A Kinetic Model For Growth And Biosynthesis Of Medium-Chain-Length Poly-(3-Hydroxyalkanoates) In Pseudomonas Putida, Mohamad Suffian Mohamad Annuar, Irene Kit Ping Tan, Shaliza Ibrahim, K.B. Ramachandran Jan 2008

A Kinetic Model For Growth And Biosynthesis Of Medium-Chain-Length Poly-(3-Hydroxyalkanoates) In Pseudomonas Putida, Mohamad Suffian Mohamad Annuar, Irene Kit Ping Tan, Shaliza Ibrahim, K.B. Ramachandran

Research Publications (2006 to 2010)

A kinetic model is presented giving a mathematical description of batch culture of Pseudomonas putida PGA1 grown using saponified palm kernel oil as carbon source and ammonium as the limiting nutrient. The growth of the micro-organism is well-described using Tessier-type model which takes into account the inhibitory effect of ammonium at high concentrations. The ammonium consumption rate by the cells is related in proportion to the rate of growth. The intracellular production of medium-chain-length poly-(3-hydroxyalkanoates) (PHAMCL) by P. putida PGA1 cells is reasonably modeled by the modified Luedeking-Piret kinetics, which incorporate a function of product synthesis inhibition (or reduction) by …

Discordance In The Paraoxonase (Pon1)-192qr Polymorphism, R. Poh, S. Muniandy

Research Publications (2006 to 2010)

Paraoxonase (PON1) has been implicated to have a cardioprotective role, due to its physical attachment with high-density lipoprotein. PON1192QR is a variation of the PON1 gene, the R allele being a risk factor for cardiovascular disease. Kinetic studies resulting in a plot of paraoxon versus diazoxon hydrolysis rates may be used to accurately predict PON1192 geno-type. In this study, paraoxonase and diazoxonase activities in plasma were measured spec-trophotometrically using plasma while PCR-based PON1192 genotyping was performed us-ing polymerase chain reaction followed by restriction digestion. The two-substrate assay-derived genotypes were cross-referred with those determined by PCR-based genotyping. When results did not …

Research Publications (2006 to 2010)

A simple and rapid real-time reverse transcription-PCR (RT-PCR) technique using SYBR green fluorescence was developed to quantify Zonula Occludens-1 (ZO-1) mRNA expression in histamine-treated cultured bovine retinal microvessel endothelial cells. There was no difference between the quantity of ZO-1 mRNA expression in histamine-treated (10(-7)M to 10(-9) M) and untreated bovine retinal endothelial cells at high magnesium levels. Immunohistochemistry staining and western blot assay unequivocally demonstrated the approximately similar levels of ZO-1 protein in both histamine-treated and non-treated endothelial cells at high magnesium concentrations.

Research Publications (2006 to 2010)

An Automated Rapid Maxillary Expander (ARME), is a specially designed orthodontic appliance to overcome the shortcomings imposed by the traditional butterfly expansion appliance. It operates by automatically widening the maxilla (upper jaw) by expanding the midpalatal suture [1]. This procedure is not feasible after late teenage years due to more rigid facial skeleton features. According to the study by Cozzani [2], the activation of the device could produce 0.25 mm of expansion per day. Over the period of 2 weeks, the expansion produced is up to 7 mm in total [1]. For this study, we explored the mechanism involved in …

Trends In Selenium Utilization In Marine Microbial World Revealed Through The Analysis Of The Global Ocean Sampling (Gos) Project, Yan Zhang, Vadim N. Gladyshev

Vadim Gladyshev Publications

Selenium is an important trace element that occurs in proteins in the form of selenocysteine (Sec) and in tRNAs in the form of selenouridine. Recent large-scale metagenomics projects provide an opportunity for understanding global trends in trace element utilization. Herein, we characterized the selenoproteome of the microbial marine community derived from the Global Ocean Sampling (GOS) expedition. More than 3,600 selenoprotein gene sequences belonging to 58 protein families were detected, including sequences representing 7 newly identified selenoprotein families, such as homologs of ferredoxin– thioredoxin reductase and serine protease. In addition, a new eukaryotic selenoprotein family, thiol reductase GILT, was identified. …

Domoic acid (DA) is a potent toxin produced by bloom-forming phytoplankton in the genus Pseudo-nitzschia, which is responsible for causing amnesic shellfish poisoning (ASP) in humans. ASP symptoms include vomiting, diarrhea, and in more severe cases confusion, loss of memory, disorientation, and even coma or death. This paper describes the development and validation of a rapid, sensitive, enzyme linked immunosorbent assay test kit for detecting DA using a monoclonal antibody. The assay gives equivalent results to those obtained using standard high performance liquid chromatography, fluorenylmethoxycarbonyl high performance liquid chromatography, or liquid chromatography—mass spectrometry methods. It has a linear range …

Mass Spectrometry Of Dendrimer Metal Complexes, Jason David Batchelor Jan 2008

Mass Spectrometry Of Dendrimer Metal Complexes, Jason David Batchelor

Theses, Dissertations and Capstones

Dendrimers are hyper-branched macromolecules with unique properties. They form using a “cascade-like” synthesis involving repetitive reaction steps. These steps are controlled, resulting in monodisperse dendrimers. Synthesis is divergent, core to exterior, or convergent, exterior to core. Dendrimers are created from various molecules with numerous functional properties. They are used as catalysts, diagnostic agents, and others.

