Selective Extraction Of Colorful Proteins Using Reverse Micelles, Loren E. Connell
Theses and Dissertations
Biotechnology is an ever expanding field of science. As the field has expanded, research into bioseparation processes has also expanded. The need for more cost effective ways to deliver products that are in high demand is greater now than ever. Bioseparation processes are a very costly part of the overall process of product development. While traditional separation process are effective, there is promise in many new techniques being developed which have the potential to lower the costs of bioseparations.
The continuous separation method of liquid-liquid extraction using reverse micelles is one such technique. While this technique has been researched in …

Cytosine methylation of repetitive sequences is widespread in plant genomes, occurring in both symmetric (CpG and CpNpG) as well as asymmetric sequence contexts. We used the methylation-dependent restriction enzyme McrBC to profile methylated DNA using tiling microarrays of Arabidopsis Chromosome 4 in two distinct ecotypes, Columbia and Landsbergerecta. We also used comparative genome hybridization to profile copy number polymorphisms. Repeated sequences and transposable elements (TEs), especially long terminal repeat retrotransposons, are densely methylated, but one third of genes also have low but detectable methylation in their transcribed regions. While TEs are almost always methylated, genic methylation is highly …

Crispr Recognition Tool (Crt): A Tool For Automatic Detection Of Clustered Regularly Interspaced Palindromic Repeats, Charles Band, Teresa L. Ramsey, Fareedah Sabree, Michael Lowe, Kyndall Brown, Nikos C. Kyrpides, Philip Hugenholtz
School of Computing: Faculty Publications
Background: Clustered Regularly Interspaced Palindromic Repeats (CRISPRs) are a novel type of direct repeat found in a wide range of bacteria and archaea. CRISPRs are beginning to attract attention because of their proposed mechanism; that is, defending their hosts against invading extrachromosomal elements such as viruses. Existing repeat detection tools do a poor job of identifying CRISPRs due to the presence of unique spacer sequences separating the repeats. In this study, a new tool, CRT, is introduced that rapidly and accurately identifies CRISPRs in large DNA strings, such as genomes and metagenomes.
Results: CRT was compared to CRISPR …

“Girls Play With Dolls And Boys Play With Soldiers”: Examining Teachers And Parents' Gender Beliefs And The Gender Identity Of 8-10 Year Old Jamaican Boys, Christopher Carlyle Clarke
USF Tampa Graduate Theses and Dissertations
This multi-case ethnographic study examined the gender beliefs of two teachers and 12 parents and the gender identity of thirty 8-10 year old boys in two primary schools in Jamaica. The study was conducted against the background of gross underachievement among Jamaican boys and the research literature pointing to gender socialization as a factor in the declining results and interest in academic studies. Through 10 weeks of observations, interviews and focus group discussions answers were sought for the following questions:
1. What beliefs do teachers hold about gender?
2. What beliefs do parents hold about gender?
3. What are boys' …

Restricted Cell Elongation In Arabidopsis Hypocotyls Is Associated With A Reduced Average Pectin Esterification Level, Paul Derbyshire, Maureen C. Mccann, Keith Roberts
Restricted Cell Elongation In Arabidopsis Hypocotyls Is Associated With A Reduced Average Pectin Esterification Level, Paul Derbyshire, Maureen C. Mccann, Keith Roberts
Department of Biological Sciences Faculty Publications
Background
Cell elongation is mainly limited by the extensibility of the cell wall. Dicotyledonous primary (growing) cell walls contain cellulose, xyloglucan, pectin and proteins, but little is known about how each polymer class contributes to the cell wall mechanical properties that control extensibility.
Results
We present evidence that the degree of pectin methyl-esterification (DE%) limits cell growth, and that a minimum level of about 60% DE is required for normal cell elongation in Arabidopsishypocotyls. When the average DE% falls below this level, as in two gibberellic acid (GA) mutantsga1-3 and gai, and plants expressing pectin methyl-esterase ( …
Host Resistance To Monilinia Vaccinii-Corymbosi In Flowers And Fruits Of Highbush Blueberry, Jeffrey Lehman
Host Resistance To Monilinia Vaccinii-Corymbosi In Flowers And Fruits Of Highbush Blueberry, Jeffrey Lehman
Biology and Earth Science Faculty Scholarship
Monilinia vaccinii-corymbosi, the causal agent of mummy berry disease, infects blueberry flowers via the gynoecial pathway. To describe the expression of host resistance in highbush blueberry (Vaccinium corymbosum), fungal growth in the styles and colonization of the locules were compared among five blueberry cultivars in a series of controlled greenhouse experiments. Styles were harvested 1 and 4 days postinoculation, and the length colonized by hyphae was determined using fluorescence microscopy. At 8 weeks after inoculation, fruit were harvested and scored for the presence of hyphae in the locules. The infection frequency of styles ranged from 0.33 to 0.71, and only …

