Dentin is a mesenchymal tissue, and, as such, is based on a collagenous matrix that is reinforced by apatite mineral. Collagen fibrils show piezoelectricity, a phenomenon that is used by piezoresponse force microscopy (PFM) to obtain high-resolution images. We applied PFM to image human dentin with 10-nm resolution, and to test the hypothesis that zones of piezoactivity, indicating the presence of collagen fibrils, can be distinguished in dentin. Piezoelectricity was observed by PFM in the dentin intertubular matrix, while the peritubular dentin remained without response. High-resolution imaging of chemically treated intertubular dentin attributed the piezoelectric effect to individual collagen fibrils …

Stability Testing Of Mint Oils Through Gas Chromatography Analysis, Sarah Wolf

In this experiment, the stability of mint oil was measured through gas chromatography analysis. Eight different oils were tested. Levels of key components of mint oil, including menthol and menthone for peppermint oil, and carvone and limonene for spearmint oil, was compared over time. Several packaging materials were immersed in oils in order to determine their effects, if any, on the composition of the oils. Additionally, oil was subjected to either 25 degree or 40 degree Celsius temperatures to determine if temperature affected the oil composition.

Selenoproteins are an elite group of proteins containing a rare amino acid, selenocysteine (Sec), encoded by the codon, UGA. In eukaryotes, incorporation of Sec requires a Sec insertion sequence (SECIS) element, a stem–loop structure located in the 3’-untranslated regions of selenoprotein mRNAs. Here we report identification of a noncanonical form of SECIS element in Toxoplasma gondii and Neospora canine, single-celled apicomplexan parasites of humans and domestic animals. This SECIS has a GGGA sequence in the SBP2-binding site in place of AUGA previously considered invariant. Using a combination of computational and molecular techniques, we show that Toxoplasma and Neospora possess …

Irrigation of the municipal cemetery in Brigham City, Utah resulted in stained headstones in 2001 and 2002. The water used in the irrigation came from Mantua reservoir, a medium sized impoundment situated near the mouth of Box Elder Canyon. In order for Brigham City to establish a city wide secondary pressurized irrigation system using water from Mantua reservoir, the cause and the source of staining problem must be determined. Previous research (Wallace 2006) determined that the source of the staining was the reduction of iron found in Mantua Reservoir sediments that occurred when seasonal variations in the reservoir caused anaerobic …

The hPMS2 mutation E705K is associated with Turcot syndrome. To elucidate the pathogenesis of hPMS2-E705K, we modeled this mutation in yeast and characterized its expression and effects on mutation avoidance in mammalian cells. We found that while hPMS2-E705K (pms1-E738K in yeast) did not significantly affect hPMS2 (Pms1p in yeast) stability or interaction with MLH1, it could not complement the mutator phenotype in MMR-deficient mouse or yeast cells. Furthermore, hPMS2-E705K/pms1-E738K inhibited MMR in wild-type (WT) mammalian cell extracts or yeast cells only when present in excess amounts relative to WT PMS2. This thesis was completed in order to assess and document the environmental effects that are the result of elemental phosphorus production in Pinellas County Florida. The study utilized a collection of information resources that included: personal interviews, technical references, historical documents, legal documents and field observations. By utilizing five different sources of information a broad understanding of the problem was developed.

Valuing The Cultural Monuments Of Armenia: Bayesian Updating Of Prior Beliefs In Contingent Valuation, Anna Alberini, Alberto Longo

We use contingent valuation to place a value on the conservation of built cultural heritage sites in Armenia. When we present the hypothetical scenario in the questionnaire we spell out what would happen to the monuments in the absence of the government conservation program. We posit that respondents combine such information with their own prior beliefs, which the questionnaire also elicits, and that the WTP for the good or program is likely to be affected by these updated beliefs. We propose a Bayesian updating model of prior beliefs, and empirically implement it using the data from our survey. We find …

The past few years have seen a rapid expansion in interest in the characterization of the entire complement of proteins, or proteome. Micro total analysis systems (μTAS) are an emerging promising method, offering rapid, sensitive and low sample consumption separations. I have demonstrated microchip capillary electrophoresis (CE) devices made of CaF2. New methods have been developed for micromachining enclosed capillaries in CaF2. CE analysis of fluorescently labeled amino acids was used to illustrate bioanalytical applications of these microdevices. Initial on-chip infrared spectroscopy results for qualitative analyte identification were achieved in microfluidic CaF2 channels. I have also shown the evaluation of …

Up to 20% of children have a diagnosable mental disorder, but only a small percentage of them actually receive treatment. The developmental trajectory of psychopathology suggests that children who show early signs of behavioral or emotional problems are likely to experience a continual increase in those problems as they grow. Caregivers, attempting to seek assistance for their children, must often navigate a complex mental health maze, obtain funding for treatment and negotiate additional treatment barriers. School-based interventions can remove many of these potential treatment barriers. Gas hydrate formation was studied in Gulf of Mexico (GOM) sediments. Sediments studied were from six-meter long cores from Mississippi Canyon Block 118 and a 27-meter core from a cruise in 2002 of the Marion Dufresne. These sediments retained their in situ seawater before testing. Hydrate formation rate and induction times were measured. The hydrate memory effect was studied in GOM sediments with and without in situ seawater. Hydrate induction time was short when in situ seawater was present. Bioproducts adsorbed on particles in the sediments are postulated to shorten the induction times by maintaining seawater structuring around coated particles. …
