Yeast Ste23p Shares Functional Similarities With Mammalian Insulin-Degrading Enzymes, Benjamin Alper, Jarrad W. Rowse, Walter K. Schmidt

The S. cerevisiae genome encodes two M16A enzymes: Axl1p and Ste23p. Of the two, Ste23p shares significantly higher sequence identity with M16A enzymes from other species, including mammalian insulin-degrading enzymes (IDEs). In this study, recombinant Ste23p and R. norvegicus IDE (RnIDE) were isolated from E. coli, and their enzymatic properties compared. Ste23p was found to cleave established RnIDE substrates including the amyloid-β peptide (Aβ 1-40) and insulin B chain. A novel internally quenched fluorogenic substrate (Abz-SEKKDNYIIKGVnitroY-OH) based on the polypeptide sequence of the yeast P2 a-factor mating propheromone was determined to be a suitable substrate for …

The Western Australian fishery is the only remaining significant fishery for pearl oysters in the world and comprises most of the exported pearl product of Australia, currently worth around $125 million annually. The majority of pearl shells used to culture pearls come from the pearling beds in the inshore waters near Broome. The fishery for pearl oysters preferentially targets smaller ‘culture’ shell (4-5 years age class, 120mm - 160mm shell length) that are more suitable for growing pearls, leaving larger MOP oysters (6+ age class, 175mm+) on the fishing grounds.

The biochemistry collection exists to support the teaching, research, and service endeavors of the Department of Biochemistry and the University of Nebraska-Lincoln community. The public may benefit from the biological sciences collection, although specific materials are not collected to meet their needs. Biochemistry-related materials not available in the library’s collection are generally readily available through Interlibrary Loan. The collection is focused on the QD Library of Congress call number range, although the heavily interdisciplinary nature of scientific study necessitates some overlap with the rest of Q, R, S, T, and other areas.

The effect of cologne/perfume on attractiveness was examined, as well as the differences between the effect of cologne/perfume and sex. Twenty-seven participants ranging from 18-22 years of age were recruited for the experiment. The purpose of this study was to determine if putting cologne/perfume on an individual will make that person more attractive to the opposite sex than if he/she did not put on cologne/perfume. The rationale behind this is that there are a lot of advertisements about cologne and perfume that portray the people in the advertisements as more desirable after he/she sprays cologne on their skin. We apply the “security-hierarchy paradox” to nuclear proliferation. Global security requires a certain amount of hierarchy. A world in which no nuclear proliferation rules exist to constrain states, for example, would not be secure. Global security requires legitimate and authoritative rules, which we define as rules that are mutually negotiated, binding to all and which provide a stable social order. Too much hierarchy, however, amounts to coercion and undermines global security. Rules that are not mutually negotiated, binding to all or do not provide a stable social order are not authoritative. Loricariidae is a very diverse family of catfishes found primarily in the Amazon River basin. These catfishes have a unique characteristic feature of having fenestrae (holes) in the skull region (compound pterotic bone) adjacent to their bi-lobed swim bladder. Since the swim bladders and the compound pterotic may act as an external ear for hearing in this taxon, I hypothesized that these swim bladders structures have an acoustical functional in the loricariid Otocinclus affinis. Microbial populations founded by a single clone and propagated under resource limitation can become polymorphic. We sought to elucidate genetic mechanisms whereby a polymorphism evolved in Escherichia coli under glucose limitation and persisted because of cross-feeding among multiple adaptive clones. Apart from a 29 kb deletion in the dominant clone, no large-scale genomic changes distinguished evolved clones from their common ancestor. Using transcriptional profiling on co-evolved clones cultured separately under glucose-limitation we identified 180 genes significantly altered in expression relative to the common ancestor grown under similar conditions. In this essay I will argue that the signature methods of interrogation used by CIA and military interrogators in the Global War on Terrorism (GWOT) -- torture lite' techniques such as hypothermia and stress positions -- may constitute torture, but that the question of their legality under U.S, and international law is not as straightforward as some critics of the Bush Administration maintained. I will take up only one thread in the complex discussion of GWOT interrogation practices and law, that of the boundary between torture and lesser cruelty.

