Estuaries play a key role in the juvenile stage of gag (Myeteroperca microlepis). The use of estuarine habitats by juvenlle gag has been examined in temperate estuaries, which are at the northern limits of the range of this species, but the importance of subtropical estuaries during the early life history of this species has not been studied extensively. Gag were collected in subtropical Charlotte Harbor, Florida, during routine monthiy sampling from January 1996 to Decemher 2002. Juvenlle gag were collected using a 21.3-m seine, a 183-m haul seine, and a 183-m purse seine. A total of 738 individuals …

The Southeast Fisheries Science Center (SEFSC) of the US NOAA Fisheries provides much of the US science advice for the management of Highly Migratory Species (HMS) in the Atlantic, Gulf of Mexico and US Caribbean Sea. These stocks include the tunas, sharks, billfishes and swordfish all of which are targeted either commercially or recreationally or both in US waters. These species are managed internationally through membership to the International Commission for the Conservation of Atlantic Tunas (ICCAT); however, while the US is an ICCAT member, few Caribbean nations are members.

On 26 June 2006 an aggregation of 16 whale sharks was observed for a period of 4 hr in the north central Gulf of Mexico (GOM). The sharks remained within an area about 1.0 km² in size and continuously ram filter fed at the surface. Visual analysis of a plankton sample collected from the study site revealed the presence of copious amounts of fish eggs in mid-embryonic development and a minor amount of other zooplankton. A second plankton sample (control) collected about 3.5 km from the study site in an area where no whale sharks were present contained few …

Swordfish are known to undergo large diel vertical movements from surface waters at night to > 300 m depth during the day. Evidence presented over the past several deeades suggests the lunar cycle affects these vertical migrations. This study collected data concurrently from 7 swordfish throughout 3 consecutive lunar cycles using pop-up satellite archival tags. All individuals demonstrated an inverse relationship between recorded nocturnal depths and lunar illumination.

The genetic basis of stock structure of blue marlin (Makaira nigricans Lacepède 1802) in the Atlantic ocean was inferred from analyses of mitochondrial control region sequences. Blue marlin were collected in 1998 from 4 major geographic locations: western North Atlantic, Caribbean Sea, western South Atlantic and eastern Atlantic. Haplotype diversity (h) ranged from 0.99-1.0 and nucleotide sequence diversity (π) ranged from 0.11-0.13 within samples indicating that the control region harbors a significant amount of genetic variation. We examined data from pop-up archival transmitting (PAT) tags (n = 18) to characterize aspects of vertical habitat use by blue marlin (Makaira nigricans) from the Gulf of Mexico (GOM). Two of these tags were recovered and provided fine-scale information about diving patterns and the relationship between time at depth and temperature. Similar to previous studies, blue marlin in the GOM spent most of their time at the surface and at temperatures within 3° C of surface temperatures. Isolated and sparse reef atolls comprising the southern section of the Seaflower MPA have been traditionally exploited by artisanal fishers using handlines. These practices have severely depleted the demersal fish stock and now fishers have shifted their efforts to the pelagic stock. The pelagic fishery included the extraction of more than 25 species, with Thunnus atlanticus, Acanthocybium solandri, Coryphaena hippurus, and Katsuwonus pelamis being the most common. Three years of fishery-dependent data (2004–2006) were used to describe traditional fishing methods and techniques, and to document, for the first time, a declining trend in the large pelagic stock. …

Human papillomavirus type 16 (HPV-16) infections can in rare cases persist and cause lesions that may progress to cervical cancer. Cells in the lesions are not permissive for virus production, nor are cervical cancer cells. The intracellular environment is such that it prevents production of the highly immunogenic, viral structural proteins L1 and L2. One may speculate that inhibition of L1 and L2 expression is a prerequisite for persistence and cancer progression.We have therefore investigated how expression of HPV-16 L1 is regulated.We found that the only splice site in the HPV-16 late region, which is used to produce L1 mRNAs, …

