Lipoxygenases (LOXs) catalyze the regiospecific and stereospecific dioxygenation of polyunsaturated membrane-embedded fatty acids. A Ca 2+-dependent membrane-binding function was localized to the amino-terminal C2-like domain of 8R-lipoxygenase (8R-LOX) from the soft coral Plexaura homomalla. The 3.2 Å crystal structure of 8R-LOX and spectroscopic data suggested that Ca2+ stabilizes two membrane-insertion loops. Analysis of the protein packing contacts in the crystal lattice indicated that the conformation of one of the two loops complicated efforts to improve the resolution of the X-ray data. A deletion mutant of 8R-LOX in which the corresponding membrane-insertion loop is absent (Δ41-45:GSLOX) was engineered. Removal of the …

Ruminal methanogens reduce carbon dioxide to methane (CH4), thereby preventing hydrogen use by bacteria for VFA synthesis resulting in a 2 to 12% loss in feed gross energy. Methane is a greenhouse gas that contributes to global warming. The objectives of this work were to determine: 1) the extent to which ruminal cultures acquire resistance to a nitrofuranyl derivative of para-aminobenzoate (NFP) and an extract from the plant Yucca shidigera (Yucca); 2) the effect of distillers dried grains plus solubles (DDGS) on ruminal CH4 production; 3) the effect of brome hay-based diets, corn-based diets, and in vivo …

Background: Human mast cells are multifunctional cells capable of a wide variety of inflammatory responses. Baicalein (BAI), isolated from the traditional Chinese herbal medicine Huangqin (Scutellaria baicalensis Georgi), has been shown to have anti-inflammatory effects. We examined its effects and mechanisms on the expression of inflammatory cytokines in an IL-1β- and TNF-α-activated human mast cell line, HMC-1. Methods: HMC-1 cells were stimulated either with IL-1β (10 ng/ml) or TNF-α (100 U/ml) in the presence or absence of BAI. This paper presents the analysis, modeling and design of zero-voltage-switching (ZVS) quasi resonant boost converter. The use of M-type switch in the topology will be described, including circuitpsilas mode of operation along with the derivations of critical voltage and current equations in each mode. Computer simulation using OrCAD Pspice and hardware development for the resonant converter with digital control will be presented. Results from both simulation and laboratory measurements to compare with theoretical analysis will be discussed.

Influence of Uracil in Fermentation Media on β-Glucan Production by Agrobacterium Radiobacter A 1.5 and Agrobacterium sp. Bro 1.2.1. Optimum β-glucan production can be achieved by an optimum condition in the fermentation media. Uracil, as a precursor of UDP-glucose, may act as a glucose donor in the formation of polysaccharides such as β-glucan. It is expected that addition of certain quantity of uracil into the fermentation media in a suitable growth phase of Agrobacterium radiobacter A 1.5 and Agrobacterium sp. Selection and test of L-histidine decarboxylase enzyme activity of six isolates of histamine forming bacteria. Six isolates of histamine forming bacteria were screened to see the degree of ability in producing histamine on modified Niven’s medium. The result showed that the six bacteria were able to produce histamine by giving a pinkish color on the medium, which could be used as a preliminary identification of histamine-forming bacteria (HFB). The isolates were grown in liquid modified Niven medium to measure the production of histamine. The histamine produced were determined by Hardy and Smith method. The result showed that all of the …