It is recognized that ocean and coastal areas of the United States contribute significantly to our nation’s overall economy. What is not completely understood is the extent to and manner in which our economy benefits from the wide range of marine and coastal activities. One area of the ocean economy that has not been collectively measured or examined is the contribution of marine research and education institutions. The goal of the project was to create a prototype strategy at a local level for collecting data at a national level, in order to create an economic sector of these institutions and …

Considering The Community: How One Rural Superintendent Perceives Community Values And Their Effect On Decision-Making, Chris Jenkins
Considering The Community: How One Rural Superintendent Perceives Community Values And Their Effect On Decision-Making, Chris Jenkins
The Rural Educator
In rural Oklahoma, the role of the superintendent is often vastly different than that of superintendents in large cities. The superintendent is the leader of the school district, which is typically the community’s largest employer. There are a few examples of superintendents who embrace this sometimes overwhelming responsibility and who are often seen as pillars of ethics, leadership and morality in the community. Dale Carter is one example of this type of superintendent. Mr. Carter has been employed by Kenawee Public Schools since 1969. During his tenure he has been a teacher, coach, principal and superintendent. This case study will …

All Faculty Scholarship for the College of the Sciences
Background
The number of completely sequenced plastid genomes available is growing rapidly. This array of sequences presents new opportunities to perform comparative analyses. In comparative studies, it is often useful to compare across wide phylogenetic spans and, within angiosperms, to include representatives from basally diverging lineages such as the genomes reported here: Nuphar advena (from a basal-most lineage) and Ranunculus macranthus (a basal eudicot). We report these two new plastid genome sequences and make comparisons (within angiosperms, seed plants, or all photosynthetic lineages) to evaluate features such as the status of ycf15 and ycf68 as protein coding genes, the distribution …

Utility And Versatility Of Extracellular Recordings From The Cockroach For Neurophysiological Instruction And Demonstration, Raddy L. Ramos, Andrew Moiseff, Joshua C. Brumberg
Publications and Research
The principles of neurophysiology continue to be challenging topics to teach in the context of undergraduate neuroscience education. Laboratory classes containing neurophysiological demonstrations and exercises are, therefore, an important and necessary complement for covering those subjects taught in lecture-based courses. We developed a number of simple yet very instructive exercises, described below, which make use of extracellular recordings from different sensory systems of the cockroach (Periplanta americana). The compendium we developed provides students with hands-on demonstrations of several commonly taught topics of neurophysiology including sensory coding by neural activity.
Collapse Analysis Of Nanofibers, Xiang-Fa Wu, Yuris A. Dzenis
Collapse Analysis Of Nanofibers, Xiang-Fa Wu, Yuris A. Dzenis
Department of Engineering Mechanics: Faculty Publications
Continuous nanofibers fabricated by the electrospinning technique have found increasing applications (e.g., nanofiber composites, nanofiber devices, bioengineering tissue scaffolding, etc.). For a nanofiber network subjected to a small external perturbation, the fiber segments within the network may deflect and stick to each other under the condition that their surface adhesion energy overcomes the elastic strain energy induced by fiber bending. Therefore, this paper aims to study adhesion-induced nanofiber collapse and relevant criteria. A simple fiber collapse model was proposed, which is based on the contact of two deflected elastic filaments under surface adhesion. Four fundamental fiber collapse modes (i.e., fiber-flat …
Alternative Targets For The Treatment Of Stroke, Craig T. Ajmo Jr.
Alternative Targets For The Treatment Of Stroke, Craig T. Ajmo Jr.
USF Tampa Graduate Theses and Dissertations
Stroke is cerebrovascular injury that has been reported to be the third leading cause of death and the first leading cause of disability in the world (W. H.O. 2007). Currently, there is only one FDA approved treatment for stroke which is recombinant tissue plasminogen activator. This treatment has a narrow therapeutic window of three hours after ischemic stroke and can adversely cause the production of oxygen free radicals and intracranial hemorrhage. These limitations result in only 2-3% of all stroke victims as being candidates for this therapy as many patients do not arrive at the hospital in time to receive …
