & Molecular Biology
Free fatty acids (FFA) induce hepatocyte lipoapoptosis by a c-Jun N-terminal kinase (JNK)-dependent mechanism. However, the cellular processes by which JNK engages the core apoptotic machinery during lipotoxicity, especially activation of BH3-only proteins, remain incompletely understood. Thus, our aim was to determine whether JNK mediates induction of BH3-only proteins during hepatocyte lipoapoptosis. The saturated FFA palmitate, but not the monounsaturated FFA oleate, induces an increase in PUMA mRNA and protein levels. Palmitate induction of PUMA was JNK1-dependent in primary murine hepatocytes. Palmitate-mediated PUMA expression was inhibited by a dominant negative c-Jun, and direct binding of a phosphorylated c-Jun containing the …

Role Of Mammalian Ecdysoneless In Cell Cycle Regulation., Jun Hyun Kim, Channabasavaiah B. Gurumurthy, Mayumi Naramura, Ying Zhang, Andrew T. Dudley, Lynn Doglio, Hamid Band, Vimla Band
Journal Articles: Genetics, Cell Biology & Anatomy
The Ecdysoneless (Ecd) protein is required for cell-autonomous roles in development and oogenesis in Drosophila, but the function of its evolutionarily conserved mammalian orthologs is not clear. To study the cellular function of Ecd in mammalian cells, we generated Ecd(lox/lox) mouse embryonic fibroblast cells from Ecd floxed mouse embryos. Cre-mediated deletion of Ecd in Ecd(lox/lox) mouse embryonic fibroblasts led to a proliferative block due to a delay in G(1)-S cell cycle progression; this defect was reversed by the introduction of human Ecd. Loss of Ecd led to marked down-regulation of E2F target gene expression. Furthermore, Ecd directly bound to Rb …

We study the effects of the perturbation which violates the permutation symmetry of three Majorana neutrinos but preserves the well known (23) interchange symmetry. This is done in the presence of an arbitrary Majorana phase which serves to insure the degeneracy of the three neutrinos at the unperturbed level.

Attachment: The Antidote To Trauma, Joshua Straub
Faculty Publications and Presentations
Trauma and loss in life are inevitable. And all too often the traumatic experience itself can be enough to paralyze the mental, emotional, and spiritual state of any given person. Unable to interpret the traumatic experience, many instead are left defined by it. Helping clients discern the objective experience and their subjective reactions to it will help free them from the emotions and beliefs that subsequently control their lives. Based on the most relevant attachment theory research and clinical techniques, this workshop teaches the attentional strategies necessary to helping clients overcome trauma.
