The fosfomycin resistance protein, FosX, catalyzes the hydration of the antibiotic fosfomycin, (1R,25)-epoxypropylphosphonic acid. Genes encoding the enzyme are found in several pathogenic microorganisms. The structure and mechanism of action of the genomically encoded FosX enzyme from Listeria monocytogenes (FosXLMATCC) obtained from the American Type Culture Collection are reported. The gene harbors 31 point mutations, and as a consequence, the protein differs in 10 amino acid residues from the previously reported FosX encoded in the genome of the EGD strain of L. monocytogenes (FosXLMEGD). The FosXLMATCC enzyme is shown to catalyze the addition of water to the C1 position of …

Synthesis And Antiviral Evaluation Of Some 3'-Carboxymethyl-3'-Deoxyadenosine Derivatives, Houguang Shi

3'-Carboxymethyl-3'-deoxyadenosine derivatives were prepared from 2'-O-TBDMS-3'-deoxy-3'-[(ethoxycarbonyl)methyl]adenosine (1) via simple and efficient procedures. Conversion of 1 to 5'-azido-2'-O-TBDMS-3', 5'-dideoxy -3'-[(ethoxycarbonyl) methyl] adenosine (4) was accomplished via a novel one-pot method employing 5'-activation (TosCl) followed by efficient nucleophilic displacement with tetramethylguanidinium azide. Compound 4 was converted to a 5'-[(N-methylcarbamoyl)amino] derivative (5) via one-pot reduction/acylation employing H2/Pd-C followed by treatment with p-nitrophenyl N-methylcarbamate. The latter step of this two-step process required an efficient source of p-nitrophenyl N-methylcarbamate, thus a highly efficient new method for preparing p-nitrophenyl N-alkylcarbamate was developed. N6-phenylcarbamoyl groups were introduced by treatment with phenylisocyanate, and an efficient new method for …

Cigarette smoke's influence on cancer has primarily been a subject of epidemilogic and tumorigenic studies. There have been no proper investigations with interests focused on how cigarette smoke affects the cellular mechanics of metastasis. Gathering an understanding of how smoke influences metastatic invasion could be vital in regulating or possibly eliminatings cancer's ability to initiate new tumor growth sites. This project focuses on cigarette smoke's influence on cellular mechanics of endothelial cells, and the invasive potential of cancer against a fully active endothelium. In carrying out NASA’s Vision for Space Exploration, a number of different environments will be encountered that will require the crew to wear a protective space suit. Specifically, four suited mission phases are identified as Launch, Entry & Abort profiles, Contingency 0g (orbital) Extravehicular Activity (EVA), Lunar Surface EVA and Martian Surface EVA. This study presents conceptual design solutions based on a previous architecture assessment that defined space suit operational requirements for four proposed space suit configuration options. Description Of The Tadpole Of Leptodactylus Pustulatus Peters, 1870 (Anura: Leptodactylidae), Rafael O. De Sá, Reuber Brandão, Lorena Dall'ara Guimarães

The external morphology and oral disc of the tadpole of Leptodactylus pustulatus Peters, 1870, are described and illustrated for Gosner stage 39. The internal oral anatomy was analyzed with SEM at Gosner stage 36 whereas chondrocranial anatomy is reported for Gosner stage 35. The overall characteristics do not depart from those known for the genus Leptodactylus and they particularly agree for those of the melanonotus species group. The labial tooth row formula is 2(2)/3. Observations are presented that suggest that L. pustulatus engages in larval parental care.

Previous articles (van der Laan and Dudoit (2003); van der Laan et al. (2006); Sinisi et al. (2007)) advertised and theoretically validated the use of cross-validation to select among many candidate estimators to compute a so called super learner which outperforms any of the given candidate estimators. The theoretical basis was provided for this super learner based on oracle results for the cross-validation selector (e.g., van der Laan and Dudoit (2003); van der Laan et al. The identification of soybean rust, caused by Phakopsora
pachyrhizi H. Sydow & Sydow, in the southern United
States in November 2004 (Schneider et al. 2005 ), in the
Midwest in 2006 (Hartman et al. 2007 ), and elsewhere has
increased the concerns of the impact of the pathogen on US
soybean production (Miles et al. 2007 ). The rapid spread of
P. pachyrhizi and its potential to cause severe yield losses
makes this among the most destructive foliar diseases of
soybean. Yield losses of 20%–60% were reported in Asia,
with losses of 80% reported from experimental plots in

