Glycosylation Regulates Turnover Of Cyclooxygenase-2., Mary B. Sevigny, Chai-Fei Li, Monika Alas, Millie Hughes-Fulford

Cyclooxygenase-2 (COX-2) catalyzes the rate-limiting step in the prostanoid biosynthesis pathway, converting arachidonic acid into prostaglandin H(2). COX-2 exists as 72 and 74kDa glycoforms, the latter resulting from an additional oligosaccharide chain at residue Asn(580). In this study, Asn(580) was mutated to determine the biological significance of this variable glycosylation. COS-1 cells transfected with the mutant gene were unable to express the 74kDa glycoform and were found to accumulate more COX-2 protein and have five times greater COX-2 activity than cells expressing both glycoforms. Thus, COX-2 turnover appears to depend upon glycosylation of the 72kDa glycoform.

The research project 'Best Practices for Developing the Engineering Estimate' was executed by Clemson University on behalf of the South Carolina Department of Transportation (SCDOT). The primary objective of this research project was to investigate the strengths and weaknesses of two types of estimating methods being utilized today by state department of transportations (DOT) nationwide. These two types of estimating methods are the unit cost line item approach, which the SCDOT utilizes, and the cost-based estimating approach. This research report identifies accuracy obtained, resources expended, and methodologies utilized when implementing either type of estimating approach. This was done so the …

A three-dimensional wave packet generated by a local disturbance in a two-dimensional hypersonic boundary layer flow is studied with the aid of the previously solved initialvalue problem. The solution can be presented as a sum of modes consisting of continuous and discrete spectra of temporal stability theory. Two discrete modes, known as Mode S and Mode F, are of interest in high-speed flows since they may be involved in a laminar-turbulent transition scenario. The continuous and discrete spectra are analyzed numerically for a hypersonic flow. A comprehensive study of the spectrum is performed, including Reynolds number, Mach number and temperature …

The water quality in the Reedy River basin has been shown to be impacted from the diverse and highly urbanized land uses within the watershed. The objective of this study was to examine the observed toxicological effects of EDCs that may be present in the Reedy River watershed by using a suite of biomarkers that were measured in indigenous fish to characterize the exposure and biological effects of these contaminants. Bluegill were analyzed for unnatural estrogenic effects such as: 1) Vitellogenin (VTG) 2) Activity of estrogenic compounds in bile extractions 3) UDP-glucuronosyltransferase (UGT) activity. Simulation And Characterization Of Tire Nonuniformity-Induced Steering Nibble Vibrations Through Integrated Subsystems Modeling, Derek Mangun

Periodic force variations produced by nonuniform tire/wheel assemblies are known root causes of torsional steering wheel vibrations known as 'nibble'. Previous studies have sought to investigate this issue through modeling or experimentation involving the entire vehicle or specific subsystems, however a direct link between objectively measured tire force variations and the vibration levels perceived by vehicle occupants has yet to be established. Analytical models of a nonuniform tire, double-wishbone suspension system, and rack and pinion steering system are sourced from literature and integrated into a single subsystem-level model validated against experimental data obtained as part of a collaborative effort.
