The Characterization Of Trna Modifying Enzymes S-Adenosylmethionine : Trna Ribosyltransferase-Isomerase (Quea) And A Novel Type I Gtp Cyclohydrolase, Shilah Amal Bonnett

Queuosine is a hypermodified nucleoside located in the wobble position of bacterial and eukaryotic tRNAs coding for Asp, Tyr, His and Asn. The biosynthesis involves the participation of S-adenosyl-methionine:tRNA ribosyltransferase-isomerase (QueA) and a GTP Cyclohydrolase-I. QueA catalyzes the transfer and isomerization of the ribosyl moiety from AdoMet to preQ1 modified tRNA. Substrate analogs of AdoMet were used to elucidate important substrate-enzyme interactions and to test key steps in the proposed chemical mechanism. Replacing AdoMet with SeAdoMet had little effect upon substrate binding but exhibited 30-fold reduction in kcat, consistent with deprotonation at C-5' as the first catalytic step. 7-deazaAdoMet failed …

Interactions Across Spatial Scales Among Forest Dieback, Fire, And Erosion In Northern New Mexico Landscapes, Craig D. Allen

Ecosystem patterns and disturbance processes at one spatial scale often interact with processes at another scale, and the result of such cross-scale interactions can be nonlinear dynamics with thresholds. Examples of cross-scale pattern-process relationships and interactions among forest dieback, fire, and erosion are illustrated from northern New Mexico (USA) landscapes, where long-term studies have recently documented all of these disturbance processes. For example, environmental stress, operating on individual trees, can cause tree death that is amplified by insect mortality agents to propagate to patch and then landscape or even regional-scale forest dieback. Microarray time course (MTC) gene expression data are commonly collected to study the dynamic nature of biological processes. One important problem is to identify genes that show different expression profiles over time and pathways that are perturbed during a given biological process. While methods are available to identify the genes with differential expression levels over time, there is a lack of methods that can incorporate the pathway information in identifying the pathways being modified/activated during a biological process. The present invention provides a mutant oligonucleotide composition encoding a cellular c-Src tyrosine kinase oncogene. Methods for isolating, expressing and characterizing recombinant Src mutant polypeptide are also provided. The invention further relates to methods for utilizing such oligonucleotides, polypeptides, agonists and antagonists for applications, which relate to research, diagnostics, and clinical arts. More specifically, this invention provides methods of diagnosing, treating, immunizing, and creating transgenic animals based on use of such mutant Src.

Age Determination From Biological Stains Using Messenger Rns Profiling Analysis, John Ballantyne

In the present work, we report the development of two reverse transcription-quantitative polymerase chain reacton (RT-qPCR) assays for the definitve identification of newborn individuals. The methodology is based upon gene expresiion profiling analysis in which specific age groups are identified by detecting the relative quantity of expression of appropriate mRNA species. RNA is isolated from dried bloodstains is analyzed using the two duplex real-time PCR reactions for two variant novel forms of newborn specific gamma hemoglobin.

The eukaryotic nicotinamide riboside kinase (Nrk) pathway, which is induced in response to nerve damage and promotes replicative life span in yeast, converts nicotinamide riboside to nicotinamide adenine dinucleotide (NAD+) by phosphorylation and adenylylation. Crystal structures of human Nrk1 bound to nucleoside and nucleotide substrates and products revealed an enzyme structurally similar to Rossmann fold metabolite kinases and allowed the identification of active site residues, which were shown to be essential for human Nrk1 and Nrk2 activity in vivo. Although the structures account for the 500-fold discrimination between nicotinamide riboside and pyrimidine nucleosides, no enzyme feature was identified to recognize …
