Molecular Diagnostic For Boll Weevil (Coleoptera: Curculionidae) Based On Amplification Of Three Species-Specific Microsatellites, Kyung Seok Kim, Zofia Szendrei, Cesar Rodriguez-Saona, Phillip G. Mulder Jr., Thomas W. Sappington Jan 2009

Molecular Diagnostic For Boll Weevil (Coleoptera: Curculionidae) Based On Amplification Of Three Species-Specific Microsatellites, Kyung Seok Kim, Zofia Szendrei, Cesar Rodriguez-Saona, Phillip G. Mulder Jr., Thomas W. Sappington

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

The boll weevil, Anthonomus grandis grandis Boheman (Coleoptera: Curculionidae), is a serious pest of cultivated cotton, Gossypium hirsutum L., in the Americas, and reinfestation of zones from which they have been eradicated is of perpetual concern. Extensive arrays of pheromone traps monitor for reintroductions, but occasionally the traps collect nontarget weevils that can be misidentified by scouts. For example, the congeneric pepper weevil, Anthonomus eugenii Cano, and other superficially similar weevils are attracted to components of the boll weevil lure or trap color. Although morphologically distinguishable by trained personnel, the potential for misidentification is compounded when captured weevils are dismembered …

A Novel Codominant Marker For Selection Of The Null Wx-B1 Allele In Wheat Breeding Programs, Mika Saito, Patricia Vrinten, Goro Ishikawa, Robert A. Graybosch, Toshiki Nakamura Jan 2009

A Novel Codominant Marker For Selection Of The Null Wx-B1 Allele In Wheat Breeding Programs, Mika Saito, Patricia Vrinten, Goro Ishikawa, Robert A. Graybosch, Toshiki Nakamura

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Waxy protein (granule-bound starch synthase I) is a key enzyme in the synthesis of amylose in endosperm tissue. The amylose content of wheat flour plays a significant role in determining Japanese udon noodle quality. Most wheat cultivars suitable for producing udon noodles have a low amylose level due to a lack of Wx-B1 protein conditioned by null Wx-B1 alleles. It was previously determined that the entire coding region of the wheat Wx-B1 gene is deleted in the most common null allele. However, the extent and breakpoints of the deletion have not been established. In this study, the position of the …

Effects Of Spontaneous Heating On Fiber Composition, Fiber Digestibility, And In Situ Disappearance Kinetics Of Neutral Detergent Fiber For Alfalfa-Orchardgrass Hays, W. K. Coblentz, P. C. Hoffman Jan 2009

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

During 2006 and 2007, forages from 3 individual hay harvests were utilized to assess the effects of spontaneous heating on concentrations of fiber components, 48-h neutral detergent fiber (NDF) digestibility (NDFD), and in situ disappearance kinetics of NDF for large-round bales of mixed alfalfa (Medicago sativa L.) and orchardgrass (Dactylis glomerata L.). Over the 3 harvests, 96 large-round bales were made at preset bale diameters of 0.9, 1.2, or 1.5 m, and at moisture concentrations ranging from 9.3 to 46.6%. Internal bale temperatures were monitored daily during an outdoor storage period, reaching maxima (MAX) of 77.2°C and 1,997 heating degree …

Expression And Functional Characterization Of The Plant Antimicrobial Snakin-1 And Defensin Recombinant Proteins, Natalia Kovalskaya, Rosemarie W. Hammond Jan 2009

Expression And Functional Characterization Of The Plant Antimicrobial Snakin-1 And Defensin Recombinant Proteins, Natalia Kovalskaya, Rosemarie W. Hammond

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

In this study, for the first time, functionally active, recombinant, cysteine-rich plant proteins snakin-1 (SN1) and defensin (PTH1) were expressed and purified using a prokaryotic expression system. The overall level of antimicrobial activities of SN1 and PTH1 produced in Escherichia coli was commensurate with that of the same proteins previously obtained from plant tissues. Both proteins exhibited strong antibacterial activity against the phytopathogenic bacterium Clavibacter michiganensis subsp. sepedonicus (50% inhibitory concentration (IC50) 1.5–8 lM) and antifungal activity against the phytopathogenic fungi Colletotrichum coccoides and Botrytis cinerea (IC50 5–14 lM). Significantly weaker activity was observed against Pseudomonas syringae …

Efficacy And Safety Of Catnip (Nepeta Cataria) As A Novel Filth Fly Repellent, J. J. Zhu, X.-P. Zeng, D. Berkebile, H.-J. Du, Y. Tong, K. Qian Jan 2009

Efficacy And Safety Of Catnip (Nepeta Cataria) As A Novel Filth Fly Repellent, J. J. Zhu, X.-P. Zeng, D. Berkebile, H.-J. Du, Y. Tong, K. Qian

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Catnip (Nepeta cataria) is known for its pseudo-narcotic effects on cats. Recently, it has been reported as an effective mosquito repellent against several Aedes and Culex species, both topically and spatially. Our laboratory bioassays showed that catnip essential oil (at a dosage of 20 mg) resulted in average repellency rates of 96% against stable flies, Stomoxys calcitrans (L.) and 79% against houseflies, Musca domestica (L.), respectively. This finding suggested that the application of repellent could be used as part of filth fly management. Further evaluations of catnip oil toxicity were conducted to provide a broad-spectrum safety profile of …

