Mouse Preimplantation Embryo Responses To Culture Medium Osmolarity Include Increased Expression Of Ccm2 And P38 Mapk Activation., Barry Fong, Patricia H Watson, Andrew J Watson
Obstetrics & Gynaecology Publications
BACKGROUND: Mechanisms that confer an ability to respond positively to environmental osmolarity are fundamental to ensuring embryo survival during the preimplantation period. Activation of p38 mitogen-activated protein kinase (MAPK) occurs following exposure to hyperosmotic treatment. Recently, a novel scaffolding protein called Osmosensing Scaffold for MEKK3 (OSM) was linked to p38 MAPK activation in response to sorbitol-induced hypertonicity. The human ortholog of OSM is cerebral cavernous malformation 2 (CCM2). The present study was conducted to investigate whether CCM2 is expressed during mouse preimplantation development and to determine whether this scaffolding protein is associated with p38 MAPK activation following exposure of preimplantation …

Forced Mobilization Accelerates Pathogenesis: Characterization Of A Preclinical Surgical Model Of Osteoarthritis, C Thomas G Appleton, David D Mcerlain, Vasek Pitelka, Neil Schwartz
Bone and Joint Institute
Preclinical osteoarthritis (OA) models are often employed in studies investigating disease-modifying OA drugs (DMOADs). In this study we present a comprehensive, longitudinal evaluation of OA pathogenesis in a rat model of OA, including histologic and biochemical analyses of articular cartilage degradation and assessment of subchondral bone sclerosis. Male Sprague-Dawley rats underwent joint destabilization surgery by anterior cruciate ligament transection and partial medial meniscectomy. The contralateral joint was evaluated as a secondary treatment, and sham surgery was performed in a separate group of animals (controls). Furthermore, the effects of walking on a rotating cylinder (to force mobilization of the joint) on …

Assessment Of Rat Lung Volumes Using Hyperpolarized Helium-3 Mr Imaging, M. Reza Akhavan Sharif
Digitized Theses
In this thesis, the upper-limit volume (gas plus partial tissue volume) as well as absolute volume (gas only) of lungs measured using 3He Magnetic Resonance (3He-MR) imaging is compared with that determined by micro-Computed Tomography (micro-CT) and xenon-enhanced micro-CT (Xe-CT) in normal rats. The upper-limit and absolute volumes obtained from He-MR and air-CT methods were not significantly different (P>0.05). Also, no significant difference was found between the volumes measured from He-MR and Xe-CT methods (P>0.05). The good agreement between the volumes measured from He-MR and air-CT suggests that He MR imaging can be used for quantitative analysis of …

Are There Individual And/Or Roost Signatures In The Echolocation Calls Of Wild Big Brown Bats (Eptesicus Fuscus)?, Elisabeth K. Van Stam
Digitized Theses
Unique individual vocal signatures are widespread among animals living in colonies such as the maternity roosts of Eptesicusfuscus. Such individual signatures have been identified in the echolocation calls of E. fuscus in the laboratory, but have not been demonstrated in the wild. By recording known wild E. fuscus as they emerged from their roosts at dusk, I tested the hypothesis that individual and/or roost signatures exist in the echolocation calls of wild E. fuscus. Analyses of calls of 176 individuals recorded at six different locations indicate that temporal and spectral features appear to contain sufficient variation to identify both roost …
Cellular Basis For Stem Cell Mediated Regeneration Of Damaged Myocardium In A Canine Model For Myocardial Infarct, Caroline N. Culshaw
Cellular Basis For Stem Cell Mediated Regeneration Of Damaged Myocardium In A Canine Model For Myocardial Infarct, Caroline N. Culshaw
Digitized Theses
Heart failure from myocardial infarct (MI) is the leading cause of death in North America. While stem cell therapy for MI has been examined in many animal models and in clinical triais, the ability of stem cells to regenerate damaged myocardium is controversial. I have examined the ability of bone marrow and skeletal muscle derived stem cells to differentiate into cardiac and skeletal muscle following injection into infarcted dog hearts. Immunofluorescent co-localization of myosin heavy chain (MyHC) and the fluorescent cell marker PHK26 demonstrated that, while injected cells could be detected at injection sites, they did not express cardiac or …

