Control Of Myosin Motor Activity And Actin Filament Translation By Alteration Of Assay Reducing Potential, Siva Krishna Nalabotu
Control Of Myosin Motor Activity And Actin Filament Translation By Alteration Of Assay Reducing Potential, Siva Krishna Nalabotu
Theses, Dissertations and Capstones
The use of biomolecular motors for nanotechnological applications has been impaired by an inability to control biomolecular motor activity in a temporal fashion. Previous data has suggested that myosin activity can be modulated by inducing changes in the degree of myosin oxidation. The hypothesis of this study is that myosin motor activity can be regulated by altering the amount of antioxidants and reducing agents present in assay solution. To test this hypothesis we systematically altered the concentration of reducing agents and examined the effects of these changes on myosin-dependent actin filament translation. Our findings indicate that myosin activity can be …

The global epidemic of childhood and adolescent overweight has become a major public health concern. Not only are these youth more likely to become obese as adults, and thus more prone to obesity-related diseases than their non overweight peers, they are also likely to suffer emotional and social effects associated with overweight. Overweight in youth has been linked to depression, low self-esteem, eating disorders, negative body image, and stigma. It appears to be bi-directional in nature, with overweight sometimes predicting certain psychological effects and psychosocial issues sometimes predicting overweight. Effective assessment and treatment of psychological and mental health issues in …
Silicone Rubber Passive Samplers For Water Quality Monitoring Of Persistent Organic Pollutants In The Marine Environment, Susan O'Hara
Silicone Rubber Passive Samplers For Water Quality Monitoring Of Persistent Organic Pollutants In The Marine Environment, Susan O'Hara
Masters
Polychlorinated Biphenyls (PCBs) and Polyaromatic Hydrocarbons (PAHs) are internationally considered as persistent organic pollutants (POPs) which can be mutagenic, carcinogenic and may accumulate in aquatic organisms and potentially in the human chain. PCBs and PAHs are thus listed as priority pollutants in the Water Framework Directive (WFD) and under the Convention for the protection of the Marine Environment of the North-East Atlantic (Oslo Paris Convention: OSPAR). Currently most marine monitoring programmes involving such compounds rely on the collection and analysis of discrete water/sediment grab samples or on the analysis of filter feeding indicator organisms e.g. mussels. Such measurements can provide …

Oestrogens are a group of steroid hormones of which oestradiol is the major form. Oestrogens are present in men and women and some of their main physiological roles involve female and male reproduction, bone metabolism and homeostasis. Oestrogens carry out their actions by diffusing across the cell membrane and binding to their receptor called the oestrogen receptor (ER), of which two types exist, ERα and ERβ. ERs regulate gene expression through the binding of DNA, most notably the estrogen response element (ERE). Breast cancer is the most common malignancy in women in industrialised countries and 65 percent of breast cancers …
Fluoride And Phosphate Glass Whispering Gallery Micro-Resonators, Jonathan Ward
Fluoride And Phosphate Glass Whispering Gallery Micro-Resonators, Jonathan Ward
Theses
This PhD work focuses on spherical microlasers made from commercial and experimental laser glasses. The laser light in a microsphere is confined in a region called a whispering gallery mode, which has unique properties that allow for many nonlinear effects to be observed with only sub-mW pump powers. Efficient coupling of light into the microsphere is attained by using adiabatically tapered fibres fabricated using various heating methods including a CO2 laser, a sapphire tube furnace and electric heaters. However, the best result is obtained using a fibre pulling rig with an oxy-butane torch as the heat source. This system …
An Assessment Of Epiphytic Lichens, Lichen Diversity And Environmental Quality In The Semi-Natural Woodlands Of Knocksink Wood Nature Reserve, Enniskerry, County Wicklow, Lenka Mulligan
Masters
This research work adapted and applied a recently developed method for assessing epiphytic lichen species diversity to the Irish semi-natural woodlands of Knocksink Wood Nature Reserve, Enniskerry, County Wicklow. The study focused on the differences that arise in relation to acidophilous oak woodland (Blechno-Quercetum petraeae) versus ash-hazel woodland (Corylo-Fraxinetum). The research also addressed differences in relation to the mixed oak-ash-hazel woodland located in Knocksink Wood and the neighbouring woodland at the Powerscourt Waterfall. The frequency of occurrence of lichen species on a defined portion of tree bark was used as an estimate of diversity and to evaluate the degree of …

Throughout history the oceans have directly or indirectly influenced humans. The importance of knowing how to protect this valuable resource and insure it for future generations is vital. Underwater Vehicles are tools essential for this process, and therefore research and development to perfect these devices is needed. However, the main goal of these devices--to transmit images from places where humans cannot go--remains the same, and their importance to future discoveries remains vital. This article discusses the different types of underwater vehicles and describes an activity wherein students will fabricate an underwater remotely operated vehicle.

