St. John's Wort Inhibits Adipocyte Differentiation And Induces Insulin Resistance In Adipocytes, Zhaleh Amini, Bryant Boyd, Julie Doucet, David M. Ribnicky, Jacqueline M. Stephens

Adipocytes are insulin sensitive cells that play a major role in energy homeostasis. Obesity is the primary disease of fat cells and a major risk factor for the development of Type II diabetes, cardiovascular disease, and metabolic syndrome. Obesity and its related disorders result in dysregulation of the mechanisms that control adipocyte gene expression and function. To identify potential novel therapeutic modulators of adipocytes, we screened 425 botanical extracts for their ability to modulate adipogenesis and insulin sensitivity. We observed that less than 2% of the extracts had substantial effects on adipocyte differentiation of 3T3-L1 cells. Two of the botanical …

Engineers who attempt to discover and optimize the behavior of complex biochemical systems face a dauntingly difficult task. This is especially true if the systems are governed by multiple qualitative and quantitative variables that have non-linear response functions and that interact synergistically. The synthetic biology community has responded to this difficulty by promoting the use of "standard biological parts" called "BioBricks", which are supposed to make biology into traditional engineering and enable engineers to "program living organisms in the same way a computer scientists can program a computer". IkappaB kinase beta (IKKbeta) is involved in tumor development and progression through activation of the nuclear factor (NF)-kappaB pathway. However, the molecular mechanism that regulates IKKbeta degradation remains largely unknown. Here, we show that a Cullin 3 (CUL3)-based ubiquitin ligase, Kelch-like ECH-associated protein 1 (KEAP1), is responsible for IKKbeta ubiquitination. Depletion of KEAP1 led to the accumulation and stabilization of IKKbeta and to upregulation of NF-kappaB-derived tumor angiogenic factors. A systematic analysis of the CUL3, KEAP1, and RBX1 genomic loci revealed a high percentage of genome loss and missense mutations in human cancers that failed to facilitate IKKbeta degradation. Callipyge sheep exhibit extreme postnatal muscle hypertrophy in the loin and hindquarters as a result of a single nucleotide polymorphism (SNP) in the imprinted DLK1-DIO3 domain on ovine chromosome 18. The callipyge SNP up-regulates the expression of surrounding transcripts when inherited in cis without altering their allele-specific imprinting status. The callipyge phenotype exhibits polar overdominant inheritance since only paternal heterozygous animals have muscle hypertrophy. Molecular Signatures Reveal Circadian Clocks May Orchestrate The Homeorhetic Response To Lactation., Theresa Casey, Osman Patel, Karl Dykema, Heather Dover, Kyle Furge, Karen Plaut

Genes associated with lactation evolved more slowly than other genes in the mammalian genome. Higher conservation of milk and mammary genes suggest that species variation in milk composition is due in part to the environment and that we must look deeper into the genome for regulation of lactation. At the onset of lactation, metabolic changes are coordinated among multiple tissues through the endocrine system to accommodate the increased demand for nutrients and energy while allowing the animal to remain in homeostasis. A multisample, multistudy project aimed at understanding how individual differences in narcissism during early adolescence are related to distortions in the aggression, and the reactivity of the hypothalamic–pituitary–adrenal axis to negative and positive experiences. The findings indicate that individual differences in narcissism are a remarkably stable aspect of personality during early adolescence. It is predictably related to an inflated view of the self that is not warranted by objective indices of social functioning. Further evidence shows that it promotes the continuity of aggressive behavior and is more strongly related to reactive aggression than to proactive aggression and more strongly related …

Integrating experimental biology laboratory exercises with mathematical modeling can be an effective tool to enhance mathematical relevance for biologists and to emphasize biological realism for mathematicians. This paper describes a lab project de-signed for and tested in an undergraduate biomathematics course. In the lab, students follow and track the paths of individual brine shrimp confined in shallow salt water in a Petri dish. Students investigate the question, “Is the movement well characterized as a 2-dimensional random walk?” Through open, but directed discussions, students derive the corresponding partial differential equation, gain an understanding of the solution behavior, and model brine shrimp …

Four genera and 17 species of leucothoids, the majority of these belonging to the genus Leucothoe, are herein reported from the Great Barrier Reef. Fifteen species are new to science and only Anamixis bazimut has been previously reported from the Great Barrier Reef.
