This dissertation will focus on a constructive hermeneutic that allows one to ask: 'What is the conversation taking place during this historical moment in education, and how can communication and rhetorical studies provide one potential answer through communicative engagement with our postmodern moment?' Periplasmic nitrate reductase from Sulfurospirillum barnesii is representative of the distinct branch of epsilonproteobacteria periplasmic nitrate reductases. The gene encoding the active subunit of S. barnesii's periplasmic nitrate reductase (napA) was successfully cloned into the vector pET101/D/TOPO. Constructs were made both with and without the sequence coding for the periplasmic signal sequence. Preliminary expression and purification experiments were performed with limited success. The DNA encoding the periplasmic signal sequence proved necessary for expression. Even with the signal sequence coding region, protein expression was limited. The low levels of expression appeared due to product degradation during cell growth. Purification was incomplete, …

The importance of estrogen in bone regulation is exemplified by the rapid loss of bone density at the onset of menopause. Post-menopausal women have low levels of estrogens, but high levels of inactive sulfated steroids. These can be converted to active steroids by steroid sulfatase. While it is known that estrogens can stimulate the growth and maintenance of bone cells, it is not known if sulfated steroids can induce a similar response. Through the modification of capillary coating procedures electroosmotic flow was controlled without altering the buffer properties. Aqueous buffers of differing ionic strengths were utilized in order to develop a better understanding of the surface properties of nanocrystalline lattices known as quantum dots. Secondary determination of nanocrystal surface properties was determined through the use of a zetasizer.

Non-aqueous buffers were utilized in order to determine the effect of the coating on aseparation of hydrophobic, biologically relevant fatty acids. In Parents Involved in Community Schools (PICS) v. Seattle School Dist. No. 1, a majority of the Supreme Court reaffirmed the constitutionality of a school district's use of race-conscious strategies designed to achieve the compelling benefits of a racially diverse student body. The Court's decision turned on the question of whether the Equal Protection Clause bars a public school district from considering the race of children of the majority group as part of its effort to foster a racially integrated school. The zebrafish is becoming an almost ubiquitously accepted model for the study of comparative developmental biology, its utility coming to rival that of the fruitfly, frog, chick and mouse. Unfortunately, however, it is widely held belief that the zebrafish cardiac outflow tract makes it unusual as a model of comparative cardiogenesis and this is based, in part, on the report that bulbus arteriosus undergoes a developmental myocardium-to-smooth muscle phenotypic transition. Concern is mounting regarding the human health and environmental effects of bisphenol A (BPA), a high-production-volume chemical used in synthesis of plastics. We have reviewed the growing literature on effects of low doses of BPA, below 50 mg/kg/day, in laboratory exposures with mammalian model organisms. Many, but not all, effects of BPA are similar to effects seen in response to the model estrogens diethylstilbestrol and ethinylestradiol. For most effects, the potency of BPA is approximately 10 to 1,000-fold less than that of diethylstilbestrol or ethinylestradiol. Urge To Gamble In Problem Gamblers Exposed To A Casino Environment, Matt G. Kushner, Kenneth Abrams, Christopher Donahue, Paul Thuras, Randy Frost, Suck Won Kim

Cue-reactivity has received increased attention in addiction research, though not for gambling in particular. We examined cue reactivity in 18 problem gamblers by accompanying them to a gaming casino and measuring their subjective urge to gamble over a 1-h period. Half of the sample was additionally exposed to a gambling-specific negative mood induction (NMI) manipulation via guided imagery. Overall, about two-thirds of the sample reported moderate to high-gambling urges during the casino exposure. Additionally, the NMI reduced cue-reactivity. Finally, gambling urges in both groups decreased over the course of the exposure sessions. These findings suggest that a majority of problem …

Declines of Steller sea lion ( Eumetopias jubatus) populations in the Aleutian Islands and Gulf of Alaska could be a consequence of physical oceanographic changes associated with the 1976–77 climate regime shift. Changes in ocean climate are hypothesized to have affected the quantity, quality, and accessibility of prey, which in turn may have affected the rates of birth and death of sea lions. Recent studies of the spatial and temporal variations in the ocean climate system of the North Pacific support this hypothesis. An intensive study that was part of the Lotic Intersite Nitrogen eXperiment II (LINX II) project was conducted to determine nitrogen transformations in nine low-order streams with contrasting land use. Short term (24-hour) additions of K15NO3 and NaBr were performed on a gradient of anthropogenically impacted streams in Puerto Rico. Nitrate uptake was determined from longitudinal decline in 15NO3 and denitrification rates were determined from the longitudinal pattern of 15N 2 and 15N2O. Several physical, chemical, and biological variables were also measured to determine controlling factors. Humans with diabetes mellitus are two to five times more likely to develop cardiovascular disease and do so at a much younger age than non-diabetics. This study utilized the Syrian F1B hamster, a model of human atherosclerosis, to: (1) determine whether one intraperitoneal injection of streptozotocin (STZ) could induce a diabetic-like hyperglycemic state, (2) compare the development of atherosclerotic lesions between non-treated (N), hyperglycemic (G), hyperlipidemic (L) and combined hyperlipidemic/hyperglycemic (L+G) hamsters, and (3) determine whether lesion development was accelerated due to plasma levels of glucose, triglycerides, total cholesterol, non-HDL-C, HDL-C, or lipid hydroperoxides, or by the TC/HDL-C or TG/HDL-C …

Angular Range Analysis (ARA) is a physics-based approach to acoustic remote seafloor characterization. In order to better understand the capabilities and limitations of this technique, ARA analyses were performed on multibeam sonar data collected at the Historical Area Remediation Site, an area with high spatial variability. The remotely derived results were compared to grain size information derived from grab samples and Sediment Profile Imaging. Uncertainties in the determination of mean grain size from ground truth were identified and when possible quantified. Thamnophis sirtalis, the Common garter snake, is a wide-ranging North American snake exhibiting great variation in morphology and life-history attributes across its range. This thesis studied reproductive variation and molecular phylogeography of T. s. sirtalis, the Eastern garter snake, and T. s. pallidulus, the Maritime garter snake, in populations from New England in 2005 and 2006. Pregnant female snakes were captured and held in captivity until parturition, and measurements were taken on parent and offspring for comparisons across years and locations. The modeling of complex biological systems presents a significant challenge. Central to this challenge is striking a balance between the degree of abstraction required to facilitate analysis and understanding, and the degree of comprehensiveness required for fidelity of the model to its reference-system. It is likely necessary to utilize multiple modeling methods in order to achieve this balance. Our research created a hybrid simulation model by melding an agent-based model of acute local infection with a system dynamics model that reflects key systemic properties.
