Incorporating improved preparation techniques, modern taxonomy, and quantitative analysis of environmental variables known to influence marsh foraminifers in other coastal regions refined marsh foraminiferal biofacies of the Mississippi delta region. Elevation, pore water salinity, total carbon, and mean grain size were compared with foraminiferal distributions in a transgressive marsh system of the lower Lafourche headland of the south-central delta plain. Cluster analysis aided definition of two biofacies, one from the marsh interior and one from the marsh edge. The marsh edge biofacies was further subdivided into levee crest and bayou margin biofacies. Correlation analysis suggested that seven of the 21 …

Spx is a global regulator discovered in Bacillus subtilis to suppress basal growth and development processes and activate transcription of genes involved in thiol homeostasis when a cell encounters oxidative stress. Its activity relies on reversible thiol-disulfide bond formation and binding RNA polymerase rather than DNA. The discovery that Staphylococcus aureus global virulence regulator, SarA, is more active upon cysteine reduction suggests that redox response could mediate virulence in this important human pathogen. We describe the cloning of spx from S. aureus strain RN6390, overexpression in Escherichia coli, and purification of native protein. Antibodies against Spx were raised for western …

Gossypol and its methylated derivatives are produced in the leaves, seeds, stems, and roots of cotton plants. Although gossypol is toxic to many animals, other uses for gossypol are actively being investigated. To develop an experimental model for gossypol biosynthesis, a project to produce hairy root cultures from cotton was initiated. Hairy root cultures from two cotton species, Gossypium hirsutum (DPL 90) and Gossypium barbadense (Sea Island) were developed in this study. Gossypol was synthesized and retained by hairy root tissue at levels similar to that found in cottonseed. Cultures originating from a single transformation event were more similar in …

Novel synthetic routes to formation of gold-magnetite nanoparticles have been designed by sonochemistry. Treatment of preformed magnetite nanoparticles with ultrasound in aqueous media with dissolved tetrachloroauric acid resulted in the formation of gold-magnetite nanocomposite materials. These materials maintained the morphology of the original magnetite particles. The morphology of the gold particles could be controlled by adjusting experimental parameters, including the addition of small amounts of solvent modifiers such as methanol, diethylene glycol, and oleic acid. Further experiments were conducted with silver and titanium. This research consists of three parts. The first part is an investigation of several popular image restoration techniques. The techniques are used to restore 2-D image data, f(x, y), that has been blurred by a known point spread function (PSF), b(x, y) and corrupted by an unknown amount of noise, n(x, y). Several sample images are restored using all of the techniques. Of the methods investigated the one which produces the best restoration results was determined to be the Wiener deconvolution method. Preliminary Screening Of Soil Actinomycetes For Anti-Tyrosinase Activity, Te-Sheng Chang, Min Tseng

Journal of Marine Science and Technology–Taiwan

A bacterial strain TI-B10 was isolated from forest soil in Taiwan. The strain showed 46 U/ml of anti-tyrosinase activity. On the basis of the typical colony appearance and 16S rRNA gene sequences analysis, the strain TI-B10 belonged to the genus Streptomyces. The antityrosinase activity of the strain could be improved to 73 U/ml under optimal cultural conditions at pH 8.0 and 30°C in the YMG medium.

Role Of The Cytoplasmic Domain In Anabaena Sensory Rhodopsin Photocycling: Vectoriality Of Schiff Base Deprotonation, Oleg A Sineshchekov, Elena N Spudich, Vishwa D Trivedi, John L Spudich

Light-induced electric signals in intact E. coli cells generated by heterologously expressed full-length and C-terminally truncated versions of Anabaena sensory rhodopsin (ASR) demonstrate that the charge movements within the membrane-embedded part of the molecule are stringently controlled by the cytoplasmic domain. In particular, truncation inverts the direction of proton movement during Schiff base deprotonation from outward to cytoplasmic. Truncation also alters faster charge movements that occur before Schiff base deprotonation. Asp(217) as previously shown by FTIR serves as a proton acceptor in the truncated ASR but not in the full-length version, and its mutation to Asn restores the natural outward …

Invasive species are having severe ecological (Mack et al. 2000) and economic (Pimentel et al. 2005) impacts on ecosystems around the world. Invasive species can alter many ecosystem processes (Crooks 2002, Walker & Smith 1997) including: water and nutrient availability, such as form and amount of N if the soil (Evans et al. 2001, Sperry et al. 2006); primary productivity, through shifts in growth rates or efficiency of resource use; disturbance regimes, including the type, frequency, and severity of disturbances such as fire (D’Antonio 2002); and community dynamics, such as species replacements (Alvarez & Cushman 2002). Many factors influence a prospective college student's final college decision. As Augsburg College seeks to grow its theatre department, the question of what matters most to prospective theatre students is especially timely. This project seeks to answer the question, "'What factors make the largest impact in attracting and matriculating theatre majors to Augsburg College?" One-on-one interviews were conducted with current Augsburg theatre students. These in-depth conversations provided insight into the effectiveness of various recruiting methods and identified untapped recruitment opportunities. The purpose of this study was to determine the effectiveness that transitional activities have on student achievement and attendance when students moved from elementary school to middle school. The data were gathered from two school districts that are demographically similar. Data included average daily attendance rates, Terra Nova Achievement Scores from social studies in grades five, six, and seven, as well as math in grades five, six, and seven. There were 187 students in the Treatment Group and 147 in the Control Group for a total of 334 students. Comparison of means for demographic equivalency of the groups (t-test) and …

