Currently, it is perceived that brook trout (Salvelinus fontinalis ) are experiencing reductions in both range and abundance across their east coast distribution. Data identifying key habitat features influencing growth rates of these fish will be valuable to managers conducting remediation efforts as growth at age has important consequences to survival and fecundity. Of particular importance within Appalachia may be the influence of surficial geology as the mid Appalachian region receives some of the most acidic rain in the nation. Knowledge of the interaction between proteins is necessary for understanding how the cellular machinery operates. Multiple methods have been developed to study protein-protein interaction, but only two have been able to provide large amount of information for S. cerevisae protein-protein network. Due to the multiple spurious data coming from these methods, there is a need of new complementary methods to improve confidence of the interactions. A new method is developed in this dissertation where the protein complexes are pulled down (immunoprecipitated) using tagged single chain antibodies specific for one of the proteins. The enantioselective syntheses of digitoxigen 2,6-dideoxy-beta-L-ribo-hexopyranoside, its analogues and digitoxin monosaccharide analogues have been achieved. Key to this approach is the iterative application of the palladium-catalyzed glycosylation reaction, reductive 1,3-transposition and diastereoselective dihydroxylation. Anticancer tests showed that these analogues have even better anticancer activities.

Women are fewer than men in science and engineering, therefore, the assumption is that women generally prefer arts and humanities. But to some analysts, the reason for the low percentage of women in these careers may be due to innate mental and psychological differences between them and men. Others contend that women have the capacity to excel in any profession and that their fewness in science and engineering can be attributed to other factors. Ischemic tissue and organ dysfunction occur in a variety of cardiovascular diseases. The hypoxic environment created during ischemic injury activates cytoprotective genes, of which heme oxygenase-1 (HO-1) is of great interest. This microsomal inducible enzyme is involved in the homeostasis of prooxidant heme, which is released from heme proteins upon injury. HO-1 responds to a variety of oxidative stress associated stimuli. HO-1 expression has been implicated as a protective response in diverse disease states which has been attributed to the beneficial properties of the by products iron, carbon monoxide and biliverdin/bilirubin. O-glycosylation, a type of posttranslational modification, is abundant and dynamic in the cell, yet its function in cell signaling and destiny regulation is far from known. This study examines the effects of O-glycosylation on muscle differentiation and metabolism, and the potential mechanism behind the effects: proteasome blockade. Proteasome action is required for muscle differentiation. We previously have shown that O-GlcNAc modification of the 19S cap of the proteasome can inhibit the proteolytic function of this organelle. By changing O-GlcNAc modification levels and proteasome function independently in C2C12 skeletal myoblasts, we have found that myoblast differentiation is largely dependent on both. …

The ability to control Listeria monocytogenes on ready-to-eat poultry products using carboxymethyl-cellulose film coatings containing green tea extract (GTE), malic acid (M), nisin (N), and their combinations was evaluated. The antimicrobials (GTE: 1.0%, nisin: 10,000 IU/g, malic acid: 1.0%) were incorporated alone or in combination into a carboxymethyl cellulose film coating. Pre-inoculated, fully cooked chicken pieces (~1g, 1cm x 1cm x 1cm) were coated with the film solution. The coated chicken pieces were stored at 4°C and the inhibitory activity against Listeria monocytogenes was evaluated at 0, 7, 14, 21, and 28 days. The highest inhibitory activity was found in …

Osteosarcoma is the most predominant primary bone cancer among pediatric patients. The pathogenesis and mechanism of osteosarcoma, however, as yet remains unclear. This study investigated the hypothesis that Ca2+/calmodulin-dependent protein kinase II (CaMK II), activated by Ca2+ and its intracellular receptor, calmodulin, plays a critical role in the growth of human osteosarcoma cells. The inhibition of CaMK II with its pharmacologic antagonist KN-93 was used to investigate the role of CaMK II in the growth of human osteosarcoma cells because our preliminary data showed that =-CaMK II is expressed in several human osteosarcoma cell lines. The periodontal ligament (PDL) is thought to include progenitor cells capable of forming fibroblasts, osteoblasts and cementoblasts. However, characterization of PDL stem cell populations (SC) remains undetermined due in part, to the diversity of cell types that conform to the periodontal apparatus. Objective: To isolate and characterize PDLSC and assess their pluripotent capabilities to differentiate into bone, cartilage and adipose cell types. African Americans, compared to Caucasians, may have chronically suppressed free fatty acid (FFA) mobilization due to basal and postprandial hyperinsulinemia. The pur-pose of this study was to determine if lower FFA among African Americans are due to higher insulin concentration and if this ethnic difference in FFA, beta cell function, and insulin suppression of FFA declines with age. Subjects who qualified underwent a fre-quently-sampled intravenous glucose tolerance test (FSIGT) for determination of insulin sensitivity (Si) and the acute insulin response to glucose (AIRg). In eukaryotes, translation termination is a process which is initiated by the presence of a stop codon in the A site of the ribosome and mediated by the binding of a release factor (eRF1). In most eukaryotes, any one of three stop codons UGA, UAG, or UAA, is required for the binding of eRF1. However some organisms, such as the ciliates, have diverged from this universal coding. Sessile benthic marine organisms such as sponges are frequently colonized by a wide diversity of invertebrates. The present study focused on a quantitative analysis of the mesofauna associated with the common subtropical/tropical sponge Amphimedon viridis and explored several criteria for the basis for this mesofauna-sponge relationship. Specimens of Amphimedon viridis were hand-collected individually in bags from both Saint Joseph Bay, Florida Panhandle, and Sugar Loaf Key, Florida Keys between April - November 2006. Quantitative analyses of sponge surfaces and interiors revealed that the most common sponge-associated group was comprised of amphipods, followed by polychaetes and tenaid crustaceans. 35 percent of …

A content analysis was done on various studies involving hormonal effects on verbal and physical aggression in animals ranging from “old world monkeys” to humans in order to prove that genetics is responsible for verbal aggressiveness used by military personnel in the contemporary operating environment. Small Lytechinus variegatus were collected in October 2004 from Port St. Joseph Peninsula State Park, FL (30° N, 85.5° W) and transported to the Texas A&M Shrimp Mariculture Facility in Port Aransas, TX.
