Chiral 1, 2-Diamino Guests In Chain Replacement Peptidomimetics: A New Helical Motif, Marlon D. Jones Jan 2007

Chiral 1, 2-Diamino Guests In Chain Replacement Peptidomimetics: A New Helical Motif, Marlon D. Jones

University of Kentucky Doctoral Dissertations

Peptides are short, sequence and length specific oligomers composed of small amino acid residues. Nature has refined these peptide sequences and their endogenous function through evolution. In addition, peptides have played an important role in medicine, which has lead to further research into developing peptides as lead pharmaceuticals (therapeutic peptides). Unfortunately, therapeutic peptides are inferior as drug candidates due to their low oral bioavailability; immunogenicity and potential to be attacked by peptidases. Fortunately, peptides can be modified by steric constraints, cyclization, and/or replacement of the peptide backbone itself creating a mimic (peptidomimetic) of the original peptide. Peptidomimetics are deliberately designed …

Neonatal Immune Modulation To Improve Pneumocystis Clearance, Kerry Mcgarr Empey Jan 2007

Neonatal Immune Modulation To Improve Pneumocystis Clearance, Kerry Mcgarr Empey

University of Kentucky Doctoral Dissertations

Pneumocystis carinii is an opportunistic fungal pathogen that causes lifethreatening pneumonia in immunocompromised individuals. Infants appear to be particularly susceptible to Pneumocystis (PC) pulmonary infections. The higher incidence of PC as well as other pulmonary infections among infants is likely due to an immature immune system. The neonatal lung environment is deficient immunologically in preterm as well as term infants (1, 2). Decreased phagocytic capacity of macrophages in newborns may increase the risk of infection from inhaled pathogens (1, 2). We have previously demonstrated that there is approximately a 3-week delay in the clearance of PC organisms from pup mouse …

Influence Of The Serotonergic System On Physiology, Development, And Behavior Of Drosophila Melanogaster, Sameera Dasari Jan 2007

Influence Of The Serotonergic System On Physiology, Development, And Behavior Of Drosophila Melanogaster, Sameera Dasari

University of Kentucky Doctoral Dissertations

The regulation and modulation of the serotonergic system is clinically significant in humans. Abnormally low levels of serotonin can result in depression and conditions like panic disorder, obsessive-compulsive disorder, social anxiety disorder, sudden infant death syndrome, and eating disorders. The mechanistic role of serotonin (5-HT) on the neural circuits related with these diseases is not definitively known. Drosophila is a simple model system that provides an advantage over vertebrates to modify genetically and for electrophysiological studies on identifiable cells. In this organism the sensory-CNS-motor circuit is modulated by 5-HT, octopamine (OA), and dopamine (DA), which gives one insight that these …

Pfi-Zeke Spectroscopy And Theoretical Calculations Of Transition Metal-Aromatic Hydrocarbon Complexes, Bradford Raymond Sohnlein Jan 2007

Pfi-Zeke Spectroscopy And Theoretical Calculations Of Transition Metal-Aromatic Hydrocarbon Complexes, Bradford Raymond Sohnlein

University of Kentucky Doctoral Dissertations

Transition metal-aromatic hydrocarbon complexes were generated in a supersonic jet and studied by zero electron kinetic energy (ZEKE) photoelectron spectroscopy and theoretical calculations. The target metal complexes were identified using time-of-flight mass spectrometry, and their ionization thresholds were located via photoionization efficiency spectroscopy. ZEKE spectroscopy was used to measure the ionization energies and vibrational frequencies of the metal complexes. Their electronic states and corresponding molecular structures were determined by comparing the experimental spectra to quantum chemical calculations and Franck-Condon simulations. In this dissertation, the metal complexes of four different aromatic hydrocarbon ligands were studied: benzene (bz), naphthalene (np), biphenyl (bp) …

Investigations Of Circadian Regulation And Immune-Circadian Interaction In The Horse, Barbara Anne Murphy Jan 2007

Investigations Of Circadian Regulation And Immune-Circadian Interaction In The Horse, Barbara Anne Murphy

University of Kentucky Doctoral Dissertations

The circadian system provides animals with a means to adapt internal physiology to the constantly changing environmental stimuli that exists on a rotating planet. Light information is translated into molecular timing mechanisms within individual pacemaker cells of the mammalian hypothalamic suprachiasmatic nucleus (SCN) via transcriptionaltranslational feedback loops. Humoral and neural outputs from this master clock result in circadian rhythms of physiology and behavior. The hierarchy of the circadian system involves SCN synchronization of cellular clocks within peripheral tissues so that differential transcriptional profiles in individual organs reflect their specific function. The first step to investigating equine circadian regulation was to …

