Expression And Function Of Hypoxia Inducible Factor-1 Alpha In Human Melanoma Under Non-Hypoxic Conditions, Caroline N. Mills, Sandeep S. Joshi, Richard M. Niles

Background

Hypoxia inducible factor-1 alpha (HIF-1α) protein is rapidly degraded under normoxic conditions. When oxygen tensions fall HIF-1α protein stabilizes and transactivates genes involved in adaptation to hypoxic conditions. We have examined the normoxic expression of HIF-1α RNA and protein in normal human melanocytes and a series of human melanoma cell lines isolated from radial growth phase (RGP), vertical growth phase (VGP) and metastatic (MET) melanomas.

Results

HIF-1α mRNA and protein was increased in RGP vs melanocytes, VGP vs RGP and MET vs VGP melanoma cell lines. We also detected expression of a HIF-1α mRNA splice variant that lacks part …

The invention provides Bean pod mottle virus (BPMV) vectors useful for expression of heterologous proteins in plants such as soybean. The BPMV vectors are also useful for virus-induced gene silencing. The invention also provides methods for expressing a heterologous polypeptide in a plant such as soybean. The invention additionally provides methods for virus-induced gene silencing, particularly in a soybean plant, which can be used to determine the function of a gene of interest.

Genome databases continue to expand with no change in the basic format of sequence data. The prevalent use of the Classic alignment based search tools like BLAST have significantly pushed the limits of Genome Isolate research. The relatively new frontier of Metagenomic research deals with thousands of diverse genomes with newer demands beyond the current homologue search and analysis. Compressing sequence data into a complex form could facilitate a broader range of sequence analyses. Zooplankton were sampled on a monthly basis at the Bermuda Atlantic Time-series Study (BATS) site from January 1995 to December 1999. Samples were collected using a 1-m², 200 µm mesh net. The net sampled the water column in an oblique manner from the surface to a mean depth of 200 m. One day and one night tow from each cruise was examined microscopically to determine the community structure of the non-calanoid copepods. In addition, a three year set of nighttime samples were examined taken by 0.5-m², 20 and 35 µm mesh nets (1995-1996 and 1997, respectively) towed obliquely to 150 …

In this dissertation a methodology is proposed for simultaneously evaluating the population bioequivalence (PBE) of a generic drug to a pre-licensed drug, or the bioequivalence of two formulations of a drug using multiple correlated pharmacokinetic metrics. The univariate criterion that is accepted by the food and drug administration (FDA) for testing population bioequivalence is generalized. Very few approaches for testing multivariate extensions of PBE have appeared in the literature. One method uses the trace of the covariance matrix as a measure of total variability, and another uses a pooled variance instead of the reference variance. The former ignores the correlation …

This paper looks at the emerging conflict between natural philosophy and empirical science in the late sixteenth century by examining the events surrounding the supposed discovery of gold in northern North America by Martin Frobisher in 1577. The discovery of gold in a region thought incapable of producing the metal, and the subsequent assays of ore mined from that region served as a catalyst for conflict between different understandings of the natural world. Proponents of natural philosophy and empirical science each used their theoretical tools to prove or disprove the value of the ore, reflecting the larger discussions taking place …

This study explores the relationship between living arrangement and psychological wellbeing in retired elderly individuals living in Yancheng, Jiangsu (PR China). Data on mode of residence, socio-economic background, daily activities, and intergenerational dynamics were collected from 200 subjects, and their potential correlations with depression (assessed via the Geriatric Depression Scale Short Version) were analyzed. Univariate as well as logistic regression confirmed mode of residence as a significant predictor of depression in this group. Oyster tongers are a cultural icon of Florida’s maritime heritage and geography. Challenged for generations by the vagaries of weather, including catastrophic storms and years-long droughts, and economic uncertainties this maritime heritage is fading fast. While Florida’s north and west coasts produce 90 percent of the Florida oyster harvest and ten percent of oysters consumed in the United States, the industry is at risk today for reasons including a declining demand for Florida oysters because of health concerns; water pollution; population growth and its accompanying development of condominiums, gated communities, and retail shopping centers; and declining interest in the hard …

DNA damage is a common hazard that all cells have to combat. Saccharomyces cerevisiae HMO2 is a high mobility group protein (HMGB) that is a component of the chromatin-remodeling complex INO80, which is involved in double strand break (DSB) repair. We show here using DNA end-joining and exonuclease protection assays that HMO2 binds preferentially to DNA ends. While HMO2 binds DNA with both blunt and cohesive ends, the sequence of a single stranded overhang significantly affects binding, supporting the conclusion that HMO2 recognizes features at DNA ends. We present various methods to record cardiac function in the larval Drosophila. The approaches allow heart rate to be measured in unrestrained and restrained whole larvae For direct control of the environment around the heart another approach utilizes the dissected larvae and removal of the internal organs in order to bathe the heart in desired compounds. The exposed heart also allows membrane potentials to be monitored which can give insight of the ionic currents generated by the myocytes and for electrical conduction along the heart tube. These approaches have various advantages and disadvantages for future experiments that are discussed. …

Background: Recombination rates vary widely across the human genome, but little of that variation is correlated with known DNA sequence features. The genome contains more than one million Alu mobile element insertions, and these insertions have been implicated in non-homologous recombination, modulation of DNA methylation, and transcriptional regulation. If individual Alu insertions have even modest effects on local recombination rates, they could collectively have a significant impact on the pattern of linkage disequilibrium in the human genome and on the evolution of the Alu family itself.