Background and aims: Tissue transglutaminase (tTG) is an autoantigen in coeliac disease and the related disorder, dermatitis herpetiformis. The detection of autoantibodies directed against tTG is a highly specific marker of coeliac disease; however, it is unclear if there is a role for these autoantibodies in the disease process. The aim of this study was to investigate whether the catalytic triad of tTG is targeted by coeliac disease autoantibodies. Methods: A full-length wild-type recombinant tTG and a novel site-directed mutagenic variant lacking the catalytic triad were produced in Escherichia coli. Male frogs typically call near water at dusk to attract females for breeding. During the breeding season, male treefrogs also emit diurnal "tree calls" or "rain calls" from the tops of trees. Very little is known about tree calls, although many treefrogs use them. Tree calls may be used to attract females, deter males or be triggered by weather conditions favorable for breeding: high temperature and relative humidity, and a drop in barometric pressure. In this thesis, we examine epidemiological models of two different retroviruses, which infect the human body. The two viruses under study are HIV or the human immunodefiency virus and HTLV-I, which is the human T lymphotropic virus type I. A retrovirus is a virus, which injects its RNA into the host, rather than it's DNA. We will study each of the different mathematical models for each of the viruses separately. Quantification of microbial processes involved in plant decomposition is important in understanding energy flow and nutrient recycling in wetlands. Prior studies examining emergent plant decomposition have often used litter that had been prematurely harvested (senescent) and/or manipulated (oven-dried). I examined the effects of litter manipulations on microbial decay dynamics associated with litter of the emergent macrophyte Typha angustifolia. Plant litter was harvested after senescence and after a period of standing-dead decay. This collected litter was either air-dried or oven-dried, placed into fine mesh (1 mm) litterbags, and submerged in the wetland. Environmental exposure to silicate compounds such as silica and asbestos has been associated with increased autoimmune responses and the development of autoimmune disease in humans. Residents of Libby, MT have experienced significant asbestos exposure due to an asbestos contaminated vermiculite mine near the community over several decades. Residents have developed numerous asbestos-related diseases as well as increased autoimmune responses. However, the exact mechanism by which Libby amphibole asbestos generates autoimmune responses is unclear. Acyl-CoA thioesterases (ACOTs) catalyze the hydrolysis of acyl-CoAs to free fatty acids and coenzyme A. Recent studies have demonstrated that one gene named Acot7, reported to be mainly expressed in brain and testis, is transcribed in several different isoforms by alternative usage of first exons. Strongly decreased levels of ACOT7 activity and protein in both mitochondria and cytosol was reported in patients diagnosed with fatty acid oxidation defects, linking ACOT7 function to regulation of fatty acid oxidation in other tissues. This paper presents a comprehensive review of the use of different sources of calcium to preserve fresh fruits and vegetables in order to extend the shelf life and enhance the nutritional value. Moisture content, shrinkage, water activity (Aw), color and texture of cooked chickpeas and soybeans during convective, microwave and combined microwave–convective drying were studied. Combined drying was significantly (P < 0.05) faster than either convective or microwave drying, and resulted in less shrinkage of the dehydrated product. Rapid burning occurred when samples were dried below a Aw of 0.27 ± 0.07 for chickpeas (P < 0.05), and 0.13 ± 0.04 for soybeans (P < 0.05). Both chickpeas and soybeans displayed a transitional behavior in texture when dried to a Aw below 0.40 ± 0.10 (P < 0.05) for chickpeas, and below 0.63 ± 0.15 (P < 0.05) for soybeans, when samples became brittle. Shelf stable dehydrated chickpea and soybean products with low water activity (Aw = 0.35) and good visual quality could be obtained within 14 min of combination drying.

Horseradish Peroxidase (HRP) is a commonly used enzyme in many biotechnological fields. Improvement of HRP stability would further increase its potential application range. In the present study, thirteen single- and three double- mutants of solvent exposed, proximal lysine and glutamic acid residues were analysed for enhanced H2O2 stability. Additionally, five single- and one pentuple-consensus mutants were investigated. Most mutants displayed little or no alteration in H2O2 stability; however, three (K232N, K241F and T110V) exhibit significantly increased H2O2 tolerances of 25- (T110V), 18- (K232N), and 12- fold (K241F). This improved stability may be due to an altered enzyme-H2O2 catalysis pathway or …

Comparison of bioinformatic data is a common application in the life sciences and beyond. In this communication, a novel Java based software tool, ProteinParser, is outlined. This soft- ware tool calculates a detailed consensus, or most common, amino acid at a given position in an aligned protein set, whilst also generating a full consensus protein FASTA output. A second application of this software tool, computing a consensus amino acid given a toler- ance threshold, is also demonstrated. This Comment contextualizes the issue of polygamous marriages within the South African constitutional paradigm, one committed unequivocally to the principle of equality. This Comment analyzes how South African law, European in origin, had to incorporate the laws and institutions of indigenous communities within the national legal framework, as part of the overall transformative legal project underway in the country since 1994. By focusing on the Recognition of Customary Marriages Act, this Comment examines such incorporation, while questioning its effect on the overall project of constitutionalism, human rights, and equality.