Expression of selenocysteine (Sec)-containing proteins requires the presence of a cis-acting mRNA structure, called selenocysteine insertion sequence (SECIS) element. In bacteria, this structure is located in the coding region immediately downstream of the Sec-encoding UGA codon, whereas in eukaryotes a completely different SECIS element has evolved in the 3’-untranslated region. Here, we report that SECIS elements in the coding regions of selenoprotein mRNAs support Sec insertion in higher eukaryotes. Comprehensive computational analysis of all available viral genomes revealed a SECIS element within the ORF of a naturally occurring selenoprotein homolog of glutathione peroxidase 4 in fowlpox virus. Advancements in the field of biomaterials are being made to produce an artificial silk fiber. A gene was constructed which utilized components from both the dragline silk of Nephila clavipes and nematode collagen. For this engineered protein, the yeast strain Pichia pastoris was chosen to be the host organism. Previous research has shown P. pastoris to have comparatively low amounts of specific protein productivity. Therefore, this problem must be compensated for by obtaining extremely high cell densities. Flavobacterium columnare and Edwardsiella ictaluri are the leading causes for losses in the aquaculture industry. The losses from the diseases caused by these pathogens total $2.5 million for catfish and $100 million for total aquaculture. F. columnare's infection rates and stressors associated with outbreaks have been well-characterized, but the specific inflammatory responses of catfish neutrophils to the bacteria have not been studied. Structural stormwater management practices help reduce the quantity and improve quality of stormwater runoff. This dissertation focuses on costs and cost effectiveness of these practices. Design, construction and maintenance costs data that were collected from six different sources and adjusted for purchasing power differences over time and location are analyzed using stochastic Leontief cost functions. Effects on these costs of land prices, wages for engineering, construction, and landscaping services, water storage or treatment, and differences in designs of the SMPs and the biophysical regions in which they are located are estimated with the Leontief functions. Composition, antioxidant capacities and cell inhibition properties of curcumin-free turmeric (Curcuma longa L.) oils from different sources were evaluated by chromatographic method, two different in vitro antioxidative activity assays (DPPH* free radical scavenging assay and reducing power assay) and two different cancer cell lines (Caco-2 and MCF-7). Turmeric oil A (TOA) contains zingiberene, turmerone, and ar-turmerone, while turmeric oil B (TOB) contains 1-phellandrene and α-terpinolene as the major compounds. The antioxidant tests showed that both turmeric oils possessed strong free radical scavenging activities and high reducing powers compared with standard antioxidants such as BHT and commercial rosemary oil (RO). …

The purpose of this study was to develop and assess a potential key feature in a minimally-invasive breast reconstruction device and to collect information for an in vitro tissue test system. Tannic acid is a hydrolysable plant tannin, and it has been determined that tannic acid functions as a collagen crosslinking agent through both hydrogen bonding mechanisms and hydrophobic effects. Additionally, current research suggests that tannic acid may have anti-tumor properties. The performance of Nzi traps for tabanids (Tabanus similis Macquart, T. quinquevittatus Wiedemann, Chrysops aberrans Philip, C. univittatus Macquart, C. cincticornis Walker, Hybomitra lasiophthalma (Macquart)), stable flies (Stomoxys calcitrans Linnaeus) (Diptera: Muscidae) and mosquitoes (Aedes) (Diptera: Culicidae) was investigated at various sites in Canada (Ontario, Alberta) and USA (Iowa, Florida, Louisiana). Traps made from selected fabrics, insect nettings and hand-dyed blue cotton were compared to the African design to provide practical recommendations for temperate environments. Amyloid-β (Aβ) toxicity has been postulated to initiate synaptic loss and subsequent neuronal degeneration seen in Alzheimer's disease (AD). We previously demonstrated that the standardized Ginkgo biloba extract EGb 761, commonly used to enhance memory and by AD patients for dementia, inhibits Aβ-induced apoptosis in neuroblastoma cells. In this study, we use EGb 761 and its single constituents to associate Aβ species with Aβ-induced pathological behaviors in a model organism, Caenorhabditis elegans. We report that EGb 761 and one of its components, ginkgolide A, alleviates Aβ-induced pathological behaviors, including paralysis, and reduces chemotaxis behavior and 5-HT hypersensitivity in a transgenic …

The genus Hydrospsyche Pictet sensu stricto in North America is divided into three species groups, the Hydrospyche cuanis Ross Group Ross, the Hydropsyche depravata Hagen Group Ross, and the Hydropsyche scalaris Hagen Group Banks. Thirty-one of thirty-six described species are recognized in the Hydropsyche scalaris Group. Examination of adult males resulted in 5 junior subjective synonyms and one possible new species based on a single exemplar from Sevier County, Utah. Hydropsyche bidens Ross 1938, H. orris Ross 1938, and H. alvata Denning 1949 are junior synonyms of Hydropsyche incommoda Hagen 1861. Hydrospyche rossi Flint, Voshell and Parker, 1979 and H. …

Over the past decade there has been an explosion in the study of cantilevered beams on the micron and submicron dimension. The applications and research that involve these structures include state-of-the-art electronic components, sensors, and more recently, studies aimed at elucidating the mechanical properties of cantilevered carbon nanotubes and semiconducting nanowires. In nanoelectro-mechanical systems (NEMS), it is desirable to develop a capacitive readout method involving only two electrodes that are fully compliant with standard CMOS technology.
