Blood Chimerism Confounds Genetic Relative Susceptibility Testing For Classical Scrapie In Sheep, David A. Schneider, Ahmed Tibary, Terje Taudsepp, Pranab J. Das, Katherine I. O'Rourke Jan 2009

Blood Chimerism Confounds Genetic Relative Susceptibility Testing For Classical Scrapie In Sheep, David A. Schneider, Ahmed Tibary, Terje Taudsepp, Pranab J. Das, Katherine I. O'Rourke

Zoonotics and Wildlife Disease: Publications

Classical scrapie disease is a transmissible spongiform encephalopathy of sheep that is enzootic in the United States. Susceptibility of sheep to classical scrapie is linked to single nucleotide polymorphisms in the prion protein gene (PRNP), forming the basis for genetic testing strategies used by national efforts to eradicate scrapie. Such efforts are occasionally hampered by inconclusive results stemming from the detection of ‘‘complex’’ genotypes. Naturally occurring cases of ovine chimerism are thought to account for some of these instances. In the current report, 4 naturally occurring ovine chimeras are documented through cytogenetic and molecular analyses. All 4 of these sheep …

Nor98 Scrapie Identified In The United States, Christie M. Loiacono, Bruce V. Thomsen, S. Mark Hall, Matti Kiupel, Diane Sutton, Katherine I. O'Rourke, Bradd Barr, Lucy Anthenill, Delwyn Keane

Zoonotics and Wildlife Disease: Publications

A distinct strain of scrapie identified in sheep of Norway in 1998 has since been identified in numerous countries throughout Europe. The disease is known as Nor98 or Nor98-like scrapie, among other names. Distinctions between classic scrapie and Nor98 scrapie are made based on histopathology and immunodiagnostic results. There are also differences in the epidemiology, typical signalment, and likelihood of clinical signs being observed. In addition, sheep that have genotypes associated with resistance to classic scrapie are not spared from Nor98 disease. The various differences between classic and Nor98 scrapie have been consistently reported in the vast majority of cases …

Reinvention In The Line Of Death: A Reconsideration Of Geoffrey Hill's Commemorative Verse, Michael Christopher Bartch Jan 2009

Reinvention In The Line Of Death: A Reconsideration Of Geoffrey Hill's Commemorative Verse, Michael Christopher Bartch

Graduate Student Theses, Dissertations, & Professional Papers

This paper considers the embodied ethics of Geoffrey Hill’s poetic practice. Hill stages his engagement with poetry through the idioms, images, tropes, and diction of the literary tradition. Through this pragmatic rehearsal of the language of the dead, Hill’s poetry projects the tradition into the present. Hill resists the ethical entrapments of appropriative poetry through his insistence upon the brute physicality of atrocity and through a rigorous (for both poet and reader) formal difficulty. Hill’s practice refuses to console after the models of Peter Sacks, Jahan Ramazani, or John Vickery. Instead, concerned with modernity’s disconnectedness, Hill’s poetry returns us to …

The Role Of The Cb1 Receptor In Learning, Memory And Anxiety-Like Behaviors, Loretta Lynn Bolyard Jan 2009

The Role Of The Cb1 Receptor In Learning, Memory And Anxiety-Like Behaviors, Loretta Lynn Bolyard

Graduate Student Theses, Dissertations, & Professional Papers

Several lines of evidence support a role of the endocannabinoid (eCB) system in cognition and anxiety. This study explores cognitive processes and anxiety-like behaviors in wild type (CB1+/+) and CB1-receptor-deficient (CB1-/-) mice of differing ages. Differences were observed between CB1+/+ and CB1-/- mice in a Morris Water Maze acquisition task. Furthermore, CB1-/- mice did not display deficits in extinction during reversal learning. In the Light-Dark Box and Suok tasks, the CB1-/- mice demonstrated greater anxiety-like behaviors relative the CB1+/+ mice. No differences were observed in the Open-Field task, suggesting that the observed behavioral differences may be related to anxiety rather …

Molecular Characterization Of Gastroenteritis Viruses Among Equines And Porcines In Southern Ireland From 2005-2007., P. J, Collins

Theses

Rotaviruses are a major cause of gastroenteritis in humans and animals. To date, seven serogroups (A to G) of rotavirus have been determined. Group A Rotaviruses (GARVs) are enteric pathogens, causing acute watery dehydrating diarrhea in various host species, including birds and mammals. Rotaviruses account for ~611,000 child deaths each year, mainly in developing countries. Likewise, rotavirus-associated enteritis is a major problem in young calves, weaning and post-weaning piglets and foals. Vaccines against the most important serologic group of rotaviruses (GARVs) are available for the prevention of rotavirus diseases in cows and horses, and, more recently, they have been made …

