Changes in [Ca2+]i response of individual Jurkat cells to nanosecond pulsed electric fields (nsPEFs) of 60 ns and field strengths of 25, 50, and 100 kV/cm were investigated. The magnitude of the nsPEF-induced rise in [Ca2+]i was dependent on the electric field strength. With 25 and 50 kV/cm, the [Ca2+]i response was due to the release of Ca2+ from intracellular stores and occurred in less than 18 ms. With 100 kV/cm, the increase in [Ca2+]i was due to both internal release and to influx across the plasma …

Compulsive Hoarding In Children: Six Case Studies, Edward H. Plimpton, Randy O. Frost, Brianna C. Abbey, Whitney Dorer
Psychology: Faculty Publications
Semi-structured interviews were conducted with the parents of six children who showed signs of compulsive hoarding. The cases revealed a wide variety of hoarding behaviors. Hoarding manifested in difficulties in discarding and maintaining control over possessions. Few cases had problems with clutter or excessive acquisition. In three cases hoarding was episodic and stress-related. Most of the cases showed overlapping ADHD symptoms and a majority demonstrated problems with perfectionism. Only two had clear obsessive-compulsive disorder, although concerns about others touching or moving possessions, which was present in all of the cases, may represent a form of ordering and arranging compulsions. None …

Modeling And Experiments In Microchannels For The Separation And Detection Of Bacteria Using Antibody-Coated Filter And Quantum Dots, Narendra Sharma
Open Access Dissertations
Capture and detection of biomarkers for organisms is an important aspect in the design of a Lab-on-a-Chip (LOC). In this research, modeling and experiments were conducted to separate, capture and detect E. coli K12 bacteria from water samples using microchannels. Microchannels were fabricated using the soft lithography technique. The bacteria were separated from the sample and captured at a polyester filter whose surface was modified by attaching functional groups that had affinity for antibodies specific to E. coli K12. The bacteria bound to the functionalized filter were then tagged with quantum dots conjugated with E. coli K12-specific antibodies for detection …
Dna Methylation Analysis And Identification Of A Novel Antisense Transcript In The Peg3 Imprinted Domain, Jennifer Marian Huang
Dna Methylation Analysis And Identification Of A Novel Antisense Transcript In The Peg3 Imprinted Domain, Jennifer Marian Huang
LSU Doctoral Dissertations
Genomic imprinting is a process that leads to the silencing of one allele of a gene in a parent of origin specific manner. Genes that are involved in this process are often regulated in clusters, one of which is the Peg3 (Paternally expressed gene 3) imprinted domain. We investigated this region for both CpG islands and long antisense transcripts, two common features of imprinted gene clusters. First, we performed a systematic survey of DNA methylation status of the CpG islands in this region of the mouse, cow, and human genomes. We identified two previously unreported differentially methylated regions (DMR): one …

Applying Scanning Probe Microscopy For The Investigation Of Molecular Self-Assembly Mechanisms And Properties Of Designed Nanomaterials, Algernon Tremayne Kelley
LSU Doctoral Dissertations
Scanning probe microscopy (SPM) for conducting surface characterizations of nanomaterials and molecular self-assembly processes is emerging as an important contribution in nanotechnology, especially towards the design of molecular electronic devices. Another area of importance is the characterization of the properties of nanomaterials for fundamental understanding of structure-function inter-relationships. Understanding the properties and behavior of molecules and finding approaches to control surface self-organization through nanolithography provides essential information for the development of workable applications for nanotechnology. This dissertation describes the methodologies of AFM for characterizing molecules and nanostructures produced with scanning probe lithography (SPL). Automated software for nanografting and nanoshaving produce …

Scanning Probe Microscopy Investigations Of (1) Arrays Of Cysteine-Coated Cds Nanoparticles And (2) Structures Formed During The Early Stages Of The Corrosion Of Copper Surfaces, Brian Robert Lewandowski
LSU Doctoral Dissertations
Scanning probe microscopy (SPM) characterizations are becoming more prevalent for surface investigations due to their capabilities for obtaining structural information and physical measurements. New capabilities of SPM for studying and controlling nanoscale processes are emerging as valuable assets in research. A fundamental understanding of the interactions of surface reactions provides essential information for developing workable applications for nanotechnology. Two applications of SPM are discussed in this dissertation. The first investigation uses atomic force microscopy (AFM) for the characterization of nanostructures produced with a newly developed lithographic technique called “two-particle” lithography. This new technique is based on particle lithography for the …