The purpose of the present study was to examine the effects of individual difference factors, i.e., core self evaluations, cognitive ability, and task specific self-efficacy, on self-set goals and whether those effects were moderated by an assigned team goal in a pre-team context. It was hypothesized that the relationship between individual difference factors and self-set goals for potential team members would be differentially affected by the difficulty of the assigned team goal. I assessed these relationships for individual performance and individual satisfaction. In addition, I examined whether gender, task type, and team composition interacted in their effects on self-set goals. …

Examining Cultural Competence Among Osteopathic Medical Students , Sooni Lee
PCOM Psychology Dissertations
With the growing number of racial and ethnic minorities in the United States, there also has been a rise in health disparities. Ethnic minorities have been experiencing an overabundance of medical issues, particularly chronic diseases like cardiovascular disease, diabetes, and cancer. Although these chronic diseases are preventable, ethnic minorities are at greater risk for death from them. Possible contributing factors to the increase in health disparities include limited medical attention and follow-up appointments with physicians and the physician-patient relationship. The purpose of this study was twofold: to examine cultural competence among osteopathic medical students and to examine their attitudes toward …
Perception Of Control And Awareness Of Deficit In Individuals With Neurological Impairment In A Clinical Sample , Donna Marie Struck
Perception Of Control And Awareness Of Deficit In Individuals With Neurological Impairment In A Clinical Sample , Donna Marie Struck
PCOM Psychology Dissertations
Acquired brain injury is considered to be one of the most important public health concerns in the United States, with approximately 1.2 million adults that meet the criteria for chronic brain disease, brain disorder, or injury each year. Medical costs directly related to injury, in addition to the indirect costs related to job loss, totaled an estimated 60 billion dollars in the year 2000. Subsequent impairment following brain injury yields a complicated neurobehavioral sequelae of deficits. Anosognosia, specifically, the deficit of the patient's inability to acknowledge, or even recognize impairments, is common following brain injury. In a review of the …

Studies On Drug Solubilization Mechanism In Simple Micelle Systems, Shaoxin Feng
Studies On Drug Solubilization Mechanism In Simple Micelle Systems, Shaoxin Feng
University of Kentucky Doctoral Dissertations
Poor aqueous solubilities of drug candidates limit the biopharmaceutical usefulness in either oral or parenteral dosage forms. Lipid assemblies, such as micelles, may provide a means of enhancing solubility. Despite their usefulness, little is known about the means by which micelles accomplish this result. The goal of the current dissertation is to provide the molecular level understanding of the mechanism by which simple micelle systems solubilize drugs. Specifically, the location, orientation and amount of the drug molecules in micelle systems are the focuses of the work.
Three series of model drugs, steroids, benzodiazepines and parabens, in three surfactant systems with …
Early Heidegger's Transition From Life To Being, Gilbert Vasile Lepadatu
Early Heidegger's Transition From Life To Being, Gilbert Vasile Lepadatu
University of Kentucky Doctoral Dissertations
Heidegger was not always preoccupied, as he himself would later come to believe, with the question regarding the sense of being. Eight years before he published his magnum opus, Sein und Zeit, in 1927 he was totally devoted to finding a systematic way to bringing “life” as the ultimate source of meaning to explicate itself. In the years between 1919-1923, “life”, and not “being”, is the matter of philosophy par excellence, only to be disregarded, even refuted as a “proper” matter of philosophy in the subsequent years. In this paper I examine the philosophical motives that led Heidegger …

