Replication Fork Regression In Vitro By The Werner Syndrome Protein (Wrn): Holliday Junction Formation, The Effect Of Leading Arm Structure And A Potential Role For Wrn Exonuclease Activity, Amrita Machwe, Liren Xiao, Robert G Lloyd, Edward Bolt, David K. Orren

Toxicology and Cancer Biology Faculty Publications

The premature aging and cancer-prone disease Werner syndrome stems from loss of WRN protein function. WRN deficiency causes replication abnormalities, sensitivity to certain genotoxic agents, genomic instability and early replicative senescence in primary fibroblasts. As a RecQ helicase family member, WRN is a DNA-dependent ATPase and unwinding enzyme, but also possesses strand annealing and exonuclease activities. RecQ helicases are postulated to participate in pathways responding to replication blockage, pathways possibly initiated by fork regression. In this study, a series of model replication fork substrates were used to examine the fork regression capability of WRN. Our results demonstrate that WRN catalyzes …

Fa2h Dependent Fatty Acid 2-Hydroxylation In The Mammalian Nervous System, Nathan Lance Alderson Jan 2007

Fa2h Dependent Fatty Acid 2-Hydroxylation In The Mammalian Nervous System, Nathan Lance Alderson

MUSC Theses and Dissertations

Myelin is formed by oligodendrocytes in the central nervous system and Schwann cells in the peripheral nervous system. This unique membrane is comprised of approximately 70% lipid and 30% protein. The high lipid content is thought to be vital for its insulatory function. Galactosylceramide (GaICer) and sulfatide make up approximately 30% of total myelin lipids, with more than half of these galactolipids containing fatty acids hydroxylated at the C[subscript 2] position (2-hydroxy fatty acids). Despite their high abundance, very little is know about the biosynthesis of these 2-hydroxy galactolipids, and specific functions of the 2-hydroxyl group in myelin galactolipids remain …

Notes On A Poetics Of Time, Lawrence Kimmel Jan 2007

Notes On A Poetics Of Time, Lawrence Kimmel

Philosophy Faculty Research

The idea of a poetics in contrast with an aesthetics of time is intended to focus on the creative possibilities of imagination in configurations of time. An aesthetics of time focusing on sensuous experience is a certainly a basic resource of creative imagination in literature. But the concept of a poetics of time, taken from the root meaning of poiesis in classical Greek thought—to make, or to bring forth—enables an inquiry into conceptions of human life and thought brought forth in various creative configurations of time in literature. This essay will analyze some of the ways in which poetic imagination …

Racketed Tail Of The Male And Female Turquoise-Browed Motmot: Male But Not Female Tail Length Correlates With Pairing Success, Performance, And Reproductive Success, Troy G. Murphy Jan 2007

Racketed Tail Of The Male And Female Turquoise-Browed Motmot: Male But Not Female Tail Length Correlates With Pairing Success, Performance, And Reproductive Success, Troy G. Murphy

Biology Faculty Research

Both males and females of many avian species maintain elaborate plumage traits, and elaborate monomorphic plumage may convey adaptive benefits to one or both sexes as inter- or intraspecific signals. Both sexes of the turquoise-browed motmot (Eumomota superciliosa) are elaborately plumed with long racket-tipped tail. I investigated whether the racketed tail functions as a sexually selected signal in one or both sexes by testing the predictions that males and/or females with the largest tails have: (1) greater pairing success, (2) greater reproductive performance (clutch-initiation date, clutch size, and hatching success), and (3) greater reproductive success. Yearling males with …

Hyperspectral Imaging: A Non-Invasive Technique To Predict Beef Tenderness, Lauren Grimes, Govindarajan Konda Naganathan, Jeyamkondan Subbiah, Chris R. Calkins

Nebraska Beef Cattle Reports

A hyperspectral imaging apparatus was developed and assembled to predict 14 d tenderness of beef steaks. USDA Choice and Select grade longissimus steaks (n=111) from between the 12th and 13th ribs were frozen at 14 days post-mortem, cut to 1-inch thickness, and thawed overnight for scanning, cooking, and obtaining slice shear force data. The model predicted three tenderness categories with 96.4% accuracy, correctly classifying 93 tender, nine intermediate and all five tough samples. One tender sample was misclassified as intermediate, and three intermediate samples were misclassified as tender. This hyperspectral imaging system was effective in predicting beef tenderness.

