At the time of fertilization, a dramatic change occurs in the oocyte that transforms this cell from a metaphase arrested state into a metabolically active and dynamic state. The view of the flow of biological processes within organisms has recently shifted from that of a linear path to a more complex network. Biological processes are no longer thought of in the simple terms of DNA to RNA, RNA to proteins, and proteins to final activity. This paper advances a capabilities conception of the individual, and considers some of the problems involved in developing such a conception. It also makes claims about the nature of the capability space as a whole, frames personal development in terms of the idea of moving though the capability space, and argues that people are alike in being increasingly heterogeneous. A key problem for a capabilities conception of the individual is that some capabilities, such as belonging to social groups and having social identities, can undermine individuality. The online harassment of women exemplifies twenty-first century behavior that profoundly harms women yet too often remains overlooked and even trivialized. This harassment includes rape threats, doctored photographs portraying women being strangled, postings of women's home addresses alongside suggestions that they are interested in anonymous sex, and technological attacks that shut down blogs and websites. It impedes women's full participation in online life, often driving them offline, and undermines their autonomy, identity, dignity, and well-being. The philosophical concept of analogy is fundamental to the theory of imaging that characterizes Plato’s metaphysics, cosmology, and methodology. While Plato never explicitly conceptualizes the philosophical role of analogy, his dialogues are rife with analogies and images that are often pivotal to the thought expressed there. An analysis of celebrated analogies such as the sun and the good in the Republic, the “second sailing” in the Phaedo, the “receptacle” (chōra) in the Timaeus, and the example of weaving in the Statesman reveals that even if the theory each elucidates is not explicitly concerned with analogy, the theories these images enable …

Type IV secretion systems (T4SS) translocate DNA and protein substrates across prokaryotic cell envelopes generally by a mechanism requiring direct contact with a target cell. Three types of T4SS have been described: (i) conjugation systems, operationally defined as machines that translocate DNA substrates intercellularly by a contact-dependent process; (ii) effector translocator systems, functioning to deliver proteins or other macromolecules to eukaryotic target cells; and (iii) DNA release/uptake systems, which translocate DNA to or from the extracellular milieu. Marine dissolved organic matter (DOM) comprises one of the largest carbon reservoirs on earth and has long been considered a potential energy source for marine invertebrates. The importance of DOM transport has been adequately demonstrated for unicellular organisms, where DOM can meet 100% of an organisms energy needs, but the effects of DOM uptake for marine metazoans are less well understood. In this study, three general areas involving the influence of DOM transport to marine invertebrates were explored. The arroyo toad (Anaxyrus californicus) is a federally endangered species found on Fort Hunter Liggett, Monterey County, California. The species was discovered in 1996 and was determined to occupy 26.7 km of the San Antonio River from approximately 2.4 km northwest of the San Antonio Mission de Padua, to the river delta above the San Antonio Reservoir. The construction of the San Antonio Reservoir dam in 1963 isolated this northern population of arroyo toads. Through time, the Fort Hunter Liggett landscape has changed drastically. The land was heavily grazed by cattle until 1991, which considerably reduced vegetation in riparian areas. …

The onset of osteoporosis caused by aging, disease states, and post-menopausal conditions significantly impacts patient quality of life, required healthcare funding, personal autonomy losses from increased fracture risk and the subsequent corrective surgery. Research has indicated that osteocyte apoptosis may be a key parameter in bone remodeling, raising the possibility of remodeling rate modulation for the mitigation of bone mass resorption. This study investigated the development potential of equine embryos in vitro after biopsy and vitrification. Twenty embryos were obtained from Quarter Horse, Thoroughbred, and mix-breed light mares between three and ten years old. The twenty embryos were divided into a biopsy (n=10) and control group (n=10). The biopsy group underwent microaspiration biopsy using a micromanipulator to obtain a small tissue sample from the embryo. Both groups were then vitrified using a commercially available technique originally described by Carnevale (2006) at Colorado State \ University.

This paper covers the portrayals of Captain James Cook within Hawaiian education. It begins by providing a backdrop to early European portrayals of Captain Cook and then proceeds to investigate how these portrayals changed as they were incorporated into Hawaiian textbooks. The paper then continues to illustrate the changes made in Cook’s portrayal within Hawaiian textbooks and how these changes coincide with the prevailing interests of the eras in which the authors wrote them.

Although rice straw and other grains have been used in building since pre-history, in the past two decades, there has been a move to utilize this rapidly renewable, locally available, agricultural byproduct as part of the sustainable construction movement. Up to this point, this has been done by simply stacking up the full straw bales. Stak Block, invented by Oryzatech, Inc., is a modular, interlocking block made of a composite of rice straw and binding agent that serves as an evolution in straw construction. Outward Bound and the National Outdoor Leadership School (NOLS) are two of the United State’s largest providers of outdoor education and adventure recreation programs. While many studies have examined the outcomes of the different organizations individually, there is very little comparative research. This study compared the attributes, consequences, and values obtained by 510 participants of courses 14 days or longer in the Rocky Mountain region during the summer of 2006.