Estrogenic Effect of 70% Ethanol Turmeric (Curcuma domestica Val.) extract on ovariectomized Female Mice (Mus musculus L.). The influence of extract turmeric (Curcuma domestica Val.) on endometrium thickness, vaginal epithelium, mammary gland, and protein of estrogen receptor of ovariectomized mice was examined. Twenty five ovariectomized mice which were divided into five groups, were treated by ethynilestradiol (8,4 x 10-3 g), aquades (10 ml), and turmeric extract at doses 230 mg/kg b.w.; 310 mg/kg b.w.; and 390 mg/kg b.w. for eight days. Submersible observations during four missions over the North Carolina and Virginia continental slopes (184–900 m) documented the occurrence of large aggregations of mesopelagic Wshes and macronektonic invertebrates near or on the bottom. Aggregated mesopelagics formed a layer up to tens of meters deep positioned from a few centimeters to 20 m, usually <10 >m, above the substrate. Aggregations were numerically dominated by microvores, notably the myctophid Wsh Ceratoscopelus maderensis and the penaeid shrimp Sergestes arcticus. Consistently present but in relatively lower numbers, were mesopelagic predators, including the paralepidids Notolepis rissoi and Lestidium atlanticum, the eel Nemichthys scolopaceus, the …

Many bioluminescence observations are made from the ocean's surface. However, the depth of the bioluminescent source is difficult to estimate on the basis of surface observations alone, given the variable light attenuation of unknown concentrations of water column constituents such as phytoplankton, colored dissolved organic matter, and detritus. Part 1 of this paper showed that bioluminescent water-leaving radiance signals are detectable, even in extremely turbid and dynamic coastal waters. Here, in part 2 of this paper, we analyze the water-leaving radiance patterns of bioluminescence modeled by HydroLight 4.2 to determine if the depth of the bioluminescent source can be estimated …

In this paper, we propose measures for compressed data structures, in which space usage is measured in a data-aware manner. In particular, we consider the fundamental dictionary problem on set data, where the task is to construct a data structure for representing a set S of n items out of a universe U = {0, ..., u - 1} and supporting various queries on S. Physics

Nighttime water-leaving radiance is a function of the depth-dependent distribution of both the in situ bioluminescence emissions and the absorption and scattering properties of the water. The vertical distributions of these parameters were used as inputs for a modified one-dimensional radiative transfer model to solve for spectral bioluminescence water-leaving radiance from prescribed depths of the water column. Variation in the water-leaving radiance was consistent with local episodic physical forcing events, with tidal forcing, terrestrial runoff, particulate accumulation, and biological responses influencing the shorter timescale dynamics. In this paper we define a hierarchical Bayesian model for microarray expression data collected from several studies and use it to identify genes that show differential expression between two conditions. Key features include shrinkage across both genes and studies; flexible modeling that allows for interactions between platforms and the estimated effect, and for both concordant and discordant differential expression across studies. We evaluated the performance of our model in a comprehensive fashion, using both artificial data, and a "split-sample" validation approach that provides an agnostic assessment of the model's behavior not only under the null hypothesis but also under a …

In this dissertation, several microfabricated devices are introduced to develop new applications in the area of chemical analysis. Electrochemical micropumps, chip-based liquid chromatography systems and multilayer capillary electrophoresis microdevices with crossover channels were fabricated using various substrates such as poly(dimethylsiloxane) (PDMS), glass, and poly(methyl methacrylate) (PMMA). I have demonstrated pressure-driven pumping of liquids in microfabricated channels using electrochemical actuation. PDMS-based micropumps were integrated easily with channel-containing PMMA substrates. Flow rates on the order of ~10 µL/min were achieved using low voltages (10 V). The potential of electrolysis-based pumping in microchannels was further evaluated for pressure driven microchip liquid chromatography (LC). …

Background: The blood-brain barrier (BBB), blood-spinal cord barrier (BSCB), and blood-cerebrospinal fluid barrier (BCSFB) control cerebral/spinal cord homeostasis by selective transport of molecules and cells from the systemic compartment. In the spinal cord and brain of both ALS patients and animal models, infiltration of T-cell lymphocytes, monocyte-derived macrophages and dendritic cells, and IgG deposits have been observed that may have a critical role in motor neuron damage. Additionally, increased levels of albumin and IgG have been found in the cerebrospinal fluid in ALS patients. These findings suggest altered barrier permeability in ALS.
