Rapid accurate detection is a prerequisite for the successful control of meticillin-resistant Staphylococcus aureus (MRSA). The IDI-MRSA real-time polymerase chain reaction (PCR) assay was designed to provide rapid results from nasal specimens collected in Stuart’s liquid transport medium. This study has evaluated the IDI-MRSA kit for use in a clinical laboratory by investigating the following parameters: (1) limits of detection (LoD), (2) performance with Amies’ gel-based transport medium, (3) ability to detect strains of MRSA in a collection representative of MRSA in Ireland since 1974 (n 113) and (4) performance in a clinical trial with swabs from nose, throat and …

Formate dehydrogenase H from Escherichia coli contains selenocysteine (SeCys), molybdenum, two molybdopterin guanine dinucleotide (MGD) cofactors, and an Fe4S4 cluster at the active site and catalyzes the two-electron oxidation of formate to carbon dioxide. The crystal structures of the oxidized [Mo( VI), Fe4S4(ox)] form of formate dehydrogenase H (with and without bound inhibitor) and the reduced [Mo(IV ), Fe4S4(red)] form have been determined, revealing a four-domain ab structure with the molybdenum directly coordinated to selenium and both MGD cofactors. These structures suggest a reaction mechanism that directly involves SeCys …

The authors demonstrate surface-enhanced Raman scattering (SERS) detection on an optofluidic chip. Interconnected solid- and liquid-core antiresonant reflecting optical waveguides (ARROWs) form a planar beam geometry that allows for high mode intensities along microfluidic channels containing molecules optimized for SERS. The excitation power and concentration dependence of SERS from rhodamine 6G (R6G) molecules adsorbed to silver nanoparticles were systematically studied. The data can be described by a model that takes into account the microphotonic structure. Detection sensitivity to a minimum concentration of 30 nM is found, demonstrating the suitability of ARROW-based optofluidic chips for high sensitivity detection with molecular specificity
Hysteria, Greed, And The Public Purse: The Litigation Impacting A Local Governmental Agency, Claire D. Simno
Hysteria, Greed, And The Public Purse: The Litigation Impacting A Local Governmental Agency, Claire D. Simno
LSU New Orleans Theses and Dissertations
This research is a case study of the litigation in progress by hundreds of St. Bernard Parish families against the Sewerage and Water Board of New Orleans and Browning Ferris Industries (BFI). The families are suing for personal and property damages sustained as a result of the poor operation and maintenance of the East Bank Sewerage Treatment Plant (EBSTP) and the Crescent Acres Landfill. The purpose of the study will be to examine the consequences, links, and process of interaction among three key units of analysis: the plaintiffs, their lawyers, and one of the major defendants in the litigation, the …

The Tigers And The Army: The Lsu School Of Medicine During World War Ii, Victoria Barreto
LSU New Orleans Theses and Dissertations
The art of medicine has long found its most difficult challenges on fields of battle. During World War II, medical schools like the Louisiana State University School of Medicine were forced to play a larger role in total warfare. They served the medical needs of both civilians and soldiers by providing more doctors for the homefront and sponsoring medical units that served abroad. LSU sponsored the 64th General Hospital, which served in both the North African and Italian theaters. Clearly, then, the story of the LSU School of Medicine during World War II provides a glimpse into the intricate collision …

Summer Ecology Of The Teshekpuk Caribou Herd, Lincoln Scott Parrett
Summer Ecology Of The Teshekpuk Caribou Herd, Lincoln Scott Parrett
Biological Sciences
The summer range of the Teshekpuk Caribou (Rangifer tarandus granti) Herd is currently undergoing the initial stages of petroleum exploration and development. Pre-development baseline information is necessary to interpret post-development distribution and habitat selection of caribou and to develop mitigation measures. We estimated bi-weekly distributions, diet and habitat selection by caribou during the summers, 2002-2004, based on aerial relocations of 21-49 radio-collared females. Little or no habitat selection was detected when comparing used locations to habitat available within bi-weekly utilization distributions. Habitat selection was much stronger when comparing bi-weekly utilization distributions to the remaining area of summer use. At the …

Comparative Immune Function In Wild Birds, Kevin David Matson
Comparative Immune Function In Wild Birds, Kevin David Matson
Dissertations
Over the last several decades, interest in quantifying immune function in comparative studies of wild animals has grown appreciably. Now, the field of ecological immunology is undergoing a transition, and ¿second generation¿ studies are being designed and carried out. With a greater appreciation of the complexity of immune systems, these second generation studies are commonly distinguished from their antecedents by making comparisons using multiple assays and including multiple species. I worked to advance this transition by developing novel approaches to comparative immunology, exploring the interrelationships among indices of immune function, and applying multiple indices to a question of comparative avian …