Molecular Dynamics Study Of Single Stranded Peptide Nucleic Acids, Anna K. Manukyan Jan 2009

Molecular Dynamics Study Of Single Stranded Peptide Nucleic Acids, Anna K. Manukyan

Theses and Dissertations in Biomedical Sciences

A PNA molecule is a DNA strand where the sugar-phosphate backbone has been replaced by a structurally homomorphous pseudopeptide chain consisting of N(2-aminoethyl)-glycine units. PNA binds strongly to both DNA and RNA. However, an analysis of the X-ray and NMR data show that the dihedral angles of PNA/DNA or PNA/DNA complexes are very different from those of DNA:DNA or RNA:RNA complexes. In addition, the PNA strand is very flexible. One way to improve the binding affinity of PNA for DNA/RNA is to design a more pre-organized PNA structure. An effective way to rigidify the PNA strand is to introduce …

Detection Of Free-Ranging West Indian Manatees Trichechus Manatus Using Side-Scan Sonar, Daniel Gonzalez-Socoloske, Leon David Olivera-Gomez, Robert E. Ford Jan 2009

Detection Of Free-Ranging West Indian Manatees Trichechus Manatus Using Side-Scan Sonar, Daniel Gonzalez-Socoloske, Leon David Olivera-Gomez, Robert E. Ford

Faculty Publications

Turbid and tannin-stained waterways are difficult habitats to study and present problems with respect to the management of aquatic fauna, and especially of endangered and cryptic animals such as manatees Trichechus spp. Linnaeus. Throughout much of the range of the extant trichechid species (T. inunguis, T. manatus, and T. senegalensis), the scientific approaches that are used successfully to document the status of the Florida subspecies (T. m. latirostris) are not feasible, due either to constraints associated with habitat or to the high costs involved. We examined the use of side-scan sonar as a novel way to acoustically detect free-ranging West …

Impacts Of Alterations Of Organic Inputs On The Bacterial Community Within The Sediments Of Wind Cave, South Dakota, Usa, Marisa K. Chelius, Guy Beresford, Howard Horton, Meghan Quirk, Greg Selby, Rodney T. Simpson, Rodney Horrocks, John C. Moore

International Journal of Speleology

Wind Cave (WICA) in the Black Hills of South Dakota, like many mostly dry caves in temperate regions is an energy-starved system. The biotic communities that reside in these systems are low in diversity and simple in structure, and sensitive to changes in external inputs of organic matter. Caves open to tourist traffic offer an opportunity to study the impacts of organic matter amendments in the form of human and rodent hair and dander, clothing lint, material from rodent activity (nesting materials and feces), and algal growth in and around artificial lighting. This study reports on the impacts of carbon …

Productivity-Diversity Relationships From Chemolithoautotrophically Based Sulfidic Karst Systems, Megan L. Porter, Annette Summers Engel, Thomas C. Kane, Brian K. Kinkle Jan 2009

Productivity-Diversity Relationships From Chemolithoautotrophically Based Sulfidic Karst Systems, Megan L. Porter, Annette Summers Engel, Thomas C. Kane, Brian K. Kinkle

International Journal of Speleology

Although ecosystems thriving in the absence of photosynthetic processes are no longer considered unique phenomena, we have yet to understand how these ecosystems are energetically sustained via chemosynthesis. Ecosystem energetics were measured in microbial mats from active sulfidic caves (Movile Cave, Romania; Frasassi Caves, Italy; Lower Kane Cave, Wyoming, USA; and Cesspool Cave, Virginia, USA) using radiotracer techniques. We also estimated bacterial diversity using 16S rRNA sequences to relate the productivity measurements to the composition of the microbial communities. All of the microbial communities investigated were dominated by chemolithoautotrophic productivity, with the highest rates from Movile Cave at 281 g …

A Comparison Of Development Times For Protophormia Terraenovae (R-D) Reared On Different Food Substrates, J.-A. Warren, G. S. Anderson Jan 2009

A Comparison Of Development Times For Protophormia Terraenovae (R-D) Reared On Different Food Substrates, J.-A. Warren, G. S. Anderson