Dna Delivery From Matrix Metalloproteinase Degradable Poly (Ethylene Glycol) Hydrogels To Mouse Cloned Mesenchymal Stem Cells, Yuguo Lei, Tatiana Segura

Department of Chemical and Biomolecular Engineering: Faculty Publications

The ability to genetically modify mesenchymal stem cells (MSCs) seeded inside synthetic hydrogel scaffolds would offer an alternative approach to guide MSC differentiation and to study molecular pathways in three dimensions than protein delivery. In this report, we explored gene transfer to infiltrating MSCs into matrix metalloproteinase (MMP) degradable hydrogels that were loaded with DNA/poly(ethylene imine) (PEI) polyplexes. DNA/PEI polyplexes were encapsulated inside poly (ethylene glycol) (PEG) hydrogels crosslinked with MMP degradable peptides via Michael Addition chemistry. A large fraction of encapsulated polyplexes remained active after encapsulation (65%) and the mechanical properties of the hydrogels were unchanged by the encapsulation …

Reputation Versus Reality: The Impact Of Us News And World Report Rankings And Education Branding On Hiring Decisions In The Job Market, Fausto D. Capobianco Jan 2009

Reputation Versus Reality: The Impact Of Us News And World Report Rankings And Education Branding On Hiring Decisions In The Job Market, Fausto D. Capobianco

Theses and Dissertations

Typically, leading brands provide benchmarks for constructing consumer preference in the marketplace. Reputation rankings have sustained and advanced the status of brand names in higher education with an implication that the degrees awarded by higher ranked schools have added prestige, a cachet with the potential of facilitating success in the job market. This implication makes reputation rankings a dependable tool for college and university marketing departments eager to increase student enrollment and retention by communicating its superiority among its peers. By examining the influence of reputation rankings on the pre-decision preferences of human resource hiring professionals in evaluating employment applicants, …

Diversity Congruency Within Organizations: The Relationship Among Emotional Intelligence, Personality Structure, Ethnic Identity, Organizational Context And Perceptions Of Organizational Diversity, David Hurlic Jan 2009

Diversity Congruency Within Organizations: The Relationship Among Emotional Intelligence, Personality Structure, Ethnic Identity, Organizational Context And Perceptions Of Organizational Diversity, David Hurlic

Theses and Dissertations

This study explores the relationship among Emotional Intelligence (EI) (consisting of perception and appraisal, facilitating thinking, understanding emotions, and regulating emotions), Personality Structure (FFM) (consisting of extroversion, agreeableness, conscientiousness, neuroticism, and openness to experiences), Ethnic Identity, Organizational Context (OC), and Perceptions of Organizational Diversity. It is hoped that by examining the impact of individual personality structures, as well as, ethnic identity and organizational context as precursors to individual perceptions between EI, Personality Structure, and diversity and affirmative action, a more coherent method of analysis is created allowing organizations to identify specific factors contributing to affirmative action and diversity program acceptance …

Success Strategies Of High Achieving Women: A Qualitative Phenomenological Investigation, Leslie Ann Evans Jan 2009

Success Strategies Of High Achieving Women: A Qualitative Phenomenological Investigation, Leslie Ann Evans

Theses and Dissertations

According to the latest figures from the U.S. Labor Department, 50% of all managerial posts are held by women, yet few women hold a top position in their organization. Of the 1,000 largest revenue producing, publicly traded companies in the United States, only 20 are led by a woman. Women hold only 15.7% of all senior executive positions in Fortune 500 companies, yet traditional women's leadership styles of open communication and inclusion have been recognized as appropriate and even best practices. Women climbing the corporate ladder face myriad challenges. In addition to the obstacles they face, the very choices women …

The Leaders Of Bushido: A Study Of The Leadership Practices Of Black Belt Martial Artists, Maya Joko

Theses and Dissertations

Martial arts is an all-encompassing discipline in which the teacher serves as a role model, guide, and leader. Black belt martial artists, especially the ones that teach, have a strong influence on the younger generation who admire and respect their teachers. In this way, martial artists are leaders who have the power to positively influence future leaders of our generation. Despite this fact, there is limited research on how martial arts training might contribute to the development of positive and effective leadership skills. The purpose of this quantitative study was to determine if there were similarities and/or differences in the …

Socially Responsible Advertising In The Fashion And Cosmetic Industry : The Female Consumers’ Perspective, Christine Murphy Jan 2009

Socially Responsible Advertising In The Fashion And Cosmetic Industry : The Female Consumers’ Perspective, Christine Murphy

Theses

This study explores the portrayal of women in advertising in the fashion and cosmetic industry. While traditionally, companies have employed the use of highly attractive thin women known as the thin ideal in their advertisements, there have been some moves in the industry towards the use of women of various ages and sizes in an attempt to celebrate female beauty as a whole instead of focusing on the thin ideal. This has been in response to criticisms of the industry as dominant contributors to high levels of negative body images, dissatisfaction, extreme dieting and eating disorders among girls and women. …