MicroRNAs (miRNAs) are short 19- to 24-nt RNA molecules that have been shown to regulate the expression of other genes in a variety of eukaryotic systems. Abnormal expression of miRNAs has been observed in several human cancers, and furthermore, germ-line and somatic mutations in human miRNAs were recently identified in patients with chronic lymphocytic leukemia. Thus, human miRNAs can act as tumor suppressor genes or oncogenes, where mutations, deletions, or amplifications can underlie the development of certain types of leukemia. In addition, previous studies have shown that miRNA expression profiles can distinguish among human solid tumors from different organs. Because …

Sme Performance, Innovation And Networking Evidence On Complementarities For A Local Economic System, Massimiliano Mazzanti, Susanna Mancinelli
Sme Performance, Innovation And Networking Evidence On Complementarities For A Local Economic System, Massimiliano Mazzanti, Susanna Mancinelli
Fondazione Eni Enrico Mattei Working Papers
The paper addresses the relevancy of networking activities and R&D as main drivers of productivity performance and ouput innovation, for small and medium enterprises (SME) playing in a local economic system. Given the intangible nature of many techno organisational innovation and networking strategies, original recent survey data for manufacturing and services are exploited. The aim is to provide new evidence on the complementarity relationships concerning different networking activities and R&D in a local SME oriented system in Northern Italy. We first introduce a methodological framework to empirically test complementarity among R&D and networking, in a discrete setting. Secondly, we consequently …

T-Cell Dysfunction By Hcv Core Protein Involves Pd-1/Pd-L1 Signaling., Billy Ellis King
T-Cell Dysfunction By Hcv Core Protein Involves Pd-1/Pd-L1 Signaling., Billy Ellis King
Electronic Theses and Dissertations
In 1989 the hepatitis C virus was identified as a significant cause of post-transfusion hepatitis. Nearly two decades later there is still no vaccine, inadequate treatment options, and limited understanding of how the virus establishes chronicity in the majority of the people it infects. Recent reports suggest that the interaction of a negative co-stimulatory pathway mediated by PD-1 and PDL-1 is associated with persistent viral infection. The role, if any, that PD-1/PDL-1 has in HCV infection is unknown. In this study we report that PD-1 is upregulated in T-cells from persons with chronic HCV infection when compared to healthy donors. …
Examination Of Microsporidia Spore Adherence And Host Cell Infection In Vitro., Timothy Robert Southern
Examination Of Microsporidia Spore Adherence And Host Cell Infection In Vitro., Timothy Robert Southern
Electronic Theses and Dissertations
Microsporidia are obligate intracellular pathogens that cause severe disease in immunocompromised humans. While albendazole is the treatment of choice, no therapy exists that effectively treats all forms or causes of human microsporidiosis. Recent studies show that the microsporidian Encephalitozoon intestinalis binds glycosaminoglycans (GAGs) associated with the host cell surface, and that the divalent cations manganese (Mn2+) and magnesium (Mg2+) augment spore adherence to host cells by activating a constituent on the spore surface. These studies also illustrate a direct relationship between spore adherence and host cell infection; inhibition of spore adherence leads to reduced host cell …

Electronic Theses and Dissertations
Erythromycin is a macrolide antibiotic that inhibits not only mRNA translation but also 50S ribosomal subunit assembly in bacterial cells. An important mechanism of erythromycin resistance is the methylation of 23S rRNA by erm methyl transferase enzymes. We are interested in investigating the true substrate for methylation because it is known from our work and the work of others that fully assembled 50S subunits are not substrates for methylation. We have published a model for 50S ribosomal subunit formation where, the precursor particle that accumulates in erythromycin treated cells is a target for methyl transferase activity. Current studies are aimed …

The Microbiological Analysis Of Composting, Marion Shepherd
All Theses
The objectives of this study were to: 1) perform a survey of South Carolina poultry farms to determine if the methods implemented resulted in the destruction of foodborne pathogens, and 2) determine the survival of E. coli O157:H7 in a dairy manure-based compost performed in uncontrolled environmental conditions.
In the survey of poultry farms Ninety-one (91) of 141 compost samples analyzed were positive for coliform populations ranging from 1.00 to 6.00 log10 CFU/g. Among finished compost samples (n=21), ca. 62%, 33%, and 14% were positive for coliforms, presumptive Salmonella, and presumptive Listeria, respectively.
In the investigation of the survival of …
