Understanding how diversity is attained and maintained is one of the central questions in biology. In the Neotropics most attention has centered on the Amazonian lowlands, despite the recognized importance of the role of the Andes in South American diversity. In this dissertation, I address the question by using Nothoprocta tinamous as a model case for examining diversification in the Andes. As a group, they are a manageable size, they exhibit near-restriction to the Andes, they show multi-species parapatric distributions, and they have presumed limited dispersal ability. These are all useful attributes for a system to study speciation and diversification. …

Syngas produced by gasification of biomass or coal can be converted directly to ethanol and higher alcohols by processes based on heterogeneous catalysts. Ethanol can be used as a neat fuel or fuel additive or as a hydrogen carrier. Rhodium-based catalysts have been found to be most selective for the formation of C2 oxygenates from the hydrogenation of CO when suitably promoted. This work describes the application of PMMA-based microanalytical devices for the affinity-type preconcentration of posttranslational modified proteins (PTMs). The choice of poly(methyl methacrylate), PMMA, is based on its biocompatibility, its functional methyl ester group for potential modification, and its extensive applications to create biological microelectromechanical systems (BioMEMS). Developing methodologies for preconcentration of PTMs is important for cancer diagnosis due to PTMs’ influence in the regulatory mechanism underlying the early stage of apoptosis or regulated cell death. In arid and semi-arid parts of the Old World, Phlebotomus paptasi is a significant biting pest of man and is the primary vector of Leishmania major, the causative agent of zoonotic cutaneous leishmaniasis (ZCL). Phlebotomus papatasi exhibits a close association with the burrowing rodents that serve as the reservoirs of L. major. Rodent burrows are considered to be the primary habitat of immature P. papatasi in ZCL foci, and sand fly larvae have been observed feeding on rodent feces. Heme oxygenase-1 (HO-1) catalyzes the initial and rate-limiting reaction in heme catabolism (yielding equimolar quantities of iron, carbon monoxide, and biliverdin), which is further metabolized to bilirubin by biliverdin reductase. HO-1, an important enzyme with antioxidant and cytoprotective properties, functions against oxidative stress generated in response to heme and to various stress stimuli, such as zinc. The cytoprotective function of HO-1 is dependent on both the activities of the reaction products and on the inducibility of the heme oxygenase gene (hmox-1), which is regulated primarily at the level of gene transcription by two distal enhancers, E1 and E2. Oncolytic herpes simplex virus type-1 (HSV-1) has shown great potential as an effective agent for cancer therapy ¨C oncolytic virotherapy. To enhance oncolytic capabilities of previously reported agents, new oncolytic and fusogenic HSV-1 OncSyn and OncdSyn viruses were constructed based on wild type HSV-1 (F) strain. To provide for safety and tumor selectivity, the viruses carried a large deletion including one of the two ¦Ã134.5 genes. This dissertation explores the development of analytical methods for studies of CVD biomarkers and related biomolecular indicators. Initially, spectroscopic studies were conducted to investigate the chemical reactivity of homocysteine (Hcy), an independent CVD risk factor and serological biomarker. Consequently, we proposed an alternate theory for in vivo Hcy clearance based on spontaneous pyridoxal tetrahydrothiazine (PT) formation from Hcy and pyridoxal. The validity of PT-assisted Hcy clearance was further evaluated by use of capillary electrophoretic methods, which allowed rapid monitoring of protein oligomerization in PT-protein reaction mixtures. This project analyzes eight novels which represent revolt or resistance by varied Native peoples against the European and Euro-American colonization of the Americas. I take a comparative approach to literatures of the Americas because of the dearth of research examining the literatures of both continents side by side, particularly literatures by and about Indigenous Americans. This research analyzes changes in the rhetoric of a sustaining myth in order to better assess what happens when a myth is threatened. By examining American sport and its current struggle to withstand the widespread use of steroids, the author investigates how public discourse about the scandal turns athletes from mythical heroes to cheaters. Although communication sciences and disorders (COMD) research supports intensive treatment for individuals with chronic brain injury, funding to provide these services is limited. This study explored the use of interdepartmental university resources to provide an intensive, multidisciplinary chronic brain injury program. Methodologically, treatment regimes were designed with clinical faculty as practicum experiences for COMD master’s degree students. Subjects with a single onset head injury or cerebral vascular accident greater than one year were recruited to participate in the Intensive Treatment, Weekly Treatment or Control Groups. This study investigated emerging phonemic awareness skills and printed sight word recognition abilities in two-year-old toddlers using plain text and MorphoPhonic Face words on flash cards. MorphoPhonic Face words were used to determine if this learning process is enhanced when words are iconically represented to show the first sound and the word's meaning (purposefully creating idiosyncratic cues) using pictures superimposed into the letters. Experiments were conducted in the state of Louisiana to evaluate methods to asses and potentially reduce water quality issues. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry was used for off-line analysis of bioaerosols. The overall goal of the work is to develop methods and instrumentation to minimize sample treatment and maximize sensitivity and selectivity for bioaerosol analysis. This work is divided into three parts: sample preparation, test bioaerosol analysis, and instrument development. The delay in early height growth (EHG) has been a limiting factor for artificial regeneration of longleaf pine (Pinus palustris Mill.). Simple Sequence Repeat (SSR) markers have been used to map the genome and quantitative trait loci controlling the EHG in a backcross family (longleaf pine x slash pine) x longleaf pine. The dimensions of microfluidic devices closely parallel those of biological cells; thusly, they are excellent platforms for the speciation, transport, manipulation, and analysis of cells. Electrokinetic transport of Escherichia coli and Saccharomyces cerevisiae was evaluated in microfluidic devices fabricated in pristine and UV-modified poly(methylmethacrylate) and polycarbonate. The magnitude and direction of transport of the cells was dictated by the buffer composition, conduit surface chemistry, and intrinsic cellular electrical properties. Vibrio vulnificus is the leading pathogen linked to seafood-associated deaths. This naturally occurring marine bacterium presents a human health risk when it is consumed in raw oysters. Hospitalization is necessary in 90 % Vibrio vulnificus illnesses and 40% end in death. (CDC) Several guidelines and regulations have been enacted to combat this human health issue, but have failed to reduce the steadily rising illness rate. Understanding den selection for the Louisiana black bear (Ursus americanus luteolus) may provide insight into habitat requirements of the subspecies and assist in conservation and management efforts. With that goal, I assessed den selection of female Louisiana black bears at multiple spatial scales in northern and central Louisiana. I used 230 den-years to examine den type (tree or ground), microhabitat characteristics at dens, and effects of landscape characteristics on den selection. We also evaluated tree availability and reuse. Solitary and parturient females selected tree dens more frequently (65%) than ground dens. This research focused on four previously identified single nucleotide polymorphisms in a leptin gene on chromosome 4 of beef cows. The SNP were E2FB, T945M, UA1, and UA2. Speciation, geographic variation, and genetic differentiation are fundamental processes that generate diversity, and understanding these processes are major goals of evolutionary biology. Evolutionary phenomena may be more observable on islands as compared to continental landmasses as a result of small population sizes, unoccupied niches, and the relative simplicity of island systems and their populations: physical isolation, shorter (and often well documented) geologic time scale, reduced faunal diversity, and lack of outside faunal influence. Par‐4 is a pro‐apoptotic tumor suppressor that is mutated, suppressed or inactivated in cancer. Par‐4 exploits components of the extrinsic pathway to cause apoptosis selectively of cancer cells. This study identified Par‐4 as an essential component of the apoptotic pathway induced by TRAIL, which selectively targets cancer cells. RNA interference‐mediated knockdown of Par‐4 rendered cancer cells unresponsive to TRAIL‐induced apoptosis. Cells with knocked‐down levels of Par‐4 were deficient in the activation of the apoptosis‐initiator caspase‐8 and the apoptosis‐effector caspase‐3 in response to TRAIL. Par‐4 was identified as a critical mediator of membrane translocation of caspase‐8 and the adapter protein FADD. …

The rice stink bug (Oebalus pugnax) is the major pest of late-season rice in Louisiana. Prior studies indicate that large populations of mosquitoes are often present in rice fields during the time that rice fields are infested with rice stink bugs. Microcosm and mesocosm scale experiments were carried out to investigate the effects of vegetation and substrates on the performance of engineered wetland systems (EWSs) meant to treat mixtures of volatile organic compounds (cVOCs). The experiments were conducted with a view to optimizing contributions of these wetland components. Phragmites communis and Typha latifolia, the commonest wetland plant species, were compared in mesocosm scale EWSs. The study of pedestrian locomotion in crustaceans has largely focused on forward walking macrurans, or sideway walking brachyurans. The spider crab, Libinia emarginata is a brachyuran that, unlike its close relatives, preferentially walks forward. The phylogenetic position, behavioral preference, and amenability to experimental techniques make spider crabs an attractive model for comparative studies of crustacean locomotion. This dissertation looks at the neuroethology of forward walking in L. emarginata. I described the skeletal, muscular, and neural anatomy of the walking machinery of L. emarginata and found adaptations at each level that reflect its walking preference. Chlamydophila psittaci causes severe disease in birds and humans, and important economical losses in the avian companion and poultry industry. Vaccines are the most cost-effective measure to control and help prevent infectious diseases, but to date there is no commercial vaccine available. A randomized clinical trial was conducted to assess the efficacy of two recombinant DNA vaccines against C. psittaci in Cockatiels (Nymphicus hollandicus). The first recombinant DNA vaccine has a gene encoding MOMP and the immunostimulant chitosan (MOMP DNA vaccine).