Previously we have shown that nanosized drug carriers called Plurogels™ sequestered Doxorubicin (Dox) and partially released this drug with application of ultrasound (US) (Fig. 1)1. The application to chemotherapy was successfully demonstrated in a rat tumor model2. However previous studies did not examine the distribution of Dox in the insonated and control tissue. This recent work investigated the effect of two US frequencies and examined the Dox distribution in the insonated and control tumors to determine if US was depositing more Dox at the insonated site.

Background
Functional and comparative studies of insect genomes have shed light on the complement of genes, which in part, account for shared morphologies, developmental programs and life-histories. Contrasting the gene inventories of insects to those of the nematodes provides insight into the genomic changes responsible for their diversification. However, nematodes have weak relationships to insects, as each belongs to separate animal phyla. A better outgroup to distinguish lineage specific novelties would include other members of Arthropoda. For example, crustaceans are close allies to the insects (together forming Pancrustacea) and their fascinating aquatic lifestyle provides an important comparison for understanding the …

Unmanned Air Vehicles (UAVs) have recently gained attention due to their increased ability to perform sophisticated missions with less cost and/or risk than their manned counterparts. This thesis develops approaches to the use of compliant mechanisms in the design of deployable wings for small UAVs. Although deployable wings with rigid-link mechanisms have been used in the past to maintain flight endurance while minimizing required storage volume, compliant mechanisms offer many advantages in manufacturability and potential space savings due to function sharing of components. A number of compliant, deployable wing concepts are generated and a classification system for them is formed. …

Recent technological advances in fields like medicine and genomics have produced high-dimensional data sets and a challenge to correctly interpret experimental results. The Optimal Discovery Procedure (ODP) (Storey 2005) builds on the framework of Neyman-Pearson hypothesis testing to optimally test thousands of hypotheses simultaneously. The method relies on the assumption of normally distributed data; however, many applications of this method will violate this assumption. This thesis investigates the sensitivity of this method to detection of significant but nonnormal data. Overall, estimation of the ODP with the method described in this thesis is satisfactory, except when the nonnormal alternative distribution has …

The City of Portsmouth (City) is located in Rockingham County at the mouth of the Piscataqua River in the seacoast area of New Hampshire. The City of Portsmouth’s sewerage system consists of approximately 115 miles of sewers (excluding the Pease International Tradeport), 19 pumping stations, and a 4.8 million gallon per day (mgd) primary wastewater treatment facility (WWTF) located on Peirce Island. The City has an inter-municipal agreement with the Town of New Castle, an island to the northeast of Portsmouth, to treat their wastewater at the City’s Peirce Island WWTF. BACKGROUND: Researchers using RNA expression microarrays in experimental designs with more than two treatment groups often identify statistically significant genes with ANOVA approaches. However, the ANOVA test does not discriminate which of the multiple treatment groups differ from one another. Thus, post hoc tests, such as linear contrasts, template correlations, and pairwise comparisons are used. Linear contrasts and template correlations work extremely well, especially when the researcher has a priori information pointing to a particular pattern/template among the different treatment groups. Further, all pairwise comparisons can be used to identify particular, treatment group-dependent patterns of gene expression. However, these approaches …

The importance of restoring filter-feeders, such as the Eastern oyster Crassostrea virginica, to mitigate the effects of eutrophication (e.g. in Chesapeake Bay) is currently under debate. The argument that bivalve molluscs alone cannot control phytoplankton blooms and reduce hypoxia oversimplifies a more complex issue, namely that ecosystem engineering species make manifold contributions to ecosystem services. Although further discussion and research leading to a more complete understanding is required, oysters and other molluscs (e.g. mussels) in estuarine ecosystems provide services far beyond the mere top-down control of phytoplankton blooms, such as (1) seston filtration, (2) benthic–pelagic coupling, (3) creation of refugia …