Cdk5-Dependent Augmentation Of Dopamine Release At Mesolimbic Terminals May Contribute To The Enhanced Dopaminergic Activity In Schizophrenia, Kesavan Sathanantham
Digitized Theses
Schizophrenia is characterized by chronic dopaminergic hyperactivity and increased release of dopamine at mesolimbic terminals. Our lab has developed a putative animalmodelofschizophreniabyablatingthedevelopingprefrontalcortexinrats. As adults, these animals display behavioral features representing chronic mesolimbic dopaminergic hyperactivity. The role of increased levels of cyclin-dependent kinase 5 (cdk5) may play in mesolimbic dopamine release was investigated in adult rats that receivedneonatalprefrontalcorticalablation. Resultsshowedincreasedlevelsofcdk5 protein in dopamine neurons in lesioned rats, and revealed increased levels of phosphorylated dynamin-1 at serine 774 and 778, targets of cdk5, in mesolimbic dopamine neurons suggesting increased cdk5 activity. Behavioral studies have shown increased locomotor response to stress, methamphetamine or apomorphine …
Design And Performance Evaluation Of A Small-Animal Pinhole-Spect Array Insert For Field-Cycled Mri, James William Kristian Odegaard
Design And Performance Evaluation Of A Small-Animal Pinhole-Spect Array Insert For Field-Cycled Mri, James William Kristian Odegaard
Digitized Theses
Dual-modality SPECT-MRI allows the functional imaging benefits of SPECT to be overlaid on the high-resolution structural images for which MRI is known. The two modalities, however, do not coexist easily. This study examines the viability of per forming pinhole-SPECT via an array insert within an existing field-cycled MRI scan ner. The purpose of this investigation is to determine the magnitude and distribution of additional scatter introduced into the SPECT data by the presence of the field- cycled MRI hardware.
An insert consisting of twelve lead collimators with tungsten single-pinhole aper tures was designed for imaging a 1-cm-radius spherical volume. A …
Development And Characterization Of Non- Invasive Measures Of Neonatal Hypoxic- Ischemic Brain Injury, Jeff D. Winter
Development And Characterization Of Non- Invasive Measures Of Neonatal Hypoxic- Ischemic Brain Injury, Jeff D. Winter
Digitized Theses
Neonatal hypoxic-ischemic brain injury often leads to severe neurodevelopmental impairment. Early detection of injury is important for the clinical management of neonates as well as monitoring of neuroprotective therapies, which are currently being investigated. ■
Diffusion-weighted imaging (DWI) plays an important role in the assessment of neonatal patients with clinical indications of hypoxic-ischemic encephalopathy (HIE). With the goal of improving efficacy of DWI in neonatal HIE, the first two parts of this thesis are (i) the characterization of the evolution of the apparent diffusion coefficient (ADC) in neonates with HIE and poor outcome (particularly the postnatal age when ADC values …

Effects Of Intermittent Umbilical Cord Occlusions On Metabolism, Cardiovascular Responses And Brain Injury In The Ovine Fetus, Hedieh Hamrahi
Digitized Theses
Intermittent umbilical cord compressions may arise as a result of nuchal cords, uterine contractions and/or oligohydramnios. Birth asphyxia accounts for only 10% of all cases of cerebral palsy, pointing to the importance of events in the antepartum period as contributors to brain injury. The objective of this thesis was to understand more fully the effects of antenatal hypoxie preconditioning on fetal metabolism, cardiovascular responses and brain injury. Forty four fetal sheep underwent placement of vascular catheters and an umbilical cord occluder at 122d gestation (term ~ 147d). Animals were assigned to two studies, each with two experimental groups. In the …
The Ends Of Narrative, Richard Thomas Eldridge
The Ends Of Narrative, Richard Thomas Eldridge
Philosophy Faculty Works
We can begin to approach what might be called the peculiarity of literature as a form of cognitive practice by comparing how literary works end with how other pieces of intellectual work end. A proof in mathematics ends by reaching its final line, where each line that is not an axiom is generated in explicit accord with a rule of inference that in principle anyone might follow. Reports of experimental results generated in a lab specify procedures that were followed in setting up equipment and carrying out tests. While they often also offer conjectural interpretations of results and suggestions for …

Archival data from a survey of parent observations was used to determine the prevalence of social and behavioral problems in children with agenesis of the corpus callosum (ACC). Parent observations were surveyed using the Child Behavior Checklist (CBCL) for 61 children with ACC who were selected from the archive based on criteria of motor development suggesting a relatively high general level of functioning. Younger children with ACC (ages 2–5) were rated as primarily having problems with sleep. Older children with ACC (ages 6–11) manifested problems in attention, social function, thought, and somatic complaints. The older children with ACC were also …

The Drosophila melanogaster lymph gland is a haematopoietic organ1–3 in which pluripotent blood cell progenitors proliferate and mature into differentiated haemocytes. Previous work4 has defined three domains, the medullary zone, the cortical zone and the posterior signalling centre (PSC), within the developing third-instar lymph gland. The medullary zone is populated by a core of undifferentiated, slowly cycling progenitor cells, whereas mature haemocytes comprising plasmatocytes, crystal cells and lamellocytes are peripherally located in the cortical zone. The PSC comprises a third region that was first defined as a small group of cells expressing the Notch ligand Serrate5. Here we show that …