Characterization Of Porcine Adipose Tissue-Derived Stem Cells, Kellie J. Williams
Characterization Of Porcine Adipose Tissue-Derived Stem Cells, Kellie J. Williams
LSU Doctoral Dissertations
Nuclear transfer (NT) results in low success rates and research has reported problems with the welfare of cloned animals produced. Evidence suggests the use of a less differentiated donor cell, such as a stem cell, may lead to an increase in the efficiency of the NT procedure. Stem cell characteristics must be clearly defined in order to isolate, identify and optimally culture potential donor cells. Stem cell characteristics such as self-renewal and differentiation have not been identified in stem cells from porcine adipose tissue, an easily attainable tissue resource. The phenotypic profile of cell surface proteins and chromosomal stability of …
Particle-Chemical Interactions And Environmental Chemodynamics Of Fine And Ultrafine Particles In A Natural Disaster Scenario, Nicholas A. Ashley
Particle-Chemical Interactions And Environmental Chemodynamics Of Fine And Ultrafine Particles In A Natural Disaster Scenario, Nicholas A. Ashley
LSU Doctoral Dissertations
The interactions of fine and ultrafine particles with chemicals play a dominant role in determining the mobility and availability of pollutants in the environment. Fine particles in sediments can sequester chemicals from the water column, and release volatile and semi-volatile organic compounds to the gas phase upon exposure to air. Ultrafine particles which are photoreactive can degrade these vapor-phase contaminants, and may transform the molecules into species which are more toxic or hazardous than the parent. As the widespread, commercial use of ultrafine particles becomes more common, understanding the chemodynamics of these particles and their interactions with chemicals in the …
Factors That Influence College Students Who Choose Engineering As Their Major To Presist In That Major To Their Fifth Semester, Kelli Elaine Wildman
Factors That Influence College Students Who Choose Engineering As Their Major To Presist In That Major To Their Fifth Semester, Kelli Elaine Wildman
LSU Doctoral Dissertations
Individuals, institutions, and society are affected by whether or not students persist in college; therefore, persistence and retention on college campuses is an important topic for higher education systems. However, little research has been done on program retention. Since engineering as a profession is expected to increase, it is imperative to have students persist in the major. The primary purpose of this study is to determine the influence of selected demographic and academic characteristics in the decision of first time in college (FTIC) traditional-age undergraduates who declare engineering as their major at admission to persist in the curriculum from second …
A Multi-Function, Disposable, Microfluidic Module For Mutation Detection, Tae Yoon Lee
A Multi-Function, Disposable, Microfluidic Module For Mutation Detection, Tae Yoon Lee
LSU Doctoral Dissertations
Recognition of point mutations in a codon 12 of the K-ras gene, most frequently observed, is considered to be useful in the early diagnosis of several types of the human cancers. We have developed a multifunction, disposable, microfluidic module which detects low-abundant point mutations in human genomic DNA in modular architecture. Each functional component including a microfluidic PCR reactor, a passive diffusional micromixer reactor, and a microfluidic LDR reactor was separately designed and fabricated. Fluidic interconnects were also developed to make a fluidic passage between the functional components. Polycarbonate substrates were micro-molded, using hot embossing with micro-milled brass mold inserts …
The Effects Of Being Placed In Special Education Classes Versus General Education Classes And Teacher Certification On Students' High-Stakes Testing Scores, Bethany Ann Porter
The Effects Of Being Placed In Special Education Classes Versus General Education Classes And Teacher Certification On Students' High-Stakes Testing Scores, Bethany Ann Porter
LSU Doctoral Dissertations
The introduction of new laws such as the amendments to the Individuals with Disabilities Education Act (IDEA, 1997) and the No Child Left Behind Act have changed education for special education students in the United States. Students with disabilities are now more frequently being held to the same standards as those students without disabilities. These federal laws are designed to help close the achievement gap among all students regardless of race, gender, poverty, or disability status. Special education students are now required to participate in statewide high-stakes testing programs alongside their nondisabled peers. Another movement involves including students with disabilities …
Collpas As Activity Hotspots For Frugivorous Bats (Stenodermatinae) In The Peruvian Amazon: Underlying Mechanisms And Conservation Implications, Adriana Bravo Ordonez
Collpas As Activity Hotspots For Frugivorous Bats (Stenodermatinae) In The Peruvian Amazon: Underlying Mechanisms And Conservation Implications, Adriana Bravo Ordonez
LSU Doctoral Dissertations
In western Amazonia, large numbers of frugivorous bats regularly visit natural forest clearings known locally as collpas (also called clay licks or mineral licks). Bats arrive at collpas to drink water that has accumulated in depressions created by larger mammals that consume soil. Although collpa visitation by bats appears relatively common in western Amazonia, little is known about its causes and its ecological implications. In this dissertation I describe general and seasonal patterns of collpa visitation by frugivorous bats in the Peruvian Amazon, and I investigate potential explanations for this unique behavior. Regardless of season, collpas seem to be activity …
Atom-Based Computer Simulation Studies Of Gas-To-Liquid Nucleation In Atmospherically Relevant Systems: Clarifying Discrepancies And Elucidating Mechanisms, Ricky Bendanillo Nellas
Atom-Based Computer Simulation Studies Of Gas-To-Liquid Nucleation In Atmospherically Relevant Systems: Clarifying Discrepancies And Elucidating Mechanisms, Ricky Bendanillo Nellas
LSU Doctoral Dissertations
For over a century, nucleation for all systems was thought simplistically to be a process that advances through the formation of critical clusters with a well-defined composition. Our results show intriguing nucleation mechanisms that challenge the aforementioned notion. We employed the simple TraPPE-UA (transferable potential for phase equilibria – united atom) force field and the AVUS-HR approach (a combination of aggregation-volume-bias Monte Carlo, umbrella sampling, and histogram reweighting), to investigate the homogeneous vapor-to-liquid nucleation of various nucleating systems. We found out that these systems could nucleate through a variety of unique non-ideal mechanisms. Alongside existing experimental investigations, this dissertation presents …