Faculty Research, Scholarly, and Creative Activity

Experiments were conducted to determine whether a specific larval substrate impacted immature development rates. Protophormia terraenovae was raised on beef organs and compared with whole carcasses (rat), to determine if discrepancies in development times were observed. The minimum development time on beef liver was the most consistent with the rat carcass but a significant difference between all substrates was found after the third instar. These differences can be explained by the differences found between heart and muscle and the other substrates. Often length of the larvae is used to estimate insect age and so Day 2 measuring of weight, length, …

Ecology And Molecular Phylogenetics Of Hydnora (Hydnoraceae) In Southern Africa, Jay Francis Bolin Jan 2009

Ecology And Molecular Phylogenetics Of Hydnora (Hydnoraceae) In Southern Africa, Jay Francis Bolin

Biological Sciences Theses & Dissertations

The Hydnoraceae are a clade of root holoparasitic angiosperms that contain two small genera, Hydnora and Prosopanche. This study, focused on Hydnora, presents novel data regarding the pollination biology, germination ecology, parasite-host nutritional relationships, and the molecular systematics of this group. Experimental addition of the primary pollinator, Dermestes maculatus to Hydnora africana chamber flowers demonstrated beetle imprisonment during the carpellate stage. Changes in the inner surfaces of the androecial chamber allowed beetle escape after pollen release. Most beetles escaped, dusted with viable pollen, three days after pollen release. To investigate germination ecology, aqueous root extracts of host and …

Eighth Amendment Gaps: Can Conditions Of Confinement Litigation Benefit From Proportionality Theory, Alexander A. Reinert Jan 2009

Eighth Amendment Gaps: Can Conditions Of Confinement Litigation Benefit From Proportionality Theory, Alexander A. Reinert

Articles

The article explores the potential benefits of integrating proportionality theory, traditionally used in sentencing, into litigation challenging harsh prison conditions under the Eighth Amendment. It argues that this approach could offer new avenues for addressing the limitations of current conditions of confinement jurisprudence by reimagining how courts evaluate the constitutionality of prison conditions.

Development Of Pcr Assays To Detect And Characterise Salmonella Isolates From Cheese And Meat Products By Utilizing An Integrated Genomics Approach, Noelle Geraldine Mccarthy Jan 2009

Development Of Pcr Assays To Detect And Characterise Salmonella Isolates From Cheese And Meat Products By Utilizing An Integrated Genomics Approach, Noelle Geraldine Mccarthy

Theses

Salmonella is an extremely important foodborne pathogen which is responsible for millions of cases of gastroenteritis annually. Salmonella outbreaks have been associated with numerous common food stuffs such as meat, dairy and seafood. Traditional culture based methods for the detection Salmonella are labourious and costly. Rapid detection and serovar identification would dramatically decrease the time required to identify Salmonella outbreaks and potential health risks.

In the US 99% of all Salmonella infections are caused by members of Salmonella enterica subspecies 1. Serovars within subspecies 1 can vary due to host specificity and pathogenicity. DNA sequencing projects have provided valuable genomic …

The Detection And Analysis Of Selected Pathogens In The Water Supply Of Dairy Cohort Herds., Niall Drummond Jan 2009

The Detection And Analysis Of Selected Pathogens In The Water Supply Of Dairy Cohort Herds., Niall Drummond

Theses

The issue of food safety is a growing concern to Irish agriculture and to the food industry. In the last twenty years, many novel pathogens, which can cause concern to the food industry, have been identified. Organisms such as Verocytotoxigenic Escherichia coli (VTEC) cause hemorrhagic colitis and Haemolytic Uremic Syndrome (HUS), which is the major cause of acute renal failure in children in many countries, and it’s presence in food is a cause of growing concern. Other pathogens causing concern are those implicated as causative agents of Acute Gastroenteritis (AGE) and these can be bacterial, viral and parasitic organisms.

Consumption …

Renovating A Control Engineering Laboratory For Enhanced Learning, David Hamilton Jan 2009

Renovating A Control Engineering Laboratory For Enhanced Learning, David Hamilton

Theses

In an attempt to both place the student at the centre of the learning process and thus improve student engagement with the discipline, and to incorporate professional skills into the engineering curriculum at the module level, an enquiry based learning (EBL) component to enhance teamwork, problem solving skills and the student’s ability to learn independently was developed using current best practice in teaching and learning. As EBL is not widely used in engineering curricula in CIT, a pilot scale three week trial was performed during the academic year 2007-08 to evaluate the appropriateness of the pedagogy and student reactions to …
