Genomic Evidence For Independent Origins Of Β-Like Globin Genes In Monotremes And Therian Mammals, Juan C. Opazo, Federico G. Hoffmann, Jay F. Storz

Phylogenetic reconstructions of the β-globin gene family in vertebrates have revealed that developmentally regulated systems of hemoglobin synthesis have been reinvented multiple times in independent lineages. For example, the functional differentiation of embryonic and adult β-like globin genes occurred independently in birds and mammals. In both taxa, the embryonic β-globin gene is exclusively expressed in primitive erythroid cells derived from the yolk sac. However, the “ε-globin” gene in birds is not orthologous to the ε-globin gene in mammals, because they are independently derived from lineage-specific duplications of a proto β-globin gene. Here, we report evidence that the early and late …

Role Of Protein Kinase C-Iota In Prostate Cancer, Hla Yee Win
Role Of Protein Kinase C-Iota In Prostate Cancer, Hla Yee Win
USF Tampa Graduate Theses and Dissertations
Prostate cancer is one of the leading causes of death among males in the United States. In this study, we hypothesized that an activated PKC-iota-dependent anti-apoptotic pathway, drives the cell cycle proliferation and survival of prostate cancer cells.
We investigated the role of atypical PKC-iota (PKC-ι) in androgen- independent prostate DU-145 carcinoma, androgen-dependent prostate LNCaP carcinoma compared to transformed non-malignant prostate RWPE-1 cells. Western blotting and immunoprecipitations demonstrated that PKC-ι is associated with cyclin-dependent activating kinase (CAK/Cdk7) in androgen-dependent, RWPE-1 and LNCaP cells but not in androgen-independent DU-145 cells. Treatment of prostate RWPE-1 cells with PKC-ι silencing RNA (siRNA) decreased …

Exposure to ozone, which is a major component of air pollution, induces a form of asthma that occurs in the absence of adaptive immunity. Although ozone-induced asthma is characterized by airway neutrophilia, and not eosinophilia, it is nevertheless associated with airway hyperreactivity (AHR), which is a cardinal feature of asthma. Because AHR induced by allergens requires the presence of natural killer T (NKT) cells, we asked whether ozone-induced AHR had similar requirements. We found that repeated exposure of wild-type (WT) mice to ozone induced severe AHR associated with an increase in airway NKT cells, neutrophils, and macrophages. Surprisingly, NKT cell-deficient …

Mode-Locking In Driven Disordered Systems As A Boundary-Value Problem, William Kung, M. Cristina Marchetti
Mode-Locking In Driven Disordered Systems As A Boundary-Value Problem, William Kung, M. Cristina Marchetti
Physics - All Scholarship
We study mode-locking in disordered media as a boundary-value problem. Focusing on the simplest class of mode-locking models which consists of a single driven overdamped degree-of-freedom, we develop an analytical method to obtain the shape of the Arnol'd tongues in the regime of low ac-driving amplitude or high ac-driving frequency. The method is exact for a scalloped pinning potential and easily adapted to other pinning potentials. It is complementary to the analysis based on the well-known Shapiro's argument that holds in the perturbative regime of large driving amplitudes or low driving frequency, where the effect of pinning is weak.

Nematic And Polar Order In Active Filament Solutions, A. Ahmadi, Tanniemola B. Liverpool, M. Cristina Marchetti
Physics - All Scholarship
Using a microscopic model of interacting polar biofilaments and motor proteins, we characterize the phase diagram of both homogeneous and inhomogeneous states in terms of experimental parameters. The polarity of motor clusters is key in determining the organization of the filaments in homogeneous isotropic, polarized and nematic states, while motor-induced bundling yields spatially inhomogeneous structures.

Copper plays a fundamental role in regulating cell growth. Many types of human cancer tissues have higher copper levels than normal tissues. Copper can also induce gene expression. However, transcription factors that mediate copper-induced cell proliferation have not been identified in mammals. Here we show that antioxidant-1 (Atox1), previously appreciated as a copper chaperone, represents a novel copper-dependent transcription factor that mediates copper-induced cell proliferation. Stimulation of mouse embryonic fibroblasts (MEFs) with copper markedly increased cell proliferation, cyclin D1 expression, and entry into S phase, which were completely abolished in Atox1-/- MEFs. Promoter analysis and EMSA revealed that copper …
Service Learning: A Concept We Are Not As Familiar With As We Might Think, Jena Spindler
Service Learning: A Concept We Are Not As Familiar With As We Might Think, Jena Spindler
Reason and Respect
Despite a common belief, service learning is completely different from community service. Service learning is different from community service because not only is working being accomplished in a community, but also students are learning how to apply service to their everyday lives as citizens of the community. Active citizenship is an essential skill to add to higher education curriculums because the students graduating are more likely going to become our future leaders.
