Cloning And Heterologous Expression Of The Napa Gene From Sulfurospirillum Barnesii, Lina Bird

Periplasmic nitrate reductase from Sulfurospirillum barnesii is representative of the distinct branch of epsilonproteobacteria periplasmic nitrate reductases. The gene encoding the active subunit of S. barnesii's periplasmic nitrate reductase (napA) was successfully cloned into the vector pET101/D/TOPO. Constructs were made both with and without the sequence coding for the periplasmic signal sequence. Preliminary expression and purification experiments were performed with limited success. The DNA encoding the periplasmic signal sequence proved necessary for expression. Even with the signal sequence coding region, protein expression was limited. The low levels of expression appeared due to product degradation during cell growth. Purification was incomplete, …

The purpose of this study was to examine the change in preservice teachers' attitudes towards using technology to meet the needs of diverse students as noted by the National Educational Technology Standards for Teachers (NETS*S) III. B. This research included two interventions: a research paper assignment and a hands-on practice using technology to assist students with special needs. The study took place within the coursework in an instructional technology class offered for preservice teachers. The use of cocaine during adolescent development could alter the normal growth of brain regions affected by cocaine, specifically the reward system, and impact the adult mesolimbic system. However, there is scant literature aimed at determining whether animals are more vulnerable to the adverse effects of drugs during adolescence. The present study investigated whether cocaine pretreatment in either adolescence or adulthood altered the dopaminergic response to a naturally reinforcing substance in adulthood. To evaluate the responsivity of the mesolimbic system after repeated cocaine, sucrose was offered during the dialysis procedure and dialysates were collected. The Indian boy over whom the king and queen of fairies quarrel is the most important of several characters in A Midsummer Night's Dream who do not appear on stage: his parents, who form with him a missing nuclear family; a child god, Cupid; and a female authority figure, the dowager to whose property the lovers Lysander and Hermia flee. In its narrative structure the play presents a healing regression to the early mother and the primary process. The traditional research and development model of large pharmaceutical companies is arguably unsustainable in current times. For example, estimated research and development costs increased as much as twelve percent over the last year while pharmaceutical sales grew only seven percent over the same period. Current estimates put the price to develop a new drug and bring it to market between $800 million and $1.5 billion per drug. These costs are increasing, driving large pharmaceutical companies to find more cost-effective research and development models. One cost-saving initiative is to globalize the system. Concern is mounting regarding the human health and environmental effects of bisphenol A (BPA), a high-production-volume chemical used in synthesis of plastics. We have reviewed the growing literature on effects of low doses of BPA, below 50 mg/kg/day, in laboratory exposures with mammalian model organisms. Many, but not all, effects of BPA are similar to effects seen in response to the model estrogens diethylstilbestrol and ethinylestradiol. For most effects, the potency of BPA is approximately 10 to 1,000-fold less than that of diethylstilbestrol or ethinylestradiol. We have reviewed research on the effects of stress on LTP in the hippocampus, amygdala and prefrontal cortex (PFC) and present new findings which provide insight into how the attention and memory-related functions of these structures are influenced by strong emotionality. We have incorporated the stress-LTP findings into our “temporal dynamics” model, which provides a framework for understanding the neurobiological basis of flashbulb and traumatic memories, as well as stress-induced amnesia. This study tests whether the face-processing sys- tem of humans and a nonhuman primate species share char- acteristics that would allow for early and quick processing of socially salient stimuli: a sensitivity toward conspecific faces, a sensitivity toward highly practiced face stimuli, and an ability to generalize changes in the face that do not sug- gest a new identity, such as a face differently oriented. The look rates by adult tamarins and humans toward conspecific and other primate faces were examined to determine if these characteristics are shared. The Kentucky Geological Survey, University of Kentucky, and the Kentucky Division of Water (of the Kentucky Environmental and Public Protection Cabinet) are evaluating groundwater quality throughout the commonwealth to determine regional conditions, assess impacts of nonpoint-source pollutants, establish a basis for detecting changes, and provide essential information for environmental-protection and resource-management decisions.

