Mgra Represses Biofilm Formation In Staphylococcus Aureus, Maria P. Trotonda, Sandeep Tamber, Guido Memmi, Ambrose L. Cheung

MgrA is a pleiotropic regulator that controls autolysis, virulence, and efflux pump activity in Staphylococcus aureus. We recently found that mgrA mutants of strains RN6390, SH1000, and MW2 also displayed enhanced biofilm formation compared with their respective parents. The biofilms formed by mgrA mutants of RN6390 and MW2 are independent of sigB and ica loci, two genetic elements that have been previously associated with biofilm formation in S. aureus. Biofilms formed by mgrA mutants are dependent on the expression of surface proteins mediated by the sortase gene srtA. Extracellular DNA was also a crucial component of the early biofilm of …

Emerging evidence suggests that the capacity of a tumor to grow and propagate is dependent on a small subset of cells within a tumor, termed cancer stem cells. In fact, cancer cells, like stem cells, can proliferate indefinitely through a dysregulated cellular self-renewal capacity. Cancer stem cells may originate due to the distribution into self-renewal and differentiation pathways occurring in multi-potential stem cells, tissue-specific stem cells, progenitor cells and cancer cells. Recent studies have shown that ovarian cancer also contains stem cells or tumor-initiating cells. BACKGROUND: The incidence of malignant melanoma has significantly increased over the last decade. Some of these malignancies are susceptible to the growth inhibitory and pro-differentiating effects of all-trans-retinoic acid (RA). The molecular changes responsible for the biological activity of RA in melanoma are not well understood.

RESULTS: In an analysis of sequential global gene expression changes during a 4-48 h RA treatment of B16 mouse melanoma cells, we found that RA increased the expression of 757 genes and decreased the expression of 737 genes. Review Of The West Indian Species Of Efferia Coquillett (Diptera: Asilidae): Part 1. Bahamas, Cayman Islands, Cuba, And Jamaica, Aubrey G. Scarbrough, Daniel E. Perez-Gelabert

The genus Efferia Coquillett from the Bahamas, Cayman Islands, Cuba, and Jamaica is reviewed. The fauna now totals 16 species with 6 new species described (Ef. bellardii n. sp., Ef. bromleyi n. sp., Ef. hinei n. sp., Ef. insula n. sp., Ef. pina n. sp., and Ef. vinalensis n. sp.). Cuba has the greatest diversity with 10 species, Jamaica 3, the Bahamas 2, and the Cayman Islands 1. Efferia stylata (Fabricius) is removed from the species list of these West Indian islands. Taxonomic Review Of The Neotropical Tetragonoderus Quadriguttatus Assemblage (Coleoptera: Carabidae: Cyclosomini) With Description Of T. Deuvei, New Species, And New West Indian And Nearctic Locality Records, Danny Shpeley, George E. Ball

The Tetragonoderus (Peronoscelis) quadriguttatus assemblage is a postulated monophyletic part of the intersectus complex. This assemblage is characterized against a background that includes review of a classification of the genus and comparison, in the form of a key, to other unrelated Western Hemisphere genera that share elongate tibial spurs with Tetragonoderus Dejean. The principal, easily observed feature that unites the three members (T. laevigatus Chaudoir, 1876; T. deuvei, new species (type locality: Cuijaba, Mato Grosso, Brazil), and T. quadriguttatus Dejean) of the quadriguttatus assemblage is the four-spotted elytra (two spots per elytron). A New Genus Of Telephanine Silvanidae (Coleoptera: Cucujoidea), With A Diagnosis Of The Tribe And Key To Genera, Michael C. Thomas, Eugenio H. Nearns

Australophanus, new genus, is described and illustrated for Cryptamorpha redtenbacheri (Reitter). Platamus Erichson is synonymized under Telephanus Erichson, new synonymy. Euplatamus Sharp, new status, replaces Platamus Erichson as the genus name. Type species are designated for Aplatamus Grouvelle and Euplatamus Sharp. Telephanus velox (Haldeman) is synonymized under Telephanus atricapillus Erichson. A diagnosis of the tribe Telephanini, a key to the described telephanine genera of the world is presented, and a phylogeny of the family Silvanidae is proposed.

Mutation of sarA in Staphylococcus aureus results in a reduced capacity to form a biofilm, but the mechanistic basis for this remains unknown. Previous transcriptional profiling experiments identified a number of genes that are differentially expressed both in a biofilm and in a sarA mutant. This included genes involved in acid tolerance and the production of nucleolytic and proteolytic exoenzymes. Based on this we generated mutations in alsSD, nuc and sspA in the S. aureus clinical isolate UAMS-1 and its isogenic sarA mutant and assessed the impact on biofilm formation. Epidemiological and clinical studies provide compelling support for a causal relationship between Helicobacter pylori infection and endothelial dysfunction, leading to vascular diseases. However, clear biochemical evidence for this association is limited. In the present study, we have conducted a comprehensive investigation of endothelial injury in bovine aortic endothelial cells (BAECs) induced by H. pylori-conditioned medium (HPCM) prepared from H. pylori 60190 [vacuolating cytotoxin A (Vac(+))]. BAECs were treated with either unconditioned media, HPCM (0-25% vol/vol), or Escherichia coli-conditioned media for 24 h, and cell functions were monitored.