Analysis Of Security Measures For Sequences, Ramakanth Kavuluru
University of Kentucky Doctoral Dissertations
Stream ciphers are private key cryptosystems used for security in communication and data transmission systems. Because they are used to encrypt streams of data, it is necessary for stream ciphers to use primitives that are easy to implement and fast to operate. LFSRs and the recently invented FCSRs are two such primitives, which give rise to certain security measures for the cryptographic strength of sequences, which we refer to as complexity measures henceforth following the convention. The linear (resp. N-adic) complexity of a sequence is the length of the shortest LFSR (resp. FCSR) that can generate the sequence. Due …
Experimental And Molecular Dynamics Simulation Studies Of Partitioning And Transport Across Lipid Bilayer Membranes, Ravindra Wadhumal Tejwani
Experimental And Molecular Dynamics Simulation Studies Of Partitioning And Transport Across Lipid Bilayer Membranes, Ravindra Wadhumal Tejwani
University of Kentucky Doctoral Dissertations
Most drugs undergo passive transport during absorption and distribution in the body. It is desirable to predict passive permeation of future drug candidates in order to increase the productivity of the drug discovery process. Unlike drug-receptor interactions, there is no receptor map for passive permeability because the process of transport across the lipid bilayer involves multiple mechanisms. This work intends to increase the understanding of permeation of drug-like molecules through lipid bilayers.
Drug molecules in solution typically form various species due to ionization, complexation, etc. Therefore, species specific properties must be obtained to bridge the experiment and simulations. Due to …

Cultural cognition refers to the influence that individuals' values have on their perceptions of technological risk. We conducted a study to assess the cultural cognition of synthetic biology risks. Examining the attitudes of a large and diverse sample of Americans (N = 1,500), we found that hierarchical, conservative, and highly religious individuals - persons who normally are most skeptical of claims of environmental risks (including those relating to nuclear power and global warming) - are the persons most concerned about synthetic biology risks. We attribute this inversion of the normal cultural profile of risk perceptions to the seemingly anti-religious connotations …
Role Of Oxidative Stress And T Cell Homing In The Development Of Murine Syngeneic Graft-Versus-Host Disease, Jacqueline Perez-Rodriguez
Role Of Oxidative Stress And T Cell Homing In The Development Of Murine Syngeneic Graft-Versus-Host Disease, Jacqueline Perez-Rodriguez
University of Kentucky Doctoral Dissertations
Syngeneic graft-versus-host disease (SGVHD) is induced by reconstituting lethally irradiated mice with syngeneic BM cells followed by a 21 day treatment with the immunosuppressive agent cyclosporine A (CsA). Clinical symptoms of the disease appear 2-3 weeks following cessation of CsA therapy and disease-associated inflammation occurs primarily in the colon and liver.
The development of SGVHD is a complex process resulting from the cooperative interaction of multiple effector cell populations including NK cells, T cells and macrophages. TH1 cytokines (IL-12, TNF-α, IFN- γ), produced by these effector cells, serve as inflammatory mediators contributing to the pathogenesis of SGVHD. The …
Pathogenesis Of Influenza A Virus: Inhibition Of Monocyte Differetiation Into Dendritic Cell, Saikat Boliar
Pathogenesis Of Influenza A Virus: Inhibition Of Monocyte Differetiation Into Dendritic Cell, Saikat Boliar
University of Kentucky Doctoral Dissertations
Dendritic cells (DC) are a heterogeneous population of hematopoietic cells that play a versatile role in orchestrating immune responses against an array of invading pathogens, including influenza virus. These cells reside in lymphoid organs as well as in non-lymphoid tissues such as mucosal surfaces of respiratory and gastro-intestinal system. Recent investigations have suggested that in the steady state, dendritic cells are derived mainly from bone marrow precursor cells without a monocytic intermediate whereas during inflammation or infection, monocytes readily differentiate to generate monocyte derived dendritic cells (MoDC). The ability of virus infected monocytes to differentiate into MoDC was investigated and …
Embryo Exchanges And Adoption Tax Credits, Naomi R. Cahn, Sarah B. Lawsky
Embryo Exchanges And Adoption Tax Credits, Naomi R. Cahn, Sarah B. Lawsky
GW Law Faculty Publications & Other Works
The “Option of Adoption Act,” a Georgia law that was introduced by a staunchly anti-abortion Georgia state representative, establishes procedures for genetic donors to relinquish their rights to embryos before birth and permits, but does not require, embryo recipients to petition a court for recognition that they are the legal parents of a child born to them as a result of an embryo transfer. This article clears up what seems to be widespread confusion about a fairly straightforward question of tax law related to such embryo “adoptions.” Notwithstanding various sources' claims to the contrary, neither a Georgia adoption tax credit …

