Despite several investigations, the transcriptional mechanisms that regulate the expression of both type I collagen genes (COL1A1 and COL1A2) in either physiological or pathological situations, such as scleroderma, are not completely known. We have investigated the role of hc-Krox transcription factor on type I collagen expression by human dermal fibroblasts. hc-Krox exerted a stimulating effect on type I collagen protein synthesis and enhanced the corresponding mRNA steady-state levels of COL1A1 and COL1A2 in foreskin fibroblasts (FF), adult normal fibroblasts (ANF), and scleroderma fibroblasts (SF). Forced hc-Krox expression was found to up-regulate COL1A1 transcription through a -112/-61-bp sequence in FF, ANF, …

The Use Of Non-Natural Nucleotides To Probe Template-Independent Dna Synthesis, Anthony J. Berdis, David Mccutcheon
The Use Of Non-Natural Nucleotides To Probe Template-Independent Dna Synthesis, Anthony J. Berdis, David Mccutcheon
Chemistry Faculty Publications
The vast majority of DNA polymerases use the complementary templating strand of DNA to guide each nucleotide incorporation. There are instances, however, in which polymerases can efficiently incorporate nucleotides in the absence of templating information. This process, known as translesion DNA synthesis, can alter the proper genetic code of an organism. To further elucidate the mechanism of template-independent DNA synthesis, we monitored the incorporation of various nucleotides at the “blunt-end” of duplex DNA by the high-fidelity bacteriophage T4 DNA polymerase. Although natural nucleotides are not incorporated at the blunt-end, a limited subset of non-natural indolyl analogues containing extensive π-electron surface …

Differential Response Of Various Spore Species To Sporicidal Disinfectants, Michael David Pratt
Differential Response Of Various Spore Species To Sporicidal Disinfectants, Michael David Pratt
Theses and Dissertations
In the fall of 2001, letters laced with anthrax spores were delivered to various news organizations in New York and Florida, as well as to two Senators in Washington, D.C. Over 22 anthrax infections and five deaths resulted from exposure to these spores, and decontamination of the affected buildings was both time consuming and costly. Since these attacks, interest in sporicidal disinfectants has increased greatly. Many chemical sporicidal disinfectants are available commercially, but the exposure time required to sterilize can be relatively long. In addition, some spores are simply injured or inhibited by chemical disinfectants, but not necessarily killed. Studies …

Continuous Simulation Of Groundwater Use And Effluent Discharge In Catfish (Ictalurus Punctatus) Ponds At Five Locations In The Southeast U.S, Sugeng Triyono
Continuous Simulation Of Groundwater Use And Effluent Discharge In Catfish (Ictalurus Punctatus) Ponds At Five Locations In The Southeast U.S, Sugeng Triyono
Theses and Dissertations
Long-term climatological data were used to evaluate the effectiveness of a drop/add management strategy to reduce groundwater use and effluent discharge in catfish ponds in the southeast U.S.. A drop/add approach is based on the creation of a storage volume in the pond for rainfall collection. The storage volume is created by allowing water level in the pond to decrease until some minimum level is reached. When the minimum level is reached, the pond is partly refilled, leaving the remaining volume available to capture incident precipitation. In this way, the role of precipitation in the water budget is increased. In …

The Role Of A Non-Microtubule-Based Spindle Matrix In Eukaryotic Cellular Division, Melissa Gwen Chambers
The Role Of A Non-Microtubule-Based Spindle Matrix In Eukaryotic Cellular Division, Melissa Gwen Chambers
Theses and Dissertations
A non-microtubule-based spindle matrix has been observed in Drosophila melanogaster meiotic and somatic cells. This matrix comprises the proteins: Skeletor, Megator and Chromator, which localize to the nuclear periphery and chromosomes at prophase and to the microtubule spindle during mitosis and meiosis. This matrix might play a role in nuclear organization and microtubule assembly and stabilization. It is important to determine the presence or absence of this matrix in other species in order to compare form and function to that in Drosophila. These matrix proteins were studied in both normally dividing cells and cells in which division was disrupted. Our …

Economic Impacts Of White-Tailed Deer From Hunting In Mississippi, Micah Whittington Whiteside
Theses and Dissertations
White-tailed deer (Odocoileus virginianus) are an important ecological, social, and economic resource in Mississippi. Studies have been conducted on expenditures by white-tailed deer hunters, but none have administered a research-based, economic impact assessment for white-tailed deer in Mississippi. The economic impacts and associated values of white-tailed deer in Mississippi through a selfministered mail questionnaire were examined to collect white-tailed deer hunter expenditure data during the 2003/2004 hunting season. Expenditures of white-tailed deer hunters were obtained from a mail survey (N = 1,257, 38.6% response rate) and were used in an input-output model to determine economic impacts for the State. Economic …

Nmr Structure And Relaxation Studies Of Dhfr From Haloferax Volcanii At High Salt, Bulent Binbuga
Nmr Structure And Relaxation Studies Of Dhfr From Haloferax Volcanii At High Salt, Bulent Binbuga
Theses and Dissertations
The studies of enzymes from extreme sources have gained significance due to their increasing potential applications. The proteins from halophiles (salt loving) have adapted to challenging environmental conditions and require salt for their structure and function. How halophilic proteins adapt to a hypersaline environment is still an intriguing question. It is important to mimic the environmental conditions of the sample under investigation with experimental techniques. In this study, structure and dynamic features of a halophilic enzyme have been investigated under high salt conditions. The acquisition of NMR data on high salt samples has always been problematic. We have devised a …
Proteome And Phosphoproteome Dynamic Change During Cell Dedifferentiation In Arabidopsis Thaliana, Brahmananda Reddy Chitteti
Proteome And Phosphoproteome Dynamic Change During Cell Dedifferentiation In Arabidopsis Thaliana, Brahmananda Reddy Chitteti
Theses and Dissertations
Cell dedifferentiation is a cell fate switching process in which a differentiated cell reverts to a status with competence for cell division and organ regeneration like an embryonic stem cell. Although the phenomenon of cell dedifferentiation has been known for over two and a half centuries in plants, little is known of the underlying mechanisms. Here, the proteome map of Arabidopsis cotyledons has been established and investigated the dynamic change of the cotyledon proteome in the time course of cell dedifferentiation. Among the 353 distinct genes, corresponding to 500 2-DE gel protein spots identified with high confidence, 12% have over …