Statistics Used In The Nebraska Beef Report And Their Purpose

Nebraska Beef Cattle Reports

The purpose of beef cattle and beef product research at UNL is to provide reference information that represents the various populations (cows, calves, heifers, feeders, carcasses, retail products, etc.) of beef production. Obviously, the researcher cannot apply treatments to every member of a population; therefore, he or she must sample the population. The use of statistics allows the researcher and readers of the Nebraska Beef Report the opportunity to evaluate separation of random (chance) occurrences and real biological effects of a treatment. Following is a brief description of the major statistics used in the beef report. For a more detailed …

Beta-Lactamase: An Ideal Reporter System For Monitoring Gene Expression In Live Eukaryotic Cells, Sohail A. Qureshi

Department of Biological & Biomedical Sciences

To gain insightful information about the mechanisms through which genes are activated and repressed requires gene reporter systems that are sensitive, robust, and cost-effective. Although numerous reporter gene technologies are commercially available, none are as sophisticated and user-friendly as beta-lactamase (BLA) when it comes to studying gene expression in live cells. This article presents an overview of the BLA technology and describes how it can be exploited for studying rare events such as homologous recombination in somatic cells and be used to deliver any DNA sequence of choice anywhere within the genome.

Predicting student achievement is often the goal of many studies, and a frequently employed tool for constructing predictive models is multiple linear regression. This research sought to compare the performance of a three-layer back propagation neural network to that of traditional multiple linear regression in predicting math and reading proficiency from 103 non-curricular variables collected in the National Center for Educational Statistics' 2002 Educational Longitudinal Study. The neural network model was implemented using the Java programming language and the coefficients for the regression equations were produced by SPSS. The results showed that, for this data set, neither model provided an …

The Evolution Of Protein Components Of Insect Spermtail Axonemes, Lisa M. Griffith Jan 2007

The Evolution Of Protein Components Of Insect Spermtail Axonemes, Lisa M. Griffith

Graduate Theses and Dissertations

Drosophilids spermtails are considered to be the peacock feathers of the world of sperm, by virtue of their incredible length. This extreme length may be supported by their use of a unique, specialized, testis-specific Beta2 tubulin. Beta2 has a highly stringent structure/function relationship to the Drosophilid spermtail axoneme, such that even small amino acid changes render it non-functional. How then, does this protein evolve, while maintaining is function? We hypothesize that it may need to co-evolve with other axonemal proteins. Here, one and two dimensional gel electrophoresis was used to compare the testes proteomes of several fly species in an …

Sulfur-Oxidizing Bacteria In Soap Lake (Washington State), A Meromictic, Haloalkaline Lake With An Unprecedented High Sulfide Content, Dimitry Y. Sorokin, Mirjam Foti, Holly C. Pinkart, Gerard Muyzer

All Faculty Scholarship for the College of the Sciences

Culture-dependent and -independent techniques were used to study the diversity of chemolithoautotrophic sulfur-oxidizing bacteria in Soap Lake (Washington State), a meromictic, haloalkaline lake containing an unprecedentedly high sulfide concentration in the anoxic monimolimnion. Both approaches revealed the dominance of bacteria belonging to the genus Thioalkalimicrobium, which are common inhabitants of soda lakes. A dense population of Thioalkalimicrobium (up to 107 cells/ml) was found at the chemocline, which is characterized by a steep oxygen-sulfide gradient. Twelve Thioalkalimicrobium strains exhibiting three different phenotypes were isolated in pure culture from various locations in Soap Lake. The isolates fell into two groups …

Dimesionality Aspects Of Nano Micro Integrated Metal Oxide Based Early Stage Leak Detection Room Temperature Hydrogen Sensor, Sameer Arun Deshpande

Electronic Theses and Dissertations

Detection of explosive gas leaks such as hydrogen (H2) becomes key element in the wake of counter-terrorism threats, introduction of hydrogen powered vehicles and use of hydrogen as a fuel for space explorations. In recent years, a significant interest has developed on metal oxide nanostructured sensors for the detection of hydrogen gas. Gas sensors properties such as sensitivity, selectivity and response time can be enhanced by tailoring the size, the shape, the structure and the surface of the nanostructures. Sensor properties (sensitivity, selectivity and response time) are largely modulated by operating temperature of the device. Issues like instability of nanostructures …
