The Pseudomonas Aeruginosa Secreted Protein Pa2934 Decreases Apical Membrane Expression Of The Cystic Fibrosis Transmembrane Conductance Regulator, Daniel P. Maceachran, Siying Ye, Jennifer M. Bomberger, Deborah A. Hogan, Agnieszka Swiatecka-Urban, Bruce Stanton, George A. O'Toole

Dartmouth Scholarship

We previously reported that Pseudomonas aeruginosa PA14 secretes a protein that can reduce the apical membrane expression of the cystic fibrosis transmembrane conductance regulator (CFTR) protein. Here we report that we have used a proteomic approach to identify this secreted protein as PA2934 [corrected], and we have named the gene cif, for CFTR inhibitory factor. We demonstrate that Cif is a secreted protein and is found associated with outer membrane-derived vesicles. Expression of Cif in Escherichia coli and purification of the C-terminal six-His-tagged Cif protein showed that Cif is necessary and sufficient to mediate the reduction in apical membrane expression …

A Discrete Particle Simulation Study Of Solids Mixing In A Pressurized Fluidized Bed, Willem Godlieb, Niels G. Deen, J.A.M. Kuipers

The 12th International Conference on Fluidization - New Horizons in Fluidization Engineering

A fluidized bed containing polymeric particles is investigated using a state-of-the-art soft-sphere discrete particle model (DPM). The pressure dependency of particle mixing, flow patterns and bubble behaviour are analysed. It is found that with increasing pressure a less distinct bubble-emulsion structure and improved solids mixing can be observed.

Activity being performed when accident occurred: According to [Demining group] Field Officer, Mr. [Name removed], both deminers were together on the base stick when the accident occurred because acting SL [Victim no.1] was verbally instructing Deminer [Victim no.2] how to perform excavation. Deminer [Victim no.2] was performing excavation with a bayonet probe (prodder) and hit a PMN mine. It is not clear why was he using his bayonet prodder for excavation, or if he probed the soil before excavating. No MDD or MDU were in support of DT #4 on the day the accident occurred.

The Differential Effects Of Two Critical Osteoclastogenesis Stimulating Factors On Bone Biomechanics, Yuyu Yuan May 2007

The Differential Effects Of Two Critical Osteoclastogenesis Stimulating Factors On Bone Biomechanics, Yuyu Yuan

All Dissertations

Many skeletal diseases, such as osteoporosis and malignant bone metastases, are generally osteolytic and associated with increased bone resorption and decreased bone strength. Within a complex cytokine environment, the proteins RANKL and M-CSF are critical for osteoclast differentiation and activation, and thus fundamental effectors of osteolytic disorders. Previous studies showed that M-CSF stimulates the proliferation and early differentiation of osteoclast progenitors to osteoclast lineage, while RANKL targets the later stages of fusion and activation, and stimulates the formation of functional active osteoclasts. However, impacts of artificially elevated levels of these proteins on the skeleton system have not been fully characterized. …

Chronic Administration Of Methylphenidate Produces Neurophysiological And Behavioral Sensitization, Pamela B Yang, Alan C Swann, Nachum Dafny

Faculty, Staff and Student Publications

The electrophysiological properties of acute and chronic methylphenidate (MPD) on neurons of the prefrontal cortex (PFC) and caudate nucleus (CN) have not been studied in awake, freely behaving animals. The present study was designed to investigate the dose-response effects of MPD on sensory evoked potentials recorded from the PFC and CN in freely behaving rats previously implanted with permanent electrodes, as well as their behavioral (locomotor) activities. On experimental day 1, locomotor behavior of rats was recorded for 2 h post-saline injection, and sensory evoked field potentials were recorded before and after saline and 0.6, 2.5, and 10 mg/kg, i.p., …

Co-Localization Of P-Creb And P-Nr1 In Spinothalamic Neurons In A Chronic Muscle Pain Model, Marie K. Hoeger Bement, Kathleen A. Sluka

Physical Therapy Faculty Research and Publications

Activation of the cAMP pathway is an important mediator of chronic muscle pain. This study examined phosphorylation of the transcription factor cAMP-response-element-binding protein (p-CREB) and the NR1 subunit of the NMDA receptor (p-NR1) in the spinal cord. Bilateral mechanical hyperalgesia of the paw was induced by administering two injections of acidic saline, 5 days apart, into the gastrocnemius muscle of male Sprague–Dawley rats. The proportion of spinothalamic neurons that expressed p-NR1 or p-CREB did not change in the dorsal horn 24 h after the second intramuscular acid injection compared with animals that received pH 7.2 injections. This lack of change …