Over the past decades, scholars have paid greater attention to sexual violence, in both theorization and empirical analysis. One area which has been largely ignored, however, is the sexual violence during times of armed conflict. This paper examines the nature and dynamics of sexual violence as it occurred during the 1994 Rwandan genocide. Drawing upon testimonies given to the International Criminal Tribunal for Rwanda (ICTR), descriptions of rapes--both singular and mass—were qualitatively analyzed. In general, three broad types of assaults were identified: opportunistic, assaults which seemed to be a product of the disorder inherent within the conflict; episodes of sexual …

Within the tuning methodology, a key step is the utilization of a consultative survey. The objectives established in the Bologna process for the tuning survey “include gleaning current perspectives on the diversity of practice and commonality of knowledge across borders and traditions, and seeking a simple and accessible language to create a scaffolding on which the various degrees can work in comfort and trust” (Adelman, 2009, p. 49). The Minnesota tuning surveys sought the consultation of previous graduates, current students, employers of graduates, and academic faculty for each of the disciplines involved in tuning.

Through this Professional Project, the author examines circulation and suitability pertaining to bicycle planning on a college campus, in the real-world setting of the University of Georgia (UGA) main campus located in Athens, Georgia.

Based on existing on-campus activity, bicycle circulation is established. Using an analytical tool, namely the bicycle level of service (BLOS) analysis, roadway segments within the study area are evaluated for suitability with respect to adjacent motor vehicle traffic. Several improvements are suggested throughout the project to enhance the existing BLOS, and in turn enhance the bicycling conditions. Most of the suggestions are based on traditional improvements, …

Background: The mitochondrial genomes of plants generally encode 30-40 identified protein-coding genes and a large number of lineage-specific ORFs. The lack of wide conservation for most ORFs suggests they are unlikely to be functional. However, an ORF, termed orf-bryo1, was recently found to be conserved among bryophytes suggesting that it might indeed encode a functional mitochondrial protein.

Results: From a broad survey of land plants, we have found that the orf-bryo1 gene is also conserved in the mitochondria of vascular plants and charophycean green algae. This gene is actively transcribed and RNA edited in many flowering plants. Comparative sequence …

Recent scientific findings about the developing teen brain have both captured public attention and begun to percolate through legal theory and practice. Indeed, many believe that developmental neuroscience contributed to the U.S. Supreme Court's elimination of the juvenile death penalty in Roper v. Simmons. Post-Roper, scholars assert that the developmentally normal attributes of the teen brain counsel differential treatment of young offenders, and advocates increasingly make such arguments before the courts. The success of any theory, though, depends in large part on implementation, and challenges that emerge through implementation illuminate problematic aspects of the theory. In November 2009, Sandia National Laboratories hosted the Next Generation Biofuels and Advanced Engines for Tomorrow’s Transportation Needs Workshop. The event focused on the combined opportunities in biofuels and engines in the transportation sector. The workshop brought together the DOE Combustion Research Facility and the DOE Joint BioEnergy Institute along with oil companies, biofuel developers, engine manufacturers, suppliers, and experts from the university, regulatory, finance, and national laboratory communities. The intersection of biofuels and engines, if properly scaled, can meet a triad of national goals:

Sensory processing disorder, characterized by over- or under-responsivity to non-noxious environmental stimuli, is a common but poorly understood disorder. We examined the role of prenatal alcohol exposure, serotonin transporter gene polymorphic region variation (rh5-HTTLPR), and striatal dopamine (DA) function on behavioral measures of sensory responsivity to repeated non-noxious sensory stimuli in macaque monkeys. Results indicated that early gestation alcohol exposure induced behavioral under-responsivity to environmental stimuli in monkeys carrying the short (s) rh5-HTTLPR allele compared to both early-exposed monkeys homozygous for the long (l) allele and monkeys from middle-to-late exposed pregnancies and controls, regardless of genotype. …