Science, Politics, Law And The Arc Of The Clean Water Act: The Role Of Assumptions In The Adoption Of A Pollution Control Landmark, Robert L. Glicksman, Matthew R. Batzel
GW Law Faculty Publications & Other Works
This article examines the assumptions upon which Congress relied in enacting the 1972 Clean Water Act and the extent to which they have been borne out or belied as the federal and state governments have implemented their statutory responsibilities in the quest to achieve acceptably clean water. It traces the development of federal water pollution control legislation before 1972, highlighting the deficiencies that contributed to the need for a new approach in 1972. It then examines the scientific and technical, political, and legal assumptions that helped shape the 1972 Clean Water Act in an effort to determine whether the failure …
Ecosystem Resilience To Disruptions Linked To Global Climate Change: An Adaptive Approach To Federal Land Management, Robert L. Glicksman
Ecosystem Resilience To Disruptions Linked To Global Climate Change: An Adaptive Approach To Federal Land Management, Robert L. Glicksman
GW Law Faculty Publications & Other Works
Global climate change presents daunting challenges to the federal government’s ability to manage its lands and resources in ways that ensure that the priceless natural heritage that these land and resources comprise remains available in substantially unimpaired condition to both present and future generations of Americans. One of the challenges results from the fact that the laws governing the activities of federal land management agencies have outlasted the scientific assumptions on which those laws were based. In particular, Congress adopted many of those laws on the assumption that ecological systems tend toward a natural equilibrium. Subsequently, the science of ecology …
Design, Synthesis, And Characterization Of Novel Hydrophilic Fluorene-Based Derivatives For Bioimaging Applications, Dao Nguyen
Electronic Theses and Dissertations
In this work, hydrophilic fluorene-based derivatives that contain ethylene oxide substituents, have been synthesized and characterized for potential use as new fluorophores for bioimaging applications and for fluorescence sensing of heavy metals. Symmetrical and unsymmetrical fluorene derivatives based on structural types of acceptor-pi-acceptor, acceptor-pi-donor, and donor-pi-donor were characterized by TGA, UV-vis absorption, fluorescence emission, lifetime, anisotropy, and two-photon absorption (2PA) cross section. They were found to possess high thermal stability, high photostability, high fluorescence quantum yields, and generally large two-photon absorption cross sections, making them quite suitable for new probes in single-photon absorption and two-photon absorption fluorescence microscopy imaging. Novel …
Modeling Microbiological And Chemical Processes In Municipal Solid Waste Bioreactor: Development And Applications Of A Three-Pha, Nitin Gawande
Modeling Microbiological And Chemical Processes In Municipal Solid Waste Bioreactor: Development And Applications Of A Three-Pha, Nitin Gawande
Electronic Theses and Dissertations
The numerical computer models that simulate municipal solid waste (MSW) bioreactor landfills have mainly two components--a biodegradation process module and a multi-phase flow module. The biodegradation model describes the chemical and microbiological processes of solid waste biodegradation. The models available to date include predefined solid waste biodegradation reactions and participating species. In a bioreactor landfill several processes, such as anaerobic and aerobic biodegradation, nitrogen and sulfate cycling, precipitation and dissolution of metals, and adsorption and gasification of various anthropogenic organic compounds, occur simultaneously. These processes may involve reactions of several species and the available biochemical models for solid waste biodegradation …

Money, Caregiving, And Kinship: Should Paid Caregivers Be Allowed To Obtain De Facto Parental Status, Pamela Loufer-Ukeles
Missouri Law Review
The law of custody and visitation is expanding to include the possibility of non-biological and non-adoptive parents' legal access to children. The concept of the psychological parent or functional caretaker is becoming increasingly prevalent and influential in state law. Moreover, the ALI Principles of Family Dissolution include two categories of psychological parents - parents by estoppel and de facto parents - in its proposed guidelines for who can petition for custody and visitation rights to children. Yet, both state law and the ALl Principles exclude caretakers who receive compensation - including foster parents, paid child care providers and surrogate mothers …