These evaluations are being conducted in stages. Under the Kentucky Watershed management Framework, Kentucky’s 12 major river basins and tributaries of the Ohio River were grouped into five basin management units (BMU’s). Humans with diabetes mellitus are two to five times more likely to develop cardiovascular disease and do so at a much younger age than non-diabetics. This study utilized the Syrian F1B hamster, a model of human atherosclerosis, to: (1) determine whether one intraperitoneal injection of streptozotocin (STZ) could induce a diabetic-like hyperglycemic state, (2) compare the development of atherosclerotic lesions between non-treated (N), hyperglycemic (G), hyperlipidemic (L) and combined hyperlipidemic/hyperglycemic (L+G) hamsters, and (3) determine whether lesion development was accelerated due to plasma levels of glucose, triglycerides, total cholesterol, non-HDL-C, HDL-C, or lipid hydroperoxides, or by the TC/HDL-C or TG/HDL-C …

Biodiesel was produced by transesterification of waste olive oil with methanol and NovozymRTM435.

Studies were also performed to evaluate the effect at different acyl acceptors and/or solvents on biodiesel yield. Among the selections, tert-butanol with methanol as the solvent and acyl acceptor gave relatively high yield, while tert-butanol without methanol gave a very low yield. Hexane as the solvent and methanol as the acyl acceptor also gave good yield. While using methyl acetate as both the solvent and the acyl acceptor, the yield was a little low, but still much higher than in tert-butanol without methanol. However, compared to the …

The goal of this thesis is to restore complexity, normalcy, and the later development of racism to the history and culture of slavery in New Hampshire. In addition to its role as a normal aspect of life in a hierarchical Atlantic world, slavery in New Hampshire gave birth to both racism and racial consciousness after its gradual demise in the state. Thamnophis sirtalis, the Common garter snake, is a wide-ranging North American snake exhibiting great variation in morphology and life-history attributes across its range. This thesis studied reproductive variation and molecular phylogeography of T. s. sirtalis, the Eastern garter snake, and T. s. pallidulus, the Maritime garter snake, in populations from New England in 2005 and 2006. Pregnant female snakes were captured and held in captivity until parturition, and measurements were taken on parent and offspring for comparisons across years and locations. Tissue samples were taken from adult snakes for mtDNA sequencing and subsequent phylogenetic comparisons. No substantial trends were observed …

The modeling of complex biological systems presents a significant challenge. Central to this challenge is striking a balance between the degree of abstraction required to facilitate analysis and understanding, and the degree of comprehensiveness required for fidelity of the model to its reference-system. It is likely necessary to utilize multiple modeling methods in order to achieve this balance. Our research created a hybrid simulation model by melding an agent-based model of acute local infection with a system dynamics model that reflects key systemic properties. This study argues the efficacy of the phenomenological cultural work of a visual culture archaeology that liberates a political and critical identity, resistant to domination, authoring social change and its own agency through multiple and incommensurable positions. Built upon Foucauldian premises, visual culture archaeology is developed as a methodology for discursive un-naming and re-naming, and emerges from the inherence and attenuation of inscripted meanings in the reinterpretation of identity during a postmodern confluence of ideas and images. This study examines two public hearings on a zoning proposal that would allow the construction of a Super Wal-Mart Center on a field over the town’s aquifer. Many citizens speak out against the zoning change because of the risk to drinking water, as well as other issues. Citizens face the speaker’s problem of how to make their presentations convincing, given the technical matters involved and the fact that Town Board members have likely already heard about these issues. In Caenorhabditis elegans, as in other animals, Notch-type signaling mediates numerous inductive events during development. The mechanism of Notch-type signaling involves proteolytic cleavage of the receptor and subsequent transport of the receptor intracellular domain to the nucleus, where it acts as a transcriptional regulator. Notch-type signaling activity is modulated by post-translational modifications and endocytosis of ligand and receptor. We previously identified the ego-2 (enhancer of glp-1) gene as a positive regulator of germline proliferation that interacts genetically with the GLP-1/Notch signaling pathway in the germline. Drawing on the rich tradition of textile crafts in the Ottoman Empire, Turkish soldiers incarcerated in British prison camps in the Middle East during and immediately after World War I made a variety of beadwork items to relieve the boredom of their prolonged imprisonment and to barter or sell for food and other amenities. Best known are the bead crochet snakes and lizards, but the prisoners also used loomed and netting techniques to produce necklaces, belts, purses, and other small items.

Debris flows can drastically alter ecosystems and damage infrastructure, and there is an increased risk of debris flows following a wildfire. In general, runoff and erosion drastically increase following a wildfire as a result of many processes, one of them being debris flows. Large debris flows are common in the months and years after wildfire in mountainous areas throughout the western U.S.A. The progressive bulking of surface runoff may be the dominant triggering mechanism of these debris flows. Vegetative ash on the hillslope becomes entrained in the flow, along with other fine-grained sediment and increases the effective viscosity of the …