This thesis studies 1st generation poly(propylene) imine, consisting of a diaminobutane core with 4 propyl amine branches. Fragmentation of protonated and metal (Co, Ni, Cu and Zn) complexed dendrimers was analyzed by ion-trap mass spectrometry. Two isotopically labeled dendrimers were synthesized for neutral loss identification …

The Aridity Of Grace: Community And Ecofeminism In Barbara Kingsolver's Animal Dreams And Prodigal Summer, Richard M. Magee Jan 2008

The Aridity Of Grace: Community And Ecofeminism In Barbara Kingsolver's Animal Dreams And Prodigal Summer, Richard M. Magee

English Faculty Publications

In both Animal Dreams and her later novel Prodigal Summer, Kingsolver constructs narratives of community inhabited by characters with a vivid awareness of the natural world and the threats to that world; furthermore, both novels feature strong female characters who long for a more harmonious life within nature. The novels develop and present forthright ecofeminist themes, with the women in the texts representing ideals of ecologically sensitive living who seek to educate their communities about threats to the environment and the defenses against those threats.

Kingsolver's ecofeminist vision, however, is frequently complicated and contradictory; just as the desert landscape …

Terrestrial Habitat Use Of Marbled Salamanders Ambystoma Opacum: A Site Specific Approach, Frank Piccininni Jan 2008

Terrestrial Habitat Use Of Marbled Salamanders Ambystoma Opacum: A Site Specific Approach, Frank Piccininni

Theses, Dissertations and Capstones

Habitat destruction, fragmentation and degradation have contributed heavily to the decline of amphibian populations such as Marbled Salamanders (Ambystoma opacum). Often, ambystomatid water resources are conserved without consideration for the equally important terrestrial habitat. This is partly due to a lack of information regarding the relationship between ecological succession, plant community composition, microhabitat and salamander abundance. Three sampling transects consisting of drift fence arrays, vegetation assessments and microhabitat surveys were extended 100 m into the terrestrial habitat surrounding a seasonal wetland at Beech Fork State Park in Wayne County, West Virginia. Principal components analysis was used to identify …

Abstract

Background

Globin isoforms with variant properties and functions have been found in the pseudocoel, body wall and cuticle of various nematode species and even in the eyespots of the insect-parasite Mermis nigrescens. In fact, much higher levels of complexity exist, as shown by recent whole genome analysis studies. In silico analysis of the genome of Caenorhabditis elegans revealed an unexpectedly high number of globin genes featuring a remarkable diversity in gene structure, amino acid sequence and expression profiles.

Results

In the present study we have analyzed whole genomic data from C. briggsae, C. remanei, Pristionchus pacificus and Brugia malayi …

Wayne State University Associated BioMed Central Scholarship

Abstract

Elevated expression of anti-apoptotic Bcl-2 family proteins have been linked to a poor survival rate of patients with Follicular Lymphoma (FL). This prompted us to evaluate a very potent non-peptidic Small-Molecule Inhibitor (SMI) targeting Bcl-2 family proteins, Apogossypolone (ApoG2) using follicular small cleaved cell lymphoma cell line (WSU-FSCCL) and cell isolated from lymphoma patients. ApoG2 inhibited the growth of WSU-FSCCL significantly with a 50% growth inhibition of cells (IC50) of 109 nM and decreased cell number of fresh lymphoma cells. ApoG2 activated caspases-9, -3, and -8, and the cleavage of Poly (ADP-ribose) polymerase (PARP) and Apoptosis Inducing …

Abstract

Background

Pancreatic carcinoma usually is a fatal disease with no cure, mainly due to its invasion and metastasis prior to diagnosis. We analyzed the gene expression profiles of paired primary pancreatic tumors and metastatic lesions from Ela-c-myc transgenic mice in order to identify genes that may be involved in the pancreatic cancer progression. Differentially expressed selected genes were verified by semi-quantitative and quantitative RT-PCR. To further evaluate the relevance of some of the selected differentially expressed genes, we investigated their expression pattern in human pancreatic cancer cell lines with high and low metastatic potentials.

Results

Data indicate that genes …

Horace Mann's Vision In Action: Bridgewater Normal School's Female Teachers, Casey Rekowski

Undergraduate Review

Through a close reading of Horace Mann’s archival material including official documents and speeches, this paper argues that Horace Mann expanded women’s social roles in the nineteenth century. Whether he intended such an outcome or not, Horace Mann’s agenda to improve common schools increased American women’s educational and professional opportunities. Drawing upon the popular ideology of the day, Mann articulated a detailed rationale for placing women, as “natural teachers” at the center of public education in Massachusetts when he became the first secretary of the Massachusetts Board of Education in 1837. This can best be seen through his annual reports …