A Comparison Of Select Trunk Muscle Thickness Change Between Subjects With Low Back Pain Classified In The Treatment-Based Classification System And Asymptomatic Controls, Kyle Benjamin Kiesel Jan 2007

A Comparison Of Select Trunk Muscle Thickness Change Between Subjects With Low Back Pain Classified In The Treatment-Based Classification System And Asymptomatic Controls, Kyle Benjamin Kiesel

University of Kentucky Doctoral Dissertations

The purposes of this dissertation were to determine: 1) the relationship betweenmuscle thickness change (MTC) as measured by rehabilitative ultrasound imaging(RUSI) and EMG activity in the lumbar multifidus (LM), 2) if motor control changesproduced by experimentally induced pain are measurable with RUSI, 3) if a differenceexists in MTC between subjects with low back pain (LBP) classified in the treatmentbasedclassification system (TBC) system and controls, 4) if MTC improves followingintervention.Current literature suggests sub-groups of patients with LBP exist and responddifferently to treatment, challenging whether the majority of LBP is "nonspecific". TheTBC system categorizes subjects into one of four categories (stabilization, mobilization, direction …

Synthesis And Reactivity Of Membrane-Supported Bimetallic Nanoparticles For Pcb And Trichloroethylene Dechlorination, Jian Xu Jan 2007

Synthesis And Reactivity Of Membrane-Supported Bimetallic Nanoparticles For Pcb And Trichloroethylene Dechlorination, Jian Xu

University of Kentucky Doctoral Dissertations

Nanosized metal particles have become an important class of materials in the field of catalysis, optical, electronic, magnetic and biological devices due to the unique physical and chemical properties. This research deals with the synthesis of structured bimetallic nanoparticles for the dechlorination of toxic organics. Nanoparticle synthesis in aqueous phase for dechlorination studies has been reported. However, in the absence of polymers or surfactants particles can easily aggregate into large particles with wide size distribution. In this study, we report a novel in-situ synthesis method of bimetallic nanoparticles embedded in polyacrylic acid (PAA) functionalized microfiltration membranes by chemical reduction of …

Factors Regulating Arteriolar Tone During Microvascular Growth, Julie Balch Samora Jan 2007

Factors Regulating Arteriolar Tone During Microvascular Growth, Julie Balch Samora

Graduate Theses, Dissertations, and Problem Reports (ETD)

Growth of the arteriolar network is accompanied by progressive changes in pressure and flow, and in the metabolic environment to which the arterioles are exposed. This dissertation was carried out to investigate the extent to which mechanisms involved in local blood flow regulation may also change during this growth. We isolated gracilis muscle arterioles from weanling (age 25-26 days) and juvenile (44-66 days) rats, and studied the responsiveness of these vessels to both myogenic and endothelium-dependent stimuli. Overall arteriolar responses to the endothelium-dependent agonists acetylcholine (ACh), A23187, VEGF and simvastatin were not different between age groups. Responses of juvenile arterioles …

Analysis Of Motor Activity Of Recombinant Myosin-1c, Anindita Biswas Jan 2007

Analysis Of Motor Activity Of Recombinant Myosin-1c, Anindita Biswas

Graduate Theses, Dissertations, and Problem Reports (ETD)

Hair cells of the inner ear are responsible for hearing and balance. These hair cells are limited in number and do not regenerate following injury. To better appreciate the function of these cells it is important to understand the molecular basis of hearing and the roles of the various molecules involved in this process. The overall goal of our laboratory is to understand the molecular basis of mechanotransduction, a process that converts the mechanical sound wave to an electrical stimulus that is transmitted to the brain. We are specifically interested in the motor protein myosin-1c (Myo1c) that has been previously …

Please, Let's Bury The Junk: The Codis Loci And The Revelation Of Private Information, David H. Kaye Jan 2007

Please, Let's Bury The Junk: The Codis Loci And The Revelation Of Private Information, David H. Kaye

Faculty Scholarship

This Northwestern University Law Review Colloquy paper describes the four possible ways in which genetic loci could possess predictive or diagnostic value with regard to diseases and explains why these mechanisms have not led, and probably cannot lead, to useful screening tests with the Convicted Offender DNA Index System (CODIS) profiles in national, state, and local databases. It then considers the phenotypes and familial relationships that the CODIS STRs can be used to identify. The profiles carry limited information about an individual's race and familial relationships, and the article places the resulting privacy issues in perspective. Finally, the paper comments …
