Cdc45 Function Alters Cell Sensitivity To Dna Topoisomerase I Poisons, Cynthia Sue Lancaster

Eukaryotic DNA topoisomerase I (Top1) is a highly conserved enzyme that functions to manage the torsional strain of DNA during cellular processes such as transcription, replication, chromatid condensation and recombination. The enzyme binds duplex DNA and through a series of strand cleavage and religation reactions removes positive or negative supercoils relieving torsional strain. Top1 is the sole cellular target of the anticancer agent camptothecin, which stabilizes the covalent complex. CPT cytotoxicity is S-phase dependent. It has been suggested that the mechanism of this S-phase toxicity is due to the advancing replication forks either colliding with the stabilized drug-enzyme-DNA intermediate or …

Factors Influencing Topotecan Cns Penetration In Mouse Models, Jun Shen

Camptothecin analogs such as topotecan are currently tested in clinical trials for brain tumors. However the clinical outcome is far below the expectations, which are derived from the promising preclinical studies. This discrepancy could be partially attributed to the presence of two barrier systems in the brain, the blood brain barrier (BBB) and blood cerebrospinal fluid (CSF) barrier (BCB). Further investigations have demonstrated multiple ATP-binding cassette (ABC) transporters are present at the two barriers. Camptothecin analogs are well known substrates for several of the transporters. This work defined the role of two ABC transporters, Bcrp1 and P-gp in determination of …

Mammary epithelial cells are highly efficient secreting cells. This natural phenomenon has triggered research efforts to focus on using these cells to assist in the production of viable liquids. The application of this technology is nearly endless. If developed successfully, mammary epithelial cells could be used by industries from farming to pharmacology.
The feasibility of culturing epithelial mammary cells which secrete within a three-dimensional scaffold was investigated and is documented within this paper. A biocompatible film with suitable mechanical characteristics and processing properties for use in a bioreactor was developed through multiple iterations. Progress in the field of DNA testing over the last few decades has resulted in increasingly accurate results. Often this means that DNA evidence found to be inconclusive years ago may prove exculpatory to a prisoner if subjected to today’s more advanced testing. Prisoners may potentially gain access to this evidence through two methods: 1) a writ of habeas corpus; or 2) a § 1983 action. While both approaches can yield the same results, a writ of habeas corpus is subject to several procedural requirements which can delay a prisoner’s access to testing, making a § 1983 action a potentially …

Scientists are considering two factors that may be important in the pathogenesis of C. albicans', it's capability to morph from yeast to hyphal phase, and its differences in colony morphology, cell shape, cell surface and cell permeability. (5) Unfortunately, unlike the bacteria, yeasts do not yet have proven virulence factors. Falls are a continually rising issue in today's healthcare. In acute care settings, patient falls make up 38% of all adverse events in which include physical injury, undesirable emotional and financial outcomes for the client (Angn, Mordiffi, Wong, Devi, & Evans, 2007). Falls in the hospitals lead to fear, pain, decreased healing, longer in-patient stays, further health-related complications. Falls may also cause patient discomfort and affect quality of life. The structural characteristics of a healthy tendon are related to the anisotropic speckle patterns observed in ultrasonic images. This speckle orientation is disrupted upon damage to the tendon structure as observed in patients with tendinopathy. Quantification of the structural appearance of tendon shows promise in creating a tool for diagnosing, prognosing, or measuring changes in tendon organization over time. The current work describes a first step taken towards this goal—classification of Achilles tendon images into tendinopathy and control categories. Eight spatial frequency parameters were extracted from regions of interest on tendon images, filtered and classified using linear discriminant analysis. Resulting …

On June 15, 1915, the renowned Yale historian-turned-explorer Hiram Bingham III returned from a three-week trek along the Salcantay trail to find his Ollantaytambo headquarters occupied by four hostile young Peruvian men. In the name of the Instituto Histórico del Cuzco, they accused him of violating the prefect’s decree prohibiting the excavation of Inca sites in the vicinity. Their suspicions were not unwarranted. Lining the walls of the office were large wooden cases, full of archaeological specimens. Many individuals are confused as to what exactly a trickster is. This paper will attempt to explain its nature in terms of defining characteristics. The intention of this research was to bring light to the current state mandated testing, and possible solutions in assisting educators to address the issues of students not meeting the standards. In more general terms, this study is looking to prove to what extent a student's learning style has on their performance on the Palmetto Achievement Challenge Test (PACT). This study analyzed four of the twenty-six learning preference areas identified by the Learning Style Instrument (LSI) of middle school students in a traditional public middle school in seventh grade.