Analysis Of Lesion Border Segmentation Using Watershed Algorithm, Hanzheng Wang
Masters Theses
"Automatic lesion segmentation is an important part of computer-based skin cancer detection. A watershed algorithm was introduced and tested on benign and melanoma images. The average of three dermatologists' manually drawn borders was compared as the benchmark. Hair removing, black border removing and vignette removing methods were introduced in preprocessing steps. A new lesion ratio estimate was added to the merging method, which was determined by the outer bounding box ratio. In postprocessing, small blob removing and border smoothing using a peninsula removing method as well as a second order B-Spline smoothing method were included. A novel threshold was developed …

A Simple Algorithm For Triconnectivity Of A Multigraph, Abusayeed Saifullah, Alper Ungor
A Simple Algorithm For Triconnectivity Of A Multigraph, Abusayeed Saifullah, Alper Ungor
All Computer Science and Engineering Research
Vertex-connectivity and edge-connectivity represent the extent to which a graph is connected. Study of these key properties of graphs plays an important role in varieties of computer science applications. Recent years have witnessed a number of linear time 3-edge-connectivity algorithms - with increasing simplicity. In contrast, the state-of-the-art algorithm for 3-vertex-connectivity due to Hopcroft and Tarjan lacks the simplicity in the sense of ease of implementation as well as the number of passes over the graph although its time and space complexity is theoretically linear. In this paper, we propose a linear time reduction from 3-vertex-connectivity to 3-edge- connectivity of …

Flap Endonuclease 1 Ensures Telomere Replication And Stability, Abhishek Saharia
Flap Endonuclease 1 Ensures Telomere Replication And Stability, Abhishek Saharia
All Theses and Dissertations (ETDs)
Telomeres, protein-DNA structures that distinguish the end of a chromosome from a bona fide DNA double strand break, are integral to genomic stability. High fidelity replication of telomeres is indispensable for their stability. Telomere replication is a challenging task that is completed through the coordinated actions of telomere binding proteins and DNA replication and repair proteins in ways that are not well understood. This work focuses on delineating the function of one DNA replication and repair protein, Flap Endonulcease 1: FEN1), in telomere replication and maintenance. I demonstrate that FEN1 is essential for the efficient replication of telomeres through its …
The Mechanisms And Roles Of Feedback Loops For Visual Processing, Jing Shao
The Mechanisms And Roles Of Feedback Loops For Visual Processing, Jing Shao
All Theses and Dissertations (ETDs)
Signal flow in the brain is not unidirectional; feedback represents a key element in neural signal processing. To address the question on how do neural feedback loops work in terms of synapses, microcircuitry, and systems dynamics, we developed a chick midbrain slice preparation to study and characterize one important feedback loop within the avian visual system: isthmotectal feedbackloop. The isthmotectal feedback loop consists of the optic tectum: OT) and three nucleus isthmi: Imc, Ipc and SLu. The tectal layer 10 neurons project to ipsilateral Imc, Ipc and SLu in a topographic way. In turn Ipc and SLu send back topographical: …
Targeting The Dimerization Of Erbb Receptor Tyrosine Kinases, Rob Yang
Targeting The Dimerization Of Erbb Receptor Tyrosine Kinases, Rob Yang
All Theses and Dissertations (ETDs)
The epidermal growth factor receptor: EGFR) is a membrane receptor tyrosine kinase whose over-activation has been implicated to cause many human cancers. Novel strategies to inhibit the activation of EGF receptors other than the conventional antibody-based and tyrosine kinase inhibitors are virtually non-existent but could provide benefits both in the laboratory and clinical settings. In an effort to expand the current approaches, this thesis focused on targeting the homodimerization of the EGF receptors themselves and the heterodimerization of EGF receptors with the related ErbB2 receptor. Three sub-projects were completed in the process. The first project explored the feasibility of inhibiting …