Influence Of Nursery Stock, Planting Practices, Fertilization And Competition Control On Initial Survival And Growth Of Nuttall And White Oak Seedlings, Joshua Larue Moree
Theses and Dissertations
This project evaluates the survival and initial growth response of oak seedlings produced under special nursery protocols (?enhanced? seedlings) compared to nursery-run oak seedlings of the same species, and the influence of various cultural practices on these same nursery stocks. Survival did not differ significantly between nursery stocks. Significant differences in survival among competition control treatments did occur. Intensive competition control regimes using glyphosate can negatively impact seedling survival if seedlings come in contact with herbicide spray drift.Nursery-run seedlings had significantly greater growth and many were larger than enhanced seedlings after two growing seasons. Height growth was typically greater with …

Systematics Of Holarctic Teleiodini (Lepidoptera : Gelechiidae), Sangmi Lee
Theses and Dissertations
Phylogenetic relationships of 25 genera of Holarctic Teleiodini (Gelechiidae) are postulated based on morphology and molecular characters, including CO?I, CO?II, and 28S genes. The phylogenetic analysis of the morphology matrix yielded six equal most parsimonious trees (length 329 steps, CI = 0.38, RI = 0.53) and a strict consensus tree (length 342 steps, CI = 0.36, RI = 0.50) with two polytomies and two trichotomies. The phylogenetic analysis of the combined morphology matrix and the CO?I + CO?II + 28S matrix yielded two equally most parsimonious trees (length 1184 steps, CI = 0.50, RI = 0.41) and a strict consensus …
The Ecology And Morphological Variation Of Opuntia (Cactaceae) Species In The Mid-South, United States, Lucas C. Majure
The Ecology And Morphological Variation Of Opuntia (Cactaceae) Species In The Mid-South, United States, Lucas C. Majure
Theses and Dissertations
Opuntia species have been poorly studied ecologically and taxonomically in the eastern United States. This study deals with the ecology of Opuntia species in the mid-south United States and covers not only the high degree of morphological variation exhibited by taxa, but also the taxonomy and distributions of the group for Mississippi. The taxa in the mid-south have distinct habitat preferences and can be separated based on habitat characterization. Information from this work provides valuable data useful in predicting possible routes that an invasive species, Cactoblastis cactorum (the cactus moth), might use in its potential westward migration. Phenotypic plasticity exhibited …
The Effects Of Changing Technology On Average Costs For Mississippi Cotton Producers: From 1996 - 2005, William Michael Thompson Ii
The Effects Of Changing Technology On Average Costs For Mississippi Cotton Producers: From 1996 - 2005, William Michael Thompson Ii
Theses and Dissertations
Row crop production can be characterized by constant change. Agricultural technology is responsible for most of the changes in productivity observed at the field level since the advent of mechanized farm equipment. Genetically modified (GM) cotton varieties have changed many aspects of cotton production in the United States. The advent of GM varieties has been the source of altered cropping practices in cotton production. The rapid adoption of GM cotton varieties in Mississippi has allowed producers to alter certain aspects of their farming operation because of added flexibility, increased yields, and other benefits of GM varieties. This study analyses the …

The fatty …
The Influence Of Probiotic Lactobacilli On Glucose Uptake By Caco-2 Cells, Arun Kumar Rooj
The Influence Of Probiotic Lactobacilli On Glucose Uptake By Caco-2 Cells, Arun Kumar Rooj
Theses and Dissertations
The resident microbes of human gastrointestinal tract cause both harmful and beneficial effects and these effects can be modulated by the administration of beneficial probiotic bacteria. Probiotics attribute several therapeutic and preventive beneficial effects, for both humans and animals. Despite the good effects of probiotic bacteria, the role of probiotic bacteria or their metabolites on the nutrient uptake by enterocytes is very less known. Most studies describe the genomic effects of probiotic bacteria on the transport properties. This thesis describes the short term (10 min or less) non-genomic effects of probiotic bacteria on the glucose uptake by human enterocytes like …
The Metaphase Checkpoint In Cells Undergoing Mitosis Without Chromosome Duplication, Mary Kathrine Johnson
The Metaphase Checkpoint In Cells Undergoing Mitosis Without Chromosome Duplication, Mary Kathrine Johnson
Theses and Dissertations
Chinese hamster ovary cells (CHO) were arrested with hydoxyurea at the beginning of DNA synthesis. Subsequent treatment with caffeine induced cells to bypass S-phase and undergo mitosis with unreplicated genomes (MUG). Treated cells built a normal spindle and distributed unattached kinetochores to daughter cells. To determine if MUG cells obey the metaphase checkpoint, we used immunoflourescence to detect and localize known metaphase checkpoint and motor proteins. In addition, the drug taxol was used to stabilize microtubules in MUG cells. The localization of CENP- E, the presence of anaphase A, taxol arrest, and taxol release acted in a similar manner as …
