The central melanocortin system plays a critical role in the regulation of energy balance in rodents and humans. The melanocortin signals in both the hypothalamus and brainstem contribute to this regulation. However, how the melanocortin signals of the hypothalamus interact with those intrinsic to the brainstem in the regulation of energy balance is poorly understood. The brainstem raphe pallidus (RPa) and adjacent areas contain melanocortin 4 receptor (MC4-R)-bearing neurons and sympathetic premotor neurons regulating thermogenesis. Here we report that α-melanocyte-stimulating hormone (α-MSH)-immunoreactive (IR) fibers are in close apposition to MC4-R neurons in the RPa. Retrograde tracing studies revealed a unique …

Abstract Hormones and agonists that enhance cell survival through binding specific G Protein-Coupled Receptors (GPCRs) are of particular interest in cancer cell survival. LNCaP cells have been shown to express muscarinic cholinergic receptors that are responsive to the agonist, carbachol. In addition, LNCaP cells specifically express the M3-subtype of GPCR’s that may couple carbachol to Gq, increases in intracellular calcium, and activation of the intracelluar Ca2+/calmodulin-dependent protein kinases (CaM Ks). The CaM Kinase family of proteins includes CaM KII, CaM Kinase Kinase (CaM KK) and its substrates CaM KIV, CaM KI and the protein kinase AKT. AKT is an anti-apoptotic …

Mutational Analysis Of The Stability Of The H2a And H2b Histone Monomers, Matthew R. Stump, Lisa M. Gloss

The eukaryotic histone heterodimer H2A–H2B folds through an obligatory dimeric intermediate that forms in a nearly diffusion-limited association reaction in the stopped-flow dead time. It is unclear whether there is partial folding of the isolated monomers before association. To address the possible contributions of structure in the monomers to the rapid association, we characterized H2A and H2B monomers in the absence of their heterodimeric partner. By far-UV circular dichroism, the H2A and H2B monomers are 15% and 31% helical, respectively—significantly less than observed in X-ray crystal structures. Acrylamide quenching of the intrinsic Tyr fluorescence was indicative of tertiary structure. The …

Bioenergetic Profiles Of Isolated Mitochondria From Diapausing Embryos Of The Annual Killifish Austrofundulus Limnaeus, Jeff Duerr, Jason E. Podrabsky

The annual killifish, Austrofundulus limnaeus, occupy ephemeral pond habitats by producing diapausing embryos which can reduce their oxygen consumption rate by up 90% compared to developing embryos. Mitochondria isolated from whole embryos collected at 10 dpf, diapause II, post-diapause, diapause III, post-hatch larvae, and adult liver exhibited oxygen consumption rates that increased with developmental stage in the presence of glutamate or succinate. Measurements of electron transport chain activity reflected life-stage specific regulation. Minimum ETC activity was observed during diapause II: complex I activity was 12% of adult, complex II was 25% of adult, and complex IV activity was 20% of …