In her article "Russian Magical Realism and Pelevin as Its Exponent" Alexandra Berlina seeks to enrich magical realism studies as a field of comparative literature and culture by showing that, although largely ignored in scholarship, such a thing as Russian magical realism exists and provides an interesting field for analysis. Matrix extracellular phosphoglycoprotein/osteoblast factor 45 (MEPE/OF45) was cloned in 2000 with functions related to bone metabolism. We identified MEPE/OF45 for the first time as a new co-factor of CHK1 in mammalian cells to protect cells from DNA damage induced killing. We demonstrate here that MEPE/OF45 directly interacts with CHK1. Knocking down MEPE/OF45 decreases CHK1 levels and sensitizes the cells to DNA damage inducers such as ionizing radiation (IR) or camptothicin (CPT)-induced killing. Over-expressing wild-type MEPE/OF45, but not the mutant MEPE/OF45 (depleted the key domain to interact with CHK1) increases CHK1 levels in the cells and increases the resistance of the …

The amyloid-beta 42 (Abeta42) is thought to play a central role in the pathogenesis of Alzheimer's disease (AD). However, the molecular mechanisms by which Abeta42 induces neuronal dysfunction and degeneration remain elusive. Mitochondrial dysfunctions are implicated in AD brains. Whether mitochondrial dysfunctions are merely a consequence of AD pathology, or are early seminal events in AD pathogenesis remains to be determined. Here, we show that Abeta42 induces mitochondrial mislocalization, which contributes to Abeta42-induced neuronal dysfunction in a transgenic Drosophila model. The cAMP-responsive transcription factor CREB functions in adipose tissue and liver to regulate glycogen and lipid metabolism in mammals. While Drosophila has a homolog of mammalian CREB, dCREB2, its role in energy metabolism is not fully understood. Using tissue-specific expression of a dominant-negative form of CREB (DN-CREB), we have examined the effect of blocking CREB activity in neurons and in the fat body, the primary energy storage depot with functions of adipose tissue and the liver in flies, on energy balance, stress resistance and feeding behavior. Since 2000, the PARIMA project has implemented risk-management activities among semi-settled pastoralists in southern Ethiopia. The goal has been to improve human welfare via collective action and capacity building. Outcomes include progress in income generation, asset conservation, and livelihood diversification. Fifty-nine collective-action groups were created. Dominated by women, they included over 2,000 founding members and groups have recently merged to form 37 cooperatives, consistent with government policy. Creating sustainable impacts via collective action and capacity building requires many inputs. Taking raw, illiterate volunteers and transforming them into sustainable groups took up to three years, on average. Costs of implementing this …

As one of the founding members of the General Agreement on Tariffs and Trade (GATT), Taiwan (the Republic of China or ROC) - the 17th largest economy, was granted accession to the World Trade Organization (WTO) in November 2001 after its observer status of eleven years. Taiwan, classified by most commentators as an "unrecognized state" or an "entity sui generis", has been excluded from most of the major international organizations. Taiwan's accession to the WTO, therefore, is considered to be an important breakthrough in diplomacy for the past decades. Knowledge-based firms seeking competitive advantage often draw on the public knowledge stream (ideas embedded in public commons institutions) as the foundation for private knowledge (ideas firms protect through private intellectual property [IP] institutions). However, understanding of the converse relationship—the impact of private knowledge strategies on public knowledge production—is limited. We examine this question in human genetics, where policy makers debate expanding IP ownership over the human genome. Our difference-in-differences estimates show that gene patents decrease public genetic knowledge, with broader patent scope, private sector ownership, patent thickets, fragmented patent ownership, and a gene's commercial relevance exacerbating their effect.

Tuberculosis remains a major threat as drug resistance continues to increase. Pulmonary tuberculosis in adults is responsible for 80% of clinical cases and nearly 100% of transmission of infection. Unfortunately, since we have no animal models of adult type pulmonary tuberculosis, the most important type of disease remains largely out of reach of modern science and many fundamental questions remain unanswered. This paper reviews research dating back to the 1950's providing compelling evidence that cord factor (trehalose 6,6 dimycolate [TDM]) is essential for understanding tuberculosis. The study of job satisfaction for professors in the work place has been prevalent in the research for decades (COACHE, 2007). As online education grows exponentially each year (Allen & Seaman, 2006), this aspect of professorial teaching is emerging as an increasingly critical factor.