The purpose of this study was to evaluate the effectiveness of current education materials on changing maternal smoking attitudes. Children are affected by prenatal exposure to tobacco smoke. Although there are educational efforts to discourage pregnant women from smoking, the practice continues in a significant number of pregnancies. New materials, based on current research, were also evaluated for effectiveness. The purpose of the present investigation is to examine the regulation and expression of the B. burgdorferi spoT/rpoS pathway under normal growth conditions and during serum starvation in wild-type strains and in spoT, rpoS and rpoN mutant strains. Additionally the expression of B. burgdorferi's rpoN gene has been examined within a surrogate Escherichia coli system by using a GFP reporter construct in order to determine if SpoT, RpoS or RpoN affect rpoN expression. Fish oil accounts for approximately 26.8% of the operating costs of salmon aquaculture. To compensate for the increasing demand in fish oil, alternative plant and animal oils have been used to replace the fish oil. The goal of this study was to formulate a cost-effective fish feed which supports adequate growth of salmon and is less susceptible to oxidative damage while maintaining the characteristic beneficial fatty acid profile. The focus of this work centers on molecular structuring of surfaces. The surface modifications were achieved using plasma polymerization technology, specifically variable duty cycle pulsed plasmas. The results obtained demonstrate that both exacting film chemistry and film thickness control were achieved during deposition of thin polymeric films on selected substrates. After felling, it was revealed that Prometheus, an ancient bristlecone pine (Pinus longaeva D.K. Bailey) located in the Great Basin National Park, had been the oldest living organism at over 5000 years of age. Great Basin bristlecone pines are famous for their longevity, living thousands of years under extreme conditions of temperature and moisture at high altitudes. For the majority of conifer trees, a period of rapid growth in tracheid lengths has been observed from the pith up to ten to forty years of age. Transposable elements (TEs) are mobile repetitive sequences that make up large fractions of mammalian genomes, including at least 45% of the human genome. Class 2, or DNA intermediate transposons, which make up approximately 3% of human nuclear DNA, have typically been overlooked in analyses of mammalian genomes and therefore remain poorly understood. Here, we carried out the first large-scale analysis of the evolutionary history of DNA transposons throughout eutherian mammal evolution. The analysis included 18 mammals from all major branches of the eutherian tree including Afrotheria, Laurasiatheria, primates, rodents and Xenarthra. Over the last two decades, the United States and the People’s Republic of China have engaged in extensive negotiations regarding China’s protection of intellectual property rights. To date, the countries have entered into at least four substantive agreements detailing China’s commitments and obligations to enforce intellectual property rights (IPRs). Unfortunately, these commitments have not led to significant improvement in China’s enforcement of rampant piracy. Sediment transport in a coastal embayment was documented on multiple spatial and temporal scales. An inlet formed in Chatham Harbor in 1987 and as a result the primary flood-tidal delta began to migrate. The relationship between the evolution of the tidal inlet and the flood-tidal delta was documented using field studies and spatial analyses of georeferenced aerial photographs. A feedback mechanism was documented such that the migrating flood-tidal delta increases in surface area, which causes the ebb-tidal flow to become more channelized, which in turn allows more intertidal area to be created by flood-tidal currents and further channelizing ebb-tidal flow. …

The peripherally circulating peptide hormone ghrelin is purported to stimulate hunger and feeding in humans, and is affected by the energy storage state as reflected in body mass index and body composition, and by macronutrient content of the diet. There have been few studies in humans that have measured both ghrelin, in its various isoforms, and appetite. To address this, Manuscript #1 of this dissertation examined fasting and postprandial ghrelin and ratings of appetite, in addition to anthropometric and fitness measures in 50 college-age student. The following study was implemented to explore the potential for a four-week curriculum module in gerontological social work education to positively impact students’ practice-related knowledge concerning older adults, as well as their attitudes toward elderly persons and interest in working with them. Pertinent literature was reviewed to conceptualize the building of a gerontology curriculum module that could be integrated into a pre-existing social work course. The module’s design and content were reflective of its primary goal, which was to educate students about salient aspects of social work knowledge, skill, and practice with older adults utilizing experiential methods. The argument against crafting federal regulations for problems stemming from development in disaster-prone areas (nation-on-edge problems) assumes that these types of problems are essentially local problems requiring unique local solutions. In this Article, Jeffrey G. Miller challenges this assumption, reasoning that a flexible framework of federal regulations would indeed be effective at remedying these problems. He suggests that such a framework could be modeled after the Clean Water Act's (CWA's) point source pollution control regime. The Article begins by discussing the Hazelwood decision in depth. It then discusses the various contexts in which courts have applied Hazelwood and the circuit split that has developed over how broadly Hazelwood should reach. Next, it describes the circuit split over whether Hazelwood permits viewpoint-based speech restrictions, highlighting the different speech contexts in which the circuits have reached divergent conclusions. The Article then argues that the overextension of Hazelwood links the two splits.
